Service-Oriented Architecture (SOA) provides a flexible framework for service composition. Using standard-based protocols (such as SOAP and WSDL), composite services can be constructed by integrating atomic services developed independently. Algorithms are needed to select service components with various QoS levels according to some application-dependent performance requirements. We design a broker-based architecture to facilitate the selection of QoS-based services. The objective of service selection is to maximize an application-specific utility function under the end-to-end QoS constraints. The problem is modeled in two ways: the combinatorial model and the graph model. The combinatorial model defines the problem as a multidimension multichoice 0-1 knapsack problem (MMKP). The graph model defines the problem as a multiconstraint optimal path (MCOP) problem. Efficient heuristic algorithms for service processes of different composition structures are presented in this article and their performances are studied by simulations. We also compare the pros and cons between the two models.
Live-cell imaging has opened an exciting window into the role cellular heterogeneity plays in dynamic, living systems. A major critical challenge for this class of experiments is the problem of image segmentation, or determining which parts of a microscope image correspond to which individual cells. Current approaches require many hours of manual curation and depend on approaches that are difficult to share between labs. They are also unable to robustly segment the cytoplasms of mammalian cells. Here, we show that deep convolutional neural networks, a supervised machine learning method, can solve this challenge for multiple cell types across the domains of life. We demonstrate that this approach can robustly segment fluorescent images of cell nuclei as well as phase images of the cytoplasms of individual bacterial and mammalian cells from phase contrast images without the need for a fluorescent cytoplasmic marker. These networks also enable the simultaneous segmentation and identification of different mammalian cell types grown in co-culture. A quantitative comparison with prior methods demonstrates that convolutional neural networks have improved accuracy and lead to a significant reduction in curation time. We relay our experience in designing and optimizing deep convolutional neural networks for this task and outline several design rules that we found led to robust performance. We conclude that deep convolutional neural networks are an accurate method that require less curation time, are generalizable to a multiplicity of cell types, from bacteria to mammalian cells, and expand live-cell imaging capabilities to include multi-cell type systems.
During bacterial growth, a cell approximately doubles in size prior to division, upon which it splits into two daughter cells. This process is subjected to the inherent perturbations of cellular noise 1,2 and thus requires regulation for cell-size homeostasis. The mechanisms underlying cell-size control and their dynamics consequences remain poorly understood due to the difficulty in sizing individual bacteria over long periods of time in a high-throughput manner. Here, we measured and analyzed long-term, single-cell growth and division across different Escherichia coli strains and growth conditions 3 . We found that a subset of cells in a population exhibited transient oscillations in cell size with periods that stretch across multiple (>10) generations. Our analysis revealed that a simple law governing cell size control -a noisy linear map -explains the origins of these cell-size oscillations across all strains. This noisy linear map implements a negative feedback on cell-size control: a cell with a larger initial size tends to divide earlier, whereas one with a smaller initial size tends to divide later. Combining simulations of cell growth and division with experimental data, we demonstrate that this noisy linear map generates transient oscillations, not just in cell size, but also in constitutive gene expression. Two-dimensional cursor control is an important and challenging issue in EEG-based brain-computer interfaces (BCIs). To address this issue, here we propose a new approach by combining two brain signals including Mu/Beta rhythm during motor imagery and P300 potential. In particular, a motor imagery detection mechanism and a P300 potential detection mechanism are devised and integrated such that the user is able to use the two signals to control, respectively, simultaneously, and independently, the horizontal and the vertical movements of the cursor in a specially designed graphic user interface. A real-time BCI system based on this approach is implemented and evaluated through an online experiment involving six subjects performing 2-D control tasks. The results attest to the efficacy of obtaining two independent control signals by the proposed approach. Furthermore, the results show that the system has merit compared with prior systems: it allows cursor movement between arbitrary positions.
One of the promises of the service-oriented architecture (SOA) is that complex services can be composed using individual services. Individual services can be selected and integrated either statically or dynamically based on the service functionalities and performance constraints. For many distributed applications, the runtime performance (e.g. end-to-end delay, cost, reliability and availability) of complex services are very important. In our earlier work, we have studied the service selection problem for complex services with only one QoS constraint. This paper extends the service selection problem to multiple QoS constraints.
