DNA barcoding has emerged as an efficient tool for taxonomy and other biodiversity fields. The vast and speciose group of decapod crustaceans is not an exception in the current scenario and comparing short DNA fragments has enabled researchers to overcome some taxonomic impediments to help broadening knowledge on the diversity of this group of crustaceans. Brazil is considered as an important area in terms of global marine biodiversity and some regions stand out in terms of decapod fauna, such as the São Paulo coastline. Thus, the aim of this study is to obtain sequences of the mitochondrial markers (COI and 16S) for decapod crustaceans distributed at the São Paulo coastline and to test the accuracy of these markers for species identification from this region by comparing our sequences to those already present in the GenBank database. We sampled along almost the 300 km of the São Paulo coastline from estuaries to offshore islands during the development of a multidisciplinary research project that took place for 5 years. All the species were processed to obtain the DNA sequences. The diversity of the decapod fauna on the São Paulo coastline comprises at least 404 species. We were able to collect 256 of those species and sequence of at least one of the target genes from 221. By testing the accuracy of these two DNA markers as a tool for identification, we were able to check our own identifications, including new records in GenBank, spot potential mistakes in GenBank, and detect potential new species.
Marine crabs of the genus Persephona Leach, 1817 are restricted to American waters of the western Atlantic and eastern Pacific Oceans. Subfamilial assignment of this taxon has varied between authors and its species composition remain in question. We conducted a comparative study based on morphology and molecular phylogenetics for all ten recognized species of Persephona, along with Iliacantha hancocki. We tested whether Persephona finneganae, P. lichtensteinii, and P. crinita represent a single species as suggested by some authors; whether specimens identified as P. punctata, P. mediterranea, and P. aquilonaris warrant treatment as separate species; and whether I. hancocki should be regarded as a junior synonym of P. subovata. Diagnostic morphological characters (of the carapace, chelipeds, and third maxillipeds) were used along with gonopod (male first pleopod 1) features and live coloration. The 16S rRNA and the Cytochrome Oxidase I (COI) (DNA barcoding) mitochondrial genes were used as molecular markers. Both morphological and molecular analyses revealed that putative specimens of P. crinita from Brazil and those assigned to P. finneganae were no different from specimens presently assignable to P. lichtensteinii. P. finneganae is regarded as a junior synonym of P. lichtensteinii, and we apply P. crinita only to specimens we examined from the Gulf of Mexico. Specimens from Brazil previously reported as P. crinita are herewith concluded to represent P. lichtensteinii. Additionally, P. townsendi is a junior synonym of P. orbicularis, Iliacantha hancocki is concluded to be a junior synonym of P. subovata, while P. aquilonaris and P. mediterranea are found to represent separate species. On the basis of our revisions, eight species of Persephona are considered valid, and the reported distribution for P. crinita is restricted.
Recent studies based on morphological and molecular data provide a new perspective concerning taxonomic aspects of the brachyuran family Mithracidae. These studies proposed a series of nominal changes and indicated that the family is actually represented by a different number and representatives of genera than previously thought. Here, we provide a comparative description of the ultrastructure of spermatozoa and spermatophores of some species of Mithracidae in a phylogenetic context. The ultrastructure of the spermatozoa and spermatophore was observed by scanning and transmission electron microscopy. The most informative morphological characters analysed were thickness of the operculum, shape of the perforatorial chamber and shape and thickness of the inner acrosomal zone. As a framework, we used a topology based on a phylogenetic analysis using mitochondrial data obtained here and from previous studies. Our results indicate that closely related species share a series of morphological characteristics of the spermatozoa. A thick operculum, for example, is a feature observed in species of the genera Amphithrax, Teleophrys, and Omalacantha in contrast to the slender operculum observed in Mithraculus and Mithrax. Amphithrax and Teleophrys have a rhomboid perforatorial chamber, while Mithraculus, Mithrax, and Omalacantha show a wider, deltoid morphology. Furthermore, our results are in agreement with recently proposed taxonomic changes including the separation of the genera Mithrax (previously Damithrax), Amphithrax (previously Mithrax) and Mithraculus, and the synonymy of Mithrax caribbaeus with Mithrax hispidus. Overall, the spermiotaxonomy of these species of Mithracidae represent a novel set of data that corroborates the most recent taxonomic revision of the family and can be used in future taxonomic and phylogenetic studies within this family.
This checklist is the third part of a series derived from a long-term multidisciplinary project on the biodiversity of decapod crustaceans from marine and coastal environments of São Paulo state (Brazil). We integrated molecular techniques (DNA markers) and morphological analyses of adult specimens for accurate identifications. We compilated 185 species from the literature, but we confirmed the presence of 168 species: 130 of which we sampled, analyzed and obtained sequences (COI and/or 16S totalizing 113 sequences) and 38 that were not directly collected but were confirmed by analyses. In addition, 27 had their distribution removed from São Paulo due to uncertainties, and absence of material as voucher. Five species were reported for the first time on the coast of São Paulo (Calappa ocellata, Neohelice granulata, Teleophrys pococki, Teramnonotus monodi, Tetraxanthus rathbunae) and one on the Brazilian coast (Pseudomedaeus agassizi). Most of the non-sampled species previously reported on the coast of São Paulo might be considered doubtful records stablished in the past by inaccurate analyses, which assumed extended distribution to the area and/or misidentifications. At this time and based on our refined compilation, we can estimate the brachyuran diversity on the coast of São Paulo in 168 species. This detailed inventory contributes to the knowledge on the local decapod fauna by checking available dataset, adding new species records in São Paulo and new sequences to GenBank database. These data may serve as baseline for future identifications and studies on conservation, population genetics, biogeography and phylogenetics, which might flag species that deserve further investigations and concerns.
The hairy crab Pilumnus vinaceus A. Milne-Edwards, 1880, previously considered to be a junior synonym of Pilumnus dasypodus Kingsley, 1879 by Rathbun (1897) is here resurrected. Pilumnus vinaceus can be distinguished from the known western Atlantic species (including P. dasypodus) based on morphological characters and molecular markers. This action increases number of reported species of Pilumnus in the western Atlantic to nineteen.
-This study evaluated the influence of two fruits hosts (orange and papaya) on biological and behavioral parameters of two populations of Ceratitis capitata (Wiedemann) reared under laboratory conditions. One of these populations has been reared under laboratory conditions by 25 years without introduction of wild flies (Lab-pop), while the other has been maintained under the same conditions by 15 years but with occasional introduction of wild specimens (Hybrid-pop). The following parameters were analyzed: emergence percentage, life cycle duration (from eclosion to emergence), adult size, longevity, female fecundity and oviposition preference. The best performance of immatures of both populations was obtained on papaya as a host. Larvae reared on orange had longer life cycle, low emergence percentage and smaller adults. The fruit type did not affect fecundity and longevity of the Lab-pop, but in the Hybrid-pop males lived longer when reared on papaya, while females had higher longevity and fecundity when reared on orange. Females of both populations preferred to lay eggs in papaya (better host for larvae) when compared to orange. However, some eggs were deposited on orange only by females of Lab-pop, suggesting a lower ability for host selection of this population. These data are discussed regarding to the effects of continuous laboratory rearing on the biological parameters of this species.
