The role of astrocytes in brain plasticity has not been extensively studied compared with that of neurons. Here we adopted integrative translational and reverse-translational approaches to explore the role of an astrocyte-specific major water channel in the brain, aquaporin-4 (AQP4), in brain plasticity and learning. We initially identified the most prevalent genetic variant of AQP4 (single nucleotide polymorphism of rs162008 with C or T variation, which has a minor allele frequency of 0.21) from a human database (n=60 706) and examined its functionality in modulating the expression level of AQP4 in an in vitro luciferase reporter assay. In the following experiments, AQP4 knock-down in mice not only impaired hippocampal volumetric plasticity after exposure to enriched environment but also caused loss of long-term potentiation after theta-burst stimulation. In humans, there was a cross-sectional association of rs162008 with gray matter (GM) volume variation in cortices, including the vicinity of the Perisylvian heteromodal language area (Sample 1, n=650). GM volume variation in these brain regions was positively associated with the semantic verbal fluency. In a prospective follow-up study (Sample 2, n=45), the effects of an intensive 5-week foreign language (English) learning experience on regional GM volume increase were modulated by this AQP4 variant, which was also associated with verbal learning capacity change. We then delineated in mice mechanisms that included AQP4-dependent transient astrocytic volume changes and astrocytic structural elaboration. We believe our study provides the first integrative evidence for a gliogenetic basis that involves AQP4, underlying language-associated brain plasticity.
Various numerical methods have been used to simulate the rock cutting process. Numerical simulation is a useful tool for estimating the performance of a cutting tool and for understanding the mechanism of rock cutting and interaction between a cutting tool and the rock. These methods supplement the rock cutting test, which is commonly referred to as the linear cutting machine (LCM) test. Mechanical excavators, such as roadheaders, longwall shearers, and trenchers, generally use pick cutters as the cutting tool. In this study, a rock cutting simulation with a pick cutter was developed using the smooth particle hydrodynamics (SPH) technique, which is a mesh-free Lagrangian method. The Drucker–Prager (DP) strength model was used to simulate the brittle behavior of rock. The cumulative damage (CD) model was used to simulate the degraded fragmentation process of rock and the distinctive behavior of rock in the compression and tensile stress regions. In this study, an attempt was made to simulate sequential cutting by multiple pick cutters. The results showed that the numerical simulation matched the experimental results closely in terms of cutter forces, specific energy, and the fragmentation phenomenon. These results confirmed the applicability of the SPH technique in simulating the rock cutting process.
Rock mass contains various discontinuities, such as faults, joints, and bedding planes. Among them, a joint is one of the most frequently encountered discontinuities in rock engineering applications. Generally, a joint exerts great influence on the mechanical and hydraulic behavior of rock mass, since it acts as a weak plane and as a fluid path in the rock mass. Therefore, an accurate understanding on joint characteristics is important in many projects. In-situ tests on joints are sometimes consumptive in terms of time and expenses so that the features are investigated by laboratory tests, providing fundamental properties for rock mass analyses. Although the behavior of a joint is affected by both mechanical and geometric conditions, the latter are often limited, since quantitative control on the conditions is quite complicated. In this study, artificial rock joints with various geometric conditions, i.e., joint roughness, were prepared in a quantitative manner and the hydromechanical characteristics were investigated by several laboratory experiments. Based on the results, a prediction model for hydraulic aperture was proposed in the form of ( e h / e m ) 3 = exp ( − 0.0462 c ) × ( 0.8864 ) J R C , which was a function of the mechanical aperture, joint roughness, and contact area. Relatively good agreement between the experimental results and predicted value indicated that the model is capable of estimating the hydraulic aperture properly.
According to the development of optical technology, the capacity of LIDAR equipments has been greatly improved to get rock mass characteristics precisely and accurately enough and it has been lightened and popularized so that it can be easily used in the field. In this study, we examined the applicability of roughness measurement for joints in a rock slope using LIDAR technique. A triangular irregular network was constructed using LIDAR and a patch, which is a plane structure of discontinuity on rock mass measured from LIDAR scanning, was extracted to estimate the roughness of the rock slope. Four different kinds of roughness parameters were analyzed to find out their correlation with JRC for various point intervals. Among them, parameter was used to measure the roughness of a patch. Regression analysis between four roughness parameters and JRC with respect to point interval was performed. All the roughness parameters decreased with the increase of point interval. In addition, the parameter value showed greater decrease for rougher surfaces. A method of roughness measurement using parameter on rock slope discontinuities was suggested which showed slight overestimation of the real roughness value.
This study describes the results of blasting overbreak analysis using the stereo photogrammetry method in an underground mine. For comparing its quantitative measurements, LIDAR system was applied to the test site and blasting overbreak was analyzed for 4 test blasting operations. The difference in values obtained from the two methods showed only 0.81% in volume and 1.05% in area, respectively, therefore authors verify the field applicability of stereo photogrammetry method on underground mine. The volumes of overbreak measured from 4 test blastings were 29.84 m , respectively, in photogrammetry analysis on excavation surface, and it was shown that the volume of overbreak decreases with blasting sequence. From these measurements, it is concluded that the stereo photogrammetry method can describe the underground excavation surface effectively and the its quantitative data can be used for analysis of volume, area and overbreak of excavation zone. In this study, a LIDAR laser scanner was used to scan a rock slope around Mt. Gwanak and to produce point cloud from which directional information of rock joint surfaces shall be extracted. It was analyzed using two different algorithms, i.e. Ball Pivoting and Wrap algorithm, and four sampling intervals, i.e. raw, 2, 5, and 10 cm. The results of Fuzzy K-mean clustering were analyzed on the stereonet. As a result, the Ball Pivoting and Wrap algorithms were considered suitable for extraction of rock surface orientation. In the case of 5 cm sampling interval, both triangulation algorithms extracted the most number of the patch and patched area.
A rock joint is a planar discontinuity that has significant influence on the mechanical and hydraulic characteristics of rock mass. Laboratory experiments are often conducted on a joint to investigate and provide fundamental information for rock mass analysis. Although joint roughness and mechanical aperture exert great effects on the experimental results, controlling them in quantitative manner is quite complicated and consumptive in terms of specimen preparation. A new and simple method for the quantitative generation of the joint specimen was proposed in this study. Based on random midpoint displacement method, a joint specimen with a void space inside was generated. Parametric studies for the roughness and mechanical aperture were carried out, and as a result, the two joint properties could be controlled by manipulating input parameters of random midpoint displacement method. In order to validate the proposed method, two joint specimens, which had different levels of roughness and aperture, were generated and printed. Surface coordinates of the specimens were obtained by a 3D laser scanner, and calculated to make a comparison between the target values and the estimated values. Results showed that the method was capable of generating joint specimens with satisfactory precision.
