The heritability and the phenotypic and genetic correlations of down weight (DW), down fibre diameter (DFD), and coefficient of variation of the down fibre diameter (CVDFD) of Chinese Alashan Left Banner White Cashmere goat were estimated on 1375 one-year-old animals, born in 2009, 2011 and 2013 and bred at the Station for Livestock Improvement of Alashan (Left Banner, Inner Mongolia, P.R. China). For all traits, significant effects were for sex, cohort and sex-cohort interaction (p < .001). The heritability for DFD and CVDFD was high, 0.41 ± 0.08 and 0.52 ± 0.06, respectively. Heritability for the DW was low (0.12 ± 0.03). Phenotypic correlation calculated by Pearson's coefficient showed that DFD is positively correlated with both CVDFD (0.29 ± 0.07) and DW (0.20 ± 0.05). The phenotypic correlation between CVDFD and DW was negative (À0.11 ± 0.06). The genetic correlations between DW and CVDFD and between DFD and CVDFD were both high and positive (0.63 ± 0.16 and 0.39 ± 0.1, respectively) while the DW showed a negative genetic correlation with DFD (À0.27 ± 0.2). Our results suggest that the selection for reducing DFD and its CVDFD is possible and a genetic progress can be achieved quickly in the Chinese Alashan Left Banner White Cashmere goat. The cashmere hair follicle (HF) perpetually goes through cycles of growth, involution and rest. The photoperiod is the main factor in the control of seasonal coat change in cashmere goats while stem cells play a crucial role in the HF growth. Several factors, including Platelet-Derived Growth Factor A (PDGFA), Bone Morphogenetic Protein 2 (BMP2) and Lim-Homeobox gene 2 (LHX2) are implicated in HF morphogenesis and cycle. In this work, the mentioned molecules were investigated to evaluate their role in follicular cycle activation. The study was performed on skin samples collected at different periods of HF cycle and the molecular expression of PDGFA, BMP2 and LHX2 was evaluated by Real-Time PCR (qPCR) at each time point. Since PDGFA showed the most variation, the goat PDGFA gene was sequenced and the protein localization was investigated by immunohistochemistry together with PDGF receptor α (PDGFRα). PDGFA immunostaining was observed in the basal layer of the HF outer root sheath and the immunoreaction appeared stronger in the regressive HFs compared to those in the anagen phase according to qPCR analysis. PDGFRα was observed in the HF epithelium, proving the effect of PDGFA on the follicular structure. The data obtained suggest that PDGFA and BMP2 are both implicated in HF cycle in goat. In particular, PDGFA secreted by the HF is involved in the anagen activation.

Background Acute or chronic irreversible respiratory failure may occur in patients undergoing pneumonectomy. Aim of this study was to determine transcriptome expression changes after experimental pneumonectomy in swine model. Experimental left pneumonectomy was performed in five pigs under general anaesthesia. Both the resected and the remaining lung, after 60 post-operative completely uneventful days, underwent genome-wide bulk RNA-Sequencing (RNA-Seq). Results Histological analysis showed dilation of air spaces and rupture of interalveolar septa. In addition, mild inflammation, no fibrosis, radial stretch of the bronchus, strong enlargement of airspaces and thinning of the blood supply were observed. Bioinformatic analyses of bulk RNA-Seq data identified 553 Differentially Expressed Genes (DEGs) at adjusted P-value below 0.001, between pre- and post-pneumonectomy. The top 10 up-regulated DEGs were Edn1, Areg, Havcr2, Gadd45g, Depp1, Cldn4, Atf3, Myc, Gadd45b, Socs3; the top 10 down-regulated DEGs were Obscn, Cdkn2b, ENSSSCG00000015738, Prrt2, Amer1, Flrt3, Efnb2, Tox3, Znf793, Znf365. Leveraging digital cytometry tools, no difference in cellular abundance was found between the two experimental groups, while the analysis of cell type-specific gene expression patterns highlighted a striking predominance of macrophage-specific genes among the DEGs. DAVID-based gene ontology analysis showed a significant enrichment of “Extrinsic apoptotic signaling pathway” (FDR q = 7.60 × 10− 3) and “Response to insulin” (FDR q = 7.60 × 10− 3) genes, along with an enrichment of genes involved as “Negative regulators of DDX58/IFIH1 signaling” (FDR q = 7.50 × 10− 4) found by querying the REACTOME pathway database. Gene network analyses indicated a general dysregulation of gene inter-connections. Conclusion This translational genomics study highlighted the existence both of individual genes, mostly dysregulated in certain cellular populations (e.g., macrophages), and gene-networks involved in pulmonary reaction after left pneumonectomy. Their involvement in lung homeostasis is largely supported by previous studies, carried out both in humans and in other animal models (under homeostatic or disease-related conditions), that adopted candidate-gene approaches. Overall, the present findings represent a preliminary assessment for future, more focused, studies on compensatory lung adaptation, pulmonary regeneration and functional reload.

The short-haired Segugio Italiano and rough-haired Segugio Italiano are two breeds of Italian scenthound. The two breeds were subjected to divergent selection based on the type (long versus short) and pigmentation (fawn versus black & tan) of the hair, leading to the development of the two above-mentioned phenotypes. Microsatellites markers from 120 unrelated dogs were used to estimate the genetic variability within the two Segugio Italiano breeds and to assess if divergent selection for the two phenotypes has possibly led to the development of subpopulations distinguished by coat type and pigmentation. Finally, we also estimated the genetic distance between the two Segugio Italiano breeds and two other related breeds, Segugio Maremmano and Segugio dell'Appennino. The mean Fis, Fit and Fst values indicated a moderate level of inbreeding. In both breeds the mean observed heterozygosity was lower than the expected heterozygosity. The AMOVA, PCoA and STRUCTURE analyses showed lack of differentiation between the two breeds and the absence of substructuring in the population. Cavalli-Sforza chord distance, Nei's genetic distance and Reynolds-Weir Cockerham's genetic distance between the two Segugio Italiano breeds were low. Low genetic distances were also found between the Segugio Italiano and the others two related breeds indicating that there is no difference in terms of genetic distance among the three breeds despite the important phenotypic differences among them. This paper provides a basis to change the breeding guidelines of the four breeds in the light of these informations.
