Менделеева, МоскваЦелью работы было включение дельта-сон индуцирующего пептида (ДСИП) в сшитые полимерные гидрогели на основе поливинилового спирта различного строения с последующим изучением кинетики выхода из них пептида в модели in vitro.В работе были использованы изотропные и макропористые гидрогели на основе акрилового производного поливинилового спирта (Акр-ПВС), а также эпоксидсодержащие макропористые гидрогели, получаемые сополимеризацией данного макромера с глицидиловым эфиром метакриловой кислоты В макропористый гидрогель ПВС пептид включали путем нанесения его водного раствора на предварительно сформированную матрицу, в эпоксидсодержащий гель ПВС-ГМА -путём адсорбции из водного раствора, а в изотропный гель ПВС -путём введения пептида в полимерную смесь в процессе формирования гидрогеля. Кинетику выхода пептида из гидрогелей исследовали при инкубации матриц в фосфатно-солевом буфере (PBS; pH 7,4), в физ. растворе (0,9% раствор NaCl) и в воде, определяя концентрацию ДСИП в супернатантах с помощью обращённо-фазовой ВЭЖХ.Из макропористого гидрогеля ПВС в течение первых 30 мин инкубации выход ДСИП составлял 74%, 70% и 64% в воде, PBS и 0,9% растворе NaCl, соответственно, а завершался через 3 ч. Из гидрогеля, содержавшего эпоксигруппы, даже спустя 48 ч инкубации не наблюдали выделения ни пептида, ни продуктов его распада.Для свежеприготовленного изотропного геля кинетика выхода пептида выглядела следующим образом: 27% в течение первых 30 мин и 78% через 33 ч. Для лиофильно высушенных образцов гидрогелей выход пептида составлял 63% в течение первых 30 мин, в то время как высушивание образцов гидрогеля при комнатной температуре в течение 3 сут. приводило к потере значительной части пептида в результате разрушения его структуры.Ключевые слова: макропористые полимерные гидрогели, модифицированный ПВС, дельта-сон индуцирующий пептид, включение пептидов, кинетика выхода пептида in vitro, тканевая инженерия. 65 * -адресат для переписки
