RESUMO:Atualmente o uso de métodos alternativos para o controle de doenças e pragas na agricultura, visando minimizar os danos ao meio ambiente e à saúde pública é uma prática reconhecida e necessária. Este trabalho objetivou investigar a ação do óleo essencial de Syzygium aromaticum (L.) Merr. & L.M.Perry sobre o crescimento micelial in vitro dos fungos fitopatogênicos Rhizoctonia solani, Fusarium solani, Fusarium oxysporum e Macrophomina phaseolina. A análise por cromatografia gasosa acoplada com espectrometria de massa possibilitou a identificação de eugenol (83,6%), acetato de eugenila (11,6%) e cariofileno (4,2%). A avaliação microscópica dos micélios dos fungos evidenciou diversas alterações morfológicas, como a presença de vacúolos, desorganização dos conteúdos celulares, diminuição na nitidez da parede celular, intensa fragmentação e menor turgência das hifas. O óleo essencial de cravo apresentou atividade fungicida na concentração de 0,15% sobre o crescimento de R. solani, F. oxysporum e F. solani, entretanto não demonstrou essa atividade sobre M. phaseolina. Esses resultados indicam perspectivas favoráveis para posterior uso do óleo de cravo no controle desses fitopatógenos na agricultura. Analysis by gas chromatography-mass spectrometry allowed the identification of eugenol (83.6%), eugenyl acetate (11.6%) and caryophyllene (4.2%). Microscopic evaluation of mycelia showed several morphological changes such as presence of vacuoles, cell content disorganization, decreased cell wall clearness, intense fragmentation and lower turgescence of hyphae. Clove essential oil showed fungicidal activity at 0.15% on the growth of R. solani, F. oxysporum and F. solani, but not for M. phaseolina. These results indicate favorable perspectives for future use of clove essential oil to control these phytopathogens in agriculture.
RESUMO:Pterodon emarginatus Vogel (Fabaceae) é uma árvore do Cerrado conhecida popularmente como "sucupira branca, faveiro, fava de sucupira e sucupira lisa" e utilizada na medicina popular em preparações anti-reumáticas, antiinflamatórias, analgésicas e antiinfecciosas. The aim of this work was to perform the phytochemical screening of the powder and to evaluate the antimicrobial activity of crude ethanol extract from P. emarginatus barks against Gram-positive bacteria, Gram-negative bacteria and the fungus Candida albicans. The crude ethanol extract was obtained from desiccated and pulverized barks. Its minimum inhibitory concentration (MIC) was determined by using Steers inoculator. The phytochemical tests detected the presence of flavonoids, saponin heterosides, resins and traces of steroids and triterpenoids. Ethanol extract MICs were 0.18 mg mL -1 for the Gram-positive bacteria Rhodococcus equi ATCC 25923, Micrococcus luteus ATCC 9341 and Micrococcus roseus IPTSP/UFG, and for the Gramnegative bacteria Serratia marcescens ATCC 14756 and Pseudomonas aeruginosa ATCC 9027; 0.37 mg mL -1 for Enterobacter cloacae FT 505 LEMC/EPM/UFG; and 0.74 mg mL -1 for the remaining tested bacteria and for the fungus C. albicans. This study opens perspectives for the use of P. emarginatus barks as an antimicrobial drug.
RESUMO:As sementes de guaraná (Paullinia cupana Kunth, Sapindaceae) são utilizadas como estimulante e os principais constituintes químicos são as metilxantinas e os taninos, tais como catequina e epicatequina. Este trabalho teve por objetivo investigar um método por cromatografia líquida de alta eficiência (CLAE) para quantificar, simultaneamente, cafeína, teofilina, teobromina, catequina e epicatequina em pó de guaraná. Os valores obtidos foram comparados com os resultados encontrados no método espectrofotométrico da Farmacopéia Brasileira IV ed. Pelo método cromatográfico, os teores de metilxantinas e taninos totais foram de 4,04% e 1,48%, respectivamente. Os teores de metilxantinas e taninos totais, obtidos pelo método espectrofotométrico, foram de 4,88% e 4,05%, respectivamente. Acredita-se que, nas condições experimentais empregadas, o método espectrofotométrico não foi específico, quantificando outras substâncias fenólicas presentes no guaraná. Portanto, sugere-se o emprego desse método cromatográfico no controle de qualidade do guaraná em pó, por ser preciso, sensível, específico e rápido. The seeds of Paullinia cupana Kunth, Sapindaceae, are used as a stimulant and the main chemical constituents are the methylxanthines and tannins, as catechin and epicatechin. This study aimed to investigate a method by high performance liquid chromatography (HPLC) to quantify both caffeine, theophylline, theobromine, catechin and epicatechin in powdered of P. cupana. The values obtained were compared with the results of the spectrophotometric method of the Brazilian Pharmacopoeia IV ed. For the chromatographic method, the levels of methylxanthines and total tannins were 4.04% and 1.48%, respectively. The levels of methylxanthines and total tannins obtained by the spectrophotometric method, were 4.88% and 4.05%, respectively. It is believed that, under our experimental conditions, the spectrophotometric method was not specific and quantified others phenolic substances present in the P. cupana. Therefore we suggest the use of this chromatographic method in quality control of P. cupana powder, to be accurate, sensitive, specific and fast. RESUMO:A Spiranthera odoratissima A. St.