PHA-stimulated human peripheral lymphocytes were cultivated in plasma-containing and in protein-free media. During the first 3 days of cultivation a skin-reactive factor (SRF) was released which, when inoculated i.d. in normal individuals, induced a tuberculin-like reaction 4 h after administration. SRF release is dependent only on blastic transformation of lymphocytes and not on their proliferation. An immunoelectrophoretic study of protein-free media revealed in all cultures, stimulated or not, the presence of a single precipitation band in the albumin area.
