Growth is a complex trait whose variability within a population cannot be explained solely by genetic variation. Epigenetic regulation is often suggested as an important factor shaping the phenotype, but its association with growth can be highly context-and species-dependent. Nevertheless, the mechanisms involved in epigenetic regulation of growth in fish are poorly understood. We have used reduced representation bisulphite sequencing to determine the genome-wide CpG methylation patterns in male and female Nile tilapia of different sizes but at the same early stage of domestication. The average CpG methylation level in the reduced genome representation was 63% across groups but many sites displayed group-specific methylation patterns. The number of differentially methylated (DM) CpGs was much higher when the interaction between sex and weight was included rather than when these factors were considered separately. There were 1128 DM CpGs between large and small females and 970 DM CpGs between large and small males. We have found many growth-related genes associated with DM CpGs, namely map3k5 and akt3 in females and gadd45g and ppargc1a in males. Only 5% of CpG locations associated with growth were common to both sexes. In particular, the autophagyrelated gene atg14 displayed a high association of methylation with growth exclusively in males. The sexually dimorphic association between atg14 methylation and growth may uncover novel metabolic mechanisms at play during mouth brooding in Nile tilapia females. Taken together, our data suggest that epigenetic regulation of growth in Nile tilapia involves different gene networks in males and females.
Notable parts of the population in Europe suffer from allergies towards apples. To address this health problem, the analysis of the interactions of relevant allergens with other substances such as phenolic compounds is of particular importance. The aim of this study was to evaluate the correlations between the total phenolic content (TPC), polyphenol oxidase (PPO) activity, antioxidant activity (AOA), and the phenolic compound profile and the content of the allergenic protein Mal d 1 in six apple cultivars. It was found that the PPO activity and the content of individual phenolic compounds had an influence on the Mal d 1 content. With regard to the important constituents, flavan-3-ols and phenolic acids, it was found that apples with a higher content of chlorogenic acid and a low content of procyanidin trimers and/or epicatechin had a lower allergenic potential. This is probably based on the reaction of phenolic compounds (when oxidized by the endogenous PPO) with proteins, thus being able to change the conformation of the (allergenic) proteins, which further corresponds to a loss of antibody recognition. When apples were additionally biofortified with selenium, the composition of the apples, with regard to TPC, phenolic profile, AOA, and PPO, was significantly affected. Consequently, this innovative agronomic practice seems to be promising for reducing the allergenic potential of apples.
Seit mehr als 20 Jahren werden in Europa Windenergieanlagen gebaut. Die Windkraft stellt einen wichtigen Baustein zur Steigerung des Anteils der erneuerbaren Energien dar. Bis 2011 wurden Onshore-Windenergieanlagen bevorzugt in windstarken Gebieten errichtet. Die am häufigsten verbreiteten Nabenhöhen waren bis zu diesem Zeitpunkt ca. 60-105 m. Die zugehörigen Türme wurden vorrangig als Stahltürme ausgeführt. Des Weiteren kamen u. a. auch herstellerbezogen Türme in Betonbauweise (sowohl Ortbeton als auch Fertigteilbauweise) sowie als Fachwerkmaste (Stahl oder Holz) zum Einsatz. Mit der Entscheidung der Bundesrepublik Deutschland, bis 2022 aus der Kernenergie auszusteigen, wurde der flächendeckende Ausbau von Windenergieanlagen eingeleitet. Da nicht ausreichend Stromtrassen zur Verfügung stehen, reicht es nicht aus, in küstennahen Gebieten Windenergie zu gewinnen, sondern man ist auf eine dezentrale Errichtung von Windenergieanlagen angewiesen. Um auch in windschwächeren Regionen effiziente Anlagen stellen zu können, müssen in größeren Höhen Zonen mit gleichmäßigeren Windgeschwindigkeiten erschlossen werden. Auch der Ersatz von älteren, leistungsschwächeren Anlagen durch neue Anlagen (Repowering) zieht z. T. den Bau höherer Türme nach sich. Durch die größeren Turmhöhen, verbunden mit größeren Durchmessern im Turmfußbereich, wird die komplette Ausführung mit vorgefertigten Stahlelementen aus Transportgründen nahezu unmöglich. Somit werden vorgespannte Betonfertigteillösungen oder sogenannte Hybridtürme zunehmend interessanter. Bei den Hybridtürmen wird der untere Teil (ca. 60-70 % der Turmhöhe) mit Betonfertigteilelementen, die mit externen Spanngliedern vorgespannt werden, erstellt, während der obere Turmteil in herkömmlicher Stahlbauweise errichtet wird [1]. Mit steigender Bedeutung der Windkraft und Weiterentwicklung der Turbinen werden höhere Türme für Windenergieanlagen erforderlich. Verbunden damit ergeben sich Änderungen in der Bauweise der Türme. Abhängig vom verwendeten Turmtyp werden die Fundamente unterschiedlich ausgebildet. Bei Stahltürmen werden Stahlteile oder vorgespannte Ankerbolzen einbetoniert, während für den Beton-Beton-Anschluss bei Spannbeton-und Hybridtürmen andere vorgespannte Lösungen Anwendung finden. Bis vor wenigen Jahren kamen in Deutschland bei vorgespannten Türmen nur Spannglieder mit nachträglichem Verbund zum Einsatz. Weiterentwicklungen haben dazu geführt, dass jetzt auch weltweit Spannbeton-und Hybridtürme mit externen Spanngliedern vorgespannt werden. Auch wenn der grundsätzliche Aufbau der Spannglieder ähnlich wie im Brückenbau sein kann, muss die Anwendbarkeit für die abweichenden Randbedingungen im Turm einer Windenergieanlage nachgewiesen sein. Bei der Vielzahl der weltweit ausgeführten Anlagen ist es nicht ausgeblieben, dass aufgrund von Ausführungsfehlern oder während der Bemessung nicht berücksichtigter Erkenntnisse die Verstärkung vorhandener Konstruktionen erforderlich werden kann.
