Calbindin-D(9k) (CaBP-9k) and -D(28k) (CaBP-28k) are cytosolic proteins with EF-hand motifs that have a high affinity for calcium ions. Many types of calcium channels and intracellular calcium binding proteins, such as sodium/calcium exchangers (NCXs) and transient receptor potential cation channels (TRPVs), have been detected in the placenta. In this study, the expression of calcium channels involved in maternal-fetal calcium transport were investigated in wild-type mice versus CaBP-9k, CaBP-28k, and CaBP-9k/28k double knockout (KO) mouse models. The expression of calcium transport genes in three dissected sections of the placenta (maternal, central, and fetal) was examined on gestational day 19 (GD 19). The expression of CaBP-9k, TRPV6, TRPV5, and NCX1 mRNA was high in fetal compared to maternal placenta, while CaBP-28k was abundant in the maternal placenta. CaBP-9k was enhanced in all sections of placenta in CaBP-28k KO mice, whereas CaBP-28k was reduced in CaBP-9k KO mice. The expression of TRPV6, TRPV5, and NCX1 were induced in both maternal and fetal placentas in CaBP-9k KO mice, but were upregulated in maternal and central placentas of CaBP-28k KO mice. The levels of these proteins showed similar patterns with those of their mRNA. Placental CaBP-9k, TRPV6, TRPV5, and NCX1 proteins were abundantly expressed in the intraplacental yolk sac located in the fetal placenta. CaBP-28k did not colocalize with other calcium transport genes, although it was enriched in the placental trophoblasts of the decidual zone in the maternal placenta. These results indicate that placental TRPV6, TRPV5, and NCX1 compensate for CaBPs in CaBP-9k and/or CaBP-28k KO mice, and may take over the roles of CaBP-9k and CaBP-28k to transfer calcium ions in the placenta. Taken together, these results indicate that TRPV6, NCX1, and CaBP-9k in the fetal placenta and CaBP-28k in the maternal placenta may play key roles in controlling calcium transport across the placenta during pregnancy.
BACKGROUND:The objective of this study is to evaluate the soil quality to cultivate crops in urban garden. METHODS AND RESULTS: During the period of six month from March to August 2015, measured eight heavy metals, seven Organic items and Fluorine on seventeen urban sites and thirty-one suburban sites in Gwangju city. METHODS AND RESULTS: The average concentration and range of heavy metal in soil are 0.15 mg/kg(ND-0.6) for Cd, 14.9 mg/kg(1.5-33.3) for Cu, 4.4 mg/kg(0.4-71.8) for As, 0.05 mg/kg(ND-1.366) for Hg, 24.7 mg/kg(13.1-62.7) for Pb, 102.5 mg/kg(49.1-276.4) for Zn and 9.2 mg/kg(ND-90.1) for Ni but Cr 6+ is not detected. The average value and range of soil fertility items are 253.5mg/kg(76.6-1766.0) for fluorine, 6.4(4.8-7.7) for pH, 20.3 g/kg(5.0-44.0) for orangic matters, 562.7 mg/kg (28.0-1672.0) for available phosphate, 0.6 cmol + /kg (0.1-2.3) for K, 9.7 cmol + /kg (2.7-22.0) for Ca, 3.0 cmol + /kg (0.9-7.4) for Mg, 1.0 ds/m(0.2-2.9) for conductivity. The concentration of Hg in Suburban area is 0.005 mg/kg lower than 0.134 mg/kg in urban area. Also, the concentration of As, Cd, Ni and Zn is lower than urban area as 32%, 37%, 51%, 71% respectively. CONCLUSION:According to SPI index of soil contamination, 39 sites are first degree and 8 sites are second degree and 1 site(41th) is fourth degree. Pb and Cd are not detected and As is detected tiny amounts in plants grown polluted soil, so heavy metals have not moved to plants.
