Four previously identified immunodominant B-cell epitopes, located within known virulent pneumococcal proteins CbpD, PhtD, PhtE, and ZmpB, had shown promising immunological characteristics, indicating their potential to be used as vaccine antigens. In this study, we further evaluated the opsonophagocytic activity of antibodies against these epitopes and their capacity to protect mice from pneumococcal sepsis. An opsonophagocytic killing assay (OPKA) revealed that OPKA titers of human anti-peptide antibodies against pneumococcal serotypes 1, 3, and 19A were significantly higher ( < 0.001) than those of the control sera, suggesting their functional potential against virulent clinical isolates. Data obtained from mice actively immunized with any of the selected epitope analogues or with a mixture of these (G_Mix group) showed, compared to controls, enhanced survival against the highly virulent pneumococcal serotype 3 ( < 0.001). Moreover, passive transfer of hyperimmune serum from G_Mix to naive mice also conferred protection to a lethal challenge with serotype 3, which demonstrates that the observed protection was antibody mediated. All immunized murine groups elicited gradually higher antibody titers and avidity, suggesting a maturation of immune response over time. Among the tested peptides, PhD_pep19 and PhtE_pep40 peptides, which reside within the zinc-binding domains of PhtD and PhtE proteins, exhibited superior immunological characteristics. Recently it has been shown that zinc uptake is of high importance for the virulence of ; thus, our findings suggest that these epitopes deserve further evaluation as novel immunoreactive components for the development of a polysaccharide-independent pneumococcal vaccine.
Τα συζευγμένα πνευμονιοκοκκικά εμβόλια (PCVs) στις χώρες με υψηλά ποσοστά εμβολιαστικής κάλυψης μείωσαν σημαντικά τα ποσοστά της Διεισδυτικής Πνευμονιοκοκκικής Νόσου (ΔΠΝ) με αποτέλεσμα την ανάπτυξη συλλογικής ανοσίας. Ωστόσο, οι επακόλουθες μεταλλάξεις στο γεννητικό υλικό του πνευμονιοκόκκου, υπο τη συνεχή ανοσολογική πίεση, έφεραν στο προσκήνιο ταχεία αύξηση της ΔΠΝ από ορότυπους που δεν περιλαμβάνονται στα κυκλοφορούντα PCVs (φαινόμενο της αντικατάστασης) και οδήγησε στην αναζήτηση νέων εμβολίων. Οι πρωτεΐνες επιφανείας του πνευμονιοκόκκου που είναι διατηρημένες στα περισσότερα στελέχη του πνευμονιοκόκκου έχουν χρησιμοποιηθεί ως υποψήφια αντιγόνα για την ανάπτυξη πνευμονιοκοκκικών εμβολίων με ευρεία οροτυπική κάλυψη. Εντούτοις, το μεγάλο κόστος καθώς και η αδυναμία παραγωγής τους στην σταθερή και μη τοξική τους δομή δυσχεραίνουν τη χρήση τους ως υποψήφια αντιγόνα εμβολίων. Στην προσπάθεια να ξεπεραστούν οι δυσκολίες αυτές, οι επιστήμονες στράφηκαν στην αναζήτηση ανοσοκυρίαρχων επίτοπων από πρωτεΐνες επιφανείας του πνευμονιοκόκκου. Προς την κατεύθυνση αυτή, ταυτοποιήθηκαν