RESUMO -Durante a realização do presente trabalho, que visou a avaliação da produção de lacases por fungos endofíticos associados a folhas de mandioca, inicialmente foram obtidas e purificadas as linhagens de fungos endofíticos presentes em folhas da espécie. Para isso, inicialmente, foi realizado o crescimento e purificação dos fungos em meio de cultura batata dextrose agar (BDA). Obtidas as linhagens puras, as mesmas foram submetidas ao processo de cultivo em Extrato de Malte, específico para obtenção de lacase. Os resultados revelaram uma maior atividade enzimática para a amostra F3 (0,081 U/mL), proveniente do crescimento do fungo em meio BDA e, nos extratos R1, R2, R3 e R4. Assim, foi realizada uma pré-purificação do mesmo através da técnica de precipitação com etanol e, após a análise dos extratos obtidos, verificou-se uma maior atividade nos extratos R3-100 e R4-100, que mostraram uma atividade de 0,75 e 0,8333 U/mL, respectivamente. Comparando-se a atividade de lacase por grama de proteína, verificou-se um aumento da atividade significativo para o fungo 3, uma vez que foi obtida uma atividade de 0,75 U/mL, para quantidades muito baixas de conteúdo protéico. Já, para o fungo 4, verificou-se um aumento de quase 1000 % na atividade, que foi de 14,29 U/g para 154,31 U/g.