Essential oils have a sedative effect on stress, and are also known to have antibiotic and anti-carcinogenic effects. These compounds have long been used as natural microbial agents, and have recently been added to a number of pharmaceutical, food and cosmetic products. Controlling the exposure to allergens and pathogens are important factors for the treatment of allergy, and potentially reducing the risk of sensitization and infection. Low humidity, at levels under 35%, may affect human comfort and health during the winter. Patients and other individuals require optimal humidifica-tion to maintain a moisturized respiratory tract necessary for protecting against bacterial infection. We designed an analytical system to examine the effects of aromatherapeutic essential oils on airborne bacteria. The antibacterial activities of essential oils were assayed using agar plate air-sampling methods. A bacterial suspension was sprayed into a bio-clean room through the upper holes using a spray gun. Free-floating airborne bacteria were collected from the bio-clean room (blank) in blood agar plates for 10 sec using an air sampler. Three different concentrations of essential oils (0.0005, 0.005 and 0.05 ppm) were then sprayed into the bio-clean room for 5 min. Free-floating airborne bacteria were collected every 10 min for 10 sec each. Treatment with 0.0005 ppm essential oils inhibited the growth of colonies; this effect appeared to persist after 60 min. Decreased bacterial colony growth was more apparent in the presence of 0.005 ppm and 0.05 ppm essential oils than 0.0005 ppm. These effects were observed after 60 min compared to the control (distilled water). These results indicate that essential oils are able to inhibit the growth of airborne bacteria.
During pregnancy, the placenta represents the establishment of an intimate connection between mother and fetus that is specific to mammals. Calbindins [Calbindin-D9k (CaBP-9k) and -D28k (CaBP-28k)] are proteins possessing EF-hand motifs that have a high affinity for Ca2+ ions and play an important role in the regulation and buffering of Ca2+ in the various tissues. Many types of calcium channels, intracellular calcium binding proteins, Na+/Ca2+ exchangers (NCX) and transient receptor potential cation channels (TRPV) have been found in the placenta. In this study, the calcium channel in maternal-fetal Ca2+ transport was investigated using the phenotypes of wild-type, CaBP-9k, CaBP-28k and CaBP-9k/28k knockout (KO) mouse models. Expressions of calcium transport genes in 3 dissected sections of placenta (MP: maternal, CP: central, FP: fetal) were examined by real-time RT-PCR (RT-qPCR) and Western blot analysis at gestational Day 19 in these mice. The level of TRPV6 mRNA and protein was highest in the MP and CP of CaBP-28k KO mice and the FP of CaBP-9k KO mice compared with other sections of KO mice. The level of CaBP-9k was significantly induced in CaBP-28k KO mice in MP, CP and FP compared with in WT mice, which levels were elevated from maternal to fetal sections. The expression of CaBP-28k mRNA and protein was reduced in CaBP-9k KO mice compared with WT in the 3 sections of placenta. The expression of NCX1 mRNA and protein was higher in all KO mice than in WT in MP and NCX1 was highest in CaBP-28k KO mice in CP, but strong in CaBP-9k KO mice in FP compared with other strains. These results indicate that TRPV6 and NCX1 participate in transferring calcium ions between maternal and fetal compartments and alteration of CaBP-9k/28k is involved in the intracellular Ca2+ buffering system among WT and KO mice. These results taken together indicate that TRPV6 and CaBP-9k genes may play a role as a key element in controlling placental calcium transport during pregnancy.
Abstract. The sap of Acer okamotoanum has been termed 'bone-benefit-water' in Korea owing to its mineral and sugar content. In particular, the calcium (Ca) and potassium (K) concentrations of the sap of Acer okamotoanum are 40-and 20-times higher, respectively, than commercial spring water. In the present study, we examined whether Acer okamotoanum sap improves or prevents hypertension-like symptoms in a rat model. Male Sprague-Dawley rats (8-weeks-old) were provided commercial spring water supplemented with 25, 50 or 100% Acer okamotoanum sap, 3% potassium ions (K + ) or captopril, and treated daily for 2 weeks with NG-nitro-L-arginine methyl ester (L-NAME; 100 mg/kg/day) by subcutaneous injection, in order to induce hypertensive symptoms. Rats were euthanized 6 h following the final injection. To assess the effect of the sap on hypertension-like symptoms, we examined the mean blood pressure (BP), protein levels and localization of endothelial nitric oxide synthase (eNOS) in the descending aorta of the rats. BP levels were significantly lower in hypertensive rats received 25, 50 and 100% sap compared with rats who were administered only commercial spring water. Protein levels of eNOS were repressed in L-NAME-only-treated rats, but were elevated in the descending aorta of rats administered captopril, K + water and Acer okamotoanum sap (25, 50 and 100%) up to the level of the sham group provided commercial spring water, and then injected with dimethyl sulfoxide for the same period of time. Localized eNOS protein was abundantly expressed in the perivascular descending aorta adipose tissue of the rats. Taken together, these results demonstrated that the sap of Acer okamotoanum ameliorated high BP induced by L-NAME treatment in a rat model.