τέσσερις επίτοποι που εντοπίζονται στις CbpD, PhtD, PhtE και ZmpB παθογόνες πρωτεΐνες του πνευμονιοκόκκου εξετάζοντας ορό παιδιατρικών ασθενών ύστερα από διεισδυτική πνευμονική πνευμονιοκοκκική λοίμωξη. Οι επίτοποι αυτοί εμφάνισαν in vitro ενθαρρυντικά ανοσολογικά χαρακτηριστικά με αποτέλεσμα να αποτελούν υποψήφια αντιγόνα εμβολίων. Ως εκ τούτου, στόχος της παρούσας διατριβής είναι η μελέτη της οψωνοφαγοκυτταρικής ικανότητας των αντισωμάτων έναντι των πεπτιδίων αυτών και η αξιολόγηση της προστατευτικής τους ικανότητα έναντι της πνευμονιοκοκκικής σήψης σε μοντέλο ανοσοποίησης μυών. Η δοκιμασία της Οψωνοφαγοκυττάρωσης επιβεβαίωσε in vito τη λειτουργική ικανότητα των αντι-πεπτιδικών αντισωμάτων έναντι των 1, 3 και 19Α παθογόνων οροτύπων του πνευμονιοκόκκου. Τα αποτελέσματα της ενεργητικής ανοσοποίησης μυών με καθένα από τα επιλεγμένα πεπτίδια, καθώς και με το συνδυασμό αυτών (G_Mix ομάδα) απέδειξαν ότι οι ανοσοποιημένοι μύες είχαν μεγαλύτερη διάρκεια επιβίωσης σε σχέση με τους μη ανοσοποιημένους μύες έναντι του εξαιρετικά παθογόνου οροτύπου 3. Ομοίως, η παθητική ανοσοποίηση μυών με ορό της G_Mix ομάδας παρείχε μεγαλύτερη προστασία σε σχέση με τους μη αποποιημένους μύες υποδηλώνοντας ότι η παρατηρηθείσα αύξηση της επιβίωσης των ανοσοποιημένων μυών ήταν μεσολαβούμενη από τα παραγόμενα αντι-πεπτιδικά αντισώματα. Περαιτέρω αξιολόγηση της ανοσολογικής απάντησης των επιλεγμένων πεπτιδίων απέδειξε ότι οι ομάδες των ανοσοποιημένων μυών παρήγαγαν προοδευτικά υψηλότερους τίτλους αντισωμάτων με μεγαλύτερη συγγένεια αντιγόνου-αντισώματος κατά τις διαδοχικές ανοσοποιήσεις. Τα αποτελέσματα αυτά υποδεικνύουν την εγκατάσταση της ανοσολογικής μνήμης και την ωρίμανση της ανοσιακής απάντησης (Affinity Maturation).Τα PhD_pep19 και PhtE_pep40 πεπτίδια που εντοπίζονται στη θέση σύνδεσης του ψευδαργύρου στις πατρικές τους πρωτεΐνες αντίστοιχα, εμφάνισαν τα πιο ενθαρρυντικά ανοσολογικά χαρακτηριστικά. Μάλιστα, σχετικές μελέτες υποστηρίζουν ότι ο ψευδάργυρος κατέχει σημαντικό ρόλο στην ομοιόσταση και στην παθογένεια του πνευμονιοκόκκου. Επιπλέον, παρατηρήθηκε διασταυρούμενη δραστικότητα ανάμεσα στα πεπτίδια αυτά πιθανόν λόγω της κοινής αμινοξικής τους αλληλουχίας που αποτελείται από τρία κατάλοιπα ιστιδίνης σύμφωνα με το μοτίβο HxxHxH. Τέλος, απεδείχθη ότι το PhtD πεπτίδιο, σε αντίθεση με τα υπόλοιπα, περιλαμβάνει έναν τουλάχιστον Τ- κυτταρικό επίτοπο καθώς επάγει ειδικό Τ- κυτταρικό πολλαπλασιασμό. Μέχρι σήμερα πολύ λίγα πεπτίδια ή τμήματα πρωτεινών επιφανείας του πνευμονιοκόκκου εμφανίζουν παρόμοια in vitro και in vivo προστατευτικά χαρακτηριστικά με τα πεπτίδια που μελετήθηκαν. Συνεπώς, η παρούσα διατριβή παρέχει σημαντικό κίνητρο για την περαιτέρω αξιοποίηση των πεπτιδίων αυτών ως πιθανά αντιγόνα με σκοπό την ανάπτυξη ενός νέου πεπτιδικού πνευμονιοκοκκικού εμβολίου με ευρεία οροτυπική κάλυψη.