-Hil. (manacá) é um arbusto cujas raízes são utilizadas popularmente contra dores musculares, de estômago e de cabeça, disfunções hepáticas, reumatismo, e como estimulante do apetite. As folhas são empregadas como depurativo do sangue, nas afecções renais e hepáticas. O objetivo deste trabalho foi realizar o estudo morfo-anatômico de S. odoratissima, bem como a triagem fitoquímica, a determinação dos teores de água, cinzas totais e insolúveis em ácido. O material vegetal foi coletado em Leopoldo de Bulhões/ Goiás e os estudos morfo-anatômicos e a triagem fitoquímica foram realizados utilizando as técnicas convencionais. Verificou-se que a folha é hipoestomática, com estômatos anomocíticos e a epiderme apresenta papilas na face abaxial. O mesofilo é dorsiventral e a nervura principal possui drusas de oxalato de cálcio e cavidades secretoras. O pecíolo apresenta sistema vascular em forma circular. Na raiz, em crescimento primário, observou-se cilindro vascular com organização diarca. Na prospecção fitoquímica detectaram-se fenóis, taninos, cumarinas, flavonoides, triterpernos/esteróis, antraquinonas e antocianinas nas folhas e alcaloides, cumarinas, amido e saponinas nas raízes. St.-Hil. (Rutaceae). Spiranthera odoratissima A. St.-Hil. (manacá) is a shrub whose roots are popularly used against stomach pains, headache, muscle pain, liver disorders, rheumatism and as an appetite stimulant. The leaves are used as a blood depurative and for liver and kidney disease. The purpose of this work was to perform the morphological and anatomical study, the phytochemical screening and determine the water content and the levels of total and acid of S. odoratissima. The plant material was collected in Leopoldo de Bulhões/ Goiás and the morpho-anatomical studies and the phytochemical screening were performed using conventional techniques. It was verified that the leaf is hypostomatic, with anomocytic stomata and the epidermis has papillae on the abaxial surface. The mesophyll is dorsiventral and the midrib presents druses of calcium oxalate and secretory cavities. The petiole has a circular vascular system. The root in primary growth presents vascular cylinder with diarchic organization. Phenols, tannins, coumarins, flavonoids, triterpenes/sterols, anthraquinones and anthocyanins were detected in the phytochemical screening of the leaves, and alkaloids, coumarins, saponins and starch were detected in the roots. RESUMO:A Spiranthera odoratissima A. St.-Hil (manacá) é utilizada popularmente como depurativo do sangue, nas afecções renais e hepáticas (chá das folhas) para dores musculares, de estômago, de cabeça, e disfunções hepáticas (chá das raízes). O objetivo desse trabalho foi avaliar a composição química do óleo essencial e a atividade antimicrobiana do óleo essencial, do extrato etanólico bruto e frações obtidos das folhas de S. odoratissima contra bactérias Gram positivas e negativas, e Candida albicans. O extrato bruto das folhas foi obtido por maceração seguido de concentração em rotaevaporador e as frações por partição em coluna filtrante. O pó das folhas foi submetido à hidrodestilação em aparelho de Clevenger e o óleo essencial obtido foi analisado por CG/EM. A atividade antimicrobiana foi avaliada pelo método da diluição em ágar para determinar a concentração inibitória mínima (CIM). Os constituintes majoritários do óleo essencial foram β-cariofileno (20,64%), γ-muuroleno (17,70%), biciclogermacreno (14,73%), e δ-cadineno (13,40%). No estudo da atividade antimicrobiana de S. odoratissima, os principais resultados foram obtidos contra Staphylococus epidermidis (extrato etanólico bruto, CIM de 0,098 mg/mL), C. albicans (fração hexano, CIM de 0,049 mg/mL), Bacillus cereus (diclorometano, CIM de 0,098 mg/mL), Micrococcus roseus (fração acetato de etila, CIM 0,049 mg/mL), e M. roseus, Micrococus luteus, B. cereus e C. albicans (fração metanol, CIM de 0,391 mg/mL).Palavras-chave: plantas medicinais, manacá, Rutaceae, Cerrado, β-cariofileno.ABSTRACT: Chemical composition of the essential oil and evaluation of the antimicrobial activity of essential oil, crude ethanol extract and fractions of Spiranthera odoratissima A. Spiranthera odoratissima ("manacá") has been popularly used as a blood cleanser, for liver and kidney diseases (tea from the leaves), as well as for muscle and stomach pains, headache and liver disorders (tea from the roots). The aim of this study was to evaluate the chemical composition of the essential oil and the antimicrobial activity of essential oil, crude ethanol extract and fractions of S. odoratissima leaves against Gram-positive and negative bacteria and Candida albicans. The crude extract of the leaves was obtained by maceration and was concentrated in a rotavapor, while the fractions were obtained by partition on column filter. The powdered leaves underwent hydrodistillation in a Clevenger apparatus and the obtained essential oil was analyzed by GC/MS. The antimicrobial activity was evaluated by using the agar dilution method for determining the minimum inhibitory concentration (MIC). The major constituents of the essential oil were β-caryophyllene (20.64%), γ-muurolene (17.70%), bicyclogermacrene (14.73%) and δ-cadinene (13.40%).
