Human papillomavirus (HPV) E6 and E7 gene products play a central role in the induction of benign proliferation and malignant transformation by interacting with several cellular regulatory proteins such as p53, p16 INK4a , and nuclear factor κB (NF-κB). In this study, HPV DNA was detected by in situ hybridization (ISH) and p53, p16 INK4a , and NF-κB by immunochemistry in 22 penile cancer cases in Kenya. HPV DNA was found in 68.2% of the cases. There was no difference in the p53-and p16 INK4a -positivities in HPV DNA-positive and HPV DNA-negative cases. In HPV DNA-positive cases, the NF-κB positivity in the nucleus, cytoplasm, and nucleus and/or cytoplasm amounted to 73.3%, 93.3%, and 100%, respectively, while in HPV DNA-negative cases, a 28.7% NF-κB positivity of in the nucleus and/or cytoplasm was observed. It is concluded that NF-κB in penile cancer is expressed more frequently in the presence of HPV infection than in its absence.
In this study, the speed of sound (SOS) of two types of rat livers (eight normal livers, four cirrhotic livers) was measured with a scanning acoustic microscope using two transducers, one of which had an 80-MHz and the other a 250-MHz center frequency. The 250-MHz transducer had a better spatial resolution adapted to studying fiber or hepatic parenchymal cells. In normal livers, averages of the SOS values were from 1598 to 1677 m/s at 80-MHz and from 1568 to 1668 m/s at 250-MHz. In the fiber tissue of cirrhotic livers, averages of the SOS values were from 1645 to 1658 m/s at 80-MHz and from 1610 to 1695 m/s at 250-MHz, while the SOS values in the other tissue of cirrhotic livers ranged from 1644 to 1709 m/s at 80-MHz and from 1641 to 1715 m/s at 250-MHz. In one liver, SOS in fiber tissue was larger than that of tissues without fiber while in others it was lower. The resulting two-dimensional SOS maps provide a unique quantitative insight of liver acoustic microstructures in a healthy liver and in a cirrhotic ones. This study would be helpful to understand the complex relationship between acoustic properties and liver disease including fiber tissue.
Psychosocial stress is generally associated with adverse health behaviors and has been linked to the development of cardiovascular diseases (CVD). Recently, an individual's sense of coherence (SOC), which is a concept that reflects the ability to cope with psychosocial stress, has been recognized as an essential component of long-term health and stress management. The association between SOC and traditional and alternative atherosclerotic markers in a community sample, however, has not been thoroughly investigated. In the present study, we evaluated stress management capability and psychological conditions using the Japanese version of the Sense of Coherence-13 (SOC-13) Scale, supplemented by the General Health Questionnaire-12 (GHQ-12) that screens for minor psychiatric disorders. The study subjects were 511 adults, median age 64 years (range 48-70), who participated in a regular medical screening program in Nagasaki Prefecture, Japan. We then correlated our findings with atherosclerotic risk factors in the same community sample, such as body mass index (BMI) and proper and regular sleeping habits. We found that close association between good stress management capability and lower BMI and/or regular sleeping habits in elderly Japanese. This provides strong evidence that BMI and sleep management are contributory to SOC. If the ability to cope with psychosocial stress is important to the prevention of CVD, then weight control and proper sleep habits must be emphasized from a psychosocial stress-management perspective as well as a physical one.
Abstract. the causal relationship between chronic inflammation and cancer is widely accepted. Numerous investigations have identified nuclear factor-κB (nF-κB) as an important modulator in driving chronic inflammation to cancer. This study aimed to determine the prevalence of human papillomavirus (HPV) in penile cancer in Japanese patients and whether NF-κB is subsequently overexpressed in penile cancer. Thirty-four specimens of penile tissue (16 malignant and 18 benign cases) were examined to determine the association of HPV infection. An in situ hybridization (ISH) method was used to detect and localize HPV-DNA. A sensitive HPV polymerase chain reaction (PCR) procedure was used for the detection of HPV-DNA, and DNA sequencing was used to identify the HPV genotype. HPV-DNA was detected in 37.5 and 75% of cases of penile cancer, using ISH and PCR, respectively. Our efforts to detect HPV genotypes were unsuccessful as HPV-DNA could not be extracted from these materials. Using ISH, a prevalence of 68.2% of HPV infection was found in penile cancer in Kenyan patients in east Africa. In the present study, all 9 HPV-positive cases, (100%) were nF-κB-positive in the nucleus and/or cytoplasm. In contrast, of the 25 HPV-negative cases, 15 (60%) were NF-κB-positive in the nucleus and/or cytoplasm. Therefore, ISH is a method which is able to prove infection of a large quantity of HPV more effectively when compared with PCR. Thus, a large quantity of HPV infection leads to the activity of NF-κB. The most prevalent genotype was the HPV-22 found in 83.3% of the penile cancer cases. In addition, HPV-11 was found in 81.8% of the non-cancer cases. For cases with a high level of infection, the activity of NF-κB increased compared with those with a low level of HPV infection.
