Thymocyte differentiation and lineage commitment is regulated by an extensive network of transcription factors and signaling molecules among which Erk plays a central role. However, Erk effectors as well as the molecular mechanisms underlying this network are not well understood. Erf is a ubiquitously expressed transcriptional repressor regulated by Erk‐dependent phosphorylation. Here, we investigated the role of Erf in T cell maturation and lineage commitment, using a double‐fluorescent Erf‐floxed mouse to produce thymus‐specific Erf knockouts. We observed significant accumulation of thymocytes in the CD4/CD8 DP stage, followed by a significant reduction in CD4SP cells, a trend for lower CD8SP cell frequency, and an elevated percentage of γδ expressing thymocytes in Erf‐deficient mice. Also, an elevated number of CD69+TCRβ+ cells indicates that thymocytes undergoing positive selection accumulate at this stage. The expression of transcription factors Gata3, ThPOK, and Socs1 that promote CD4+ cell commitment was significantly decreased in Erf‐deficient mice. These findings suggest that Erf is involved in T cell maturation, acting as a positive regulator during CD4 and eventually CD8 lineage commitment, while negatively regulates the production of γδ T cells. In addition, Erf‐deficient mice displayed decreased percentages of CD4+ and CD8+ splenocytes and elevated levels of IL‐4 indicating that Erf may have an additional role in the homeostasis, differentiation, and immunologic response of helper and cytotoxic T cells in the periphery. Overall, our results show, for the first time, Erf's involvement in T cell biology suggesting that Erf acts as a potential regulator during thymocyte maturation and thymocyte lineage commitment, in γδ T cell generation, as well as in Th cell differentiation.
Ο μεταγραφικός παράγοντας ERF (Ets2 Repressor Factor, ERF), μέλος της οικογένειας των Ets γονιδίων, αποτελεί ένα μεταγραφικό καταστολέα, η ενεργότητα του οποίου ελέγχεται μέσω φωσφορυλίωσης από την Erk1/2, γεγονός που τροποποιεί την υποκυτταρική διαμερισματοποίησή του. Μολονότι η Erk1/2 έχει σημαντικό ρόλο στην ωρίμανση και τη διαφοροποίηση των Τ κυττάρων και ο ERF εκφράζεται στα Τ λεμφοκύτταρα, ο ρόλος του ERF στη βιολογία των Τ κυττάρων δεν έχει μελετηθεί. Στην παρούσα διατριβή μελετήσαμε το ρόλο του Erf στην ωρίμανση των Τ κυττάρων σε σύστημα διαγονιδιακών ποντικών μετά από ιστοειδική αποσιώπηση του Erf στο θύμο, ενώ σε ένα in vitro σύστημα χρησιμοποιώντας την κυτταρική σειρά HuT78 μελετήσαμε τη φωσφορυλίωση του ERF καθώς και το λειτουργικό του ρόλο μετά από υπερέκφραση. Τα αποτελέσματά μας παρέχουν βάσιμες ενδείξεις για τη συμμετοχή του Erf στη ρύθμιση της διαδικασίας καθώς διαγονιδιακοί ποντικοί με ιστοειδική αποσιώπηση του Erf στο θύμο παρουσίασαν στατιστικώς σημαντικά μειωμένα επίπεδα των κυτταρικών υποπληθυσμών CD4+ και TCRβ και στατιστικώς σημαντικά αυξημένα επίπεδα διπλά θετικών (CD4+CD8+) κυττάρων. Εν ολίγοις, υποδηλώνουν ότι ο Erf ρυθμίζει την ωρίμανση των θυμοκυττάρων κατά το στάδιο CD4+CD8+, πριν τη δέσμευση της κυτταρικής σειράς. Αναλύοντας το μοριακό μηχανισμό της δράσης του Erf, μελετήσαμε την έκφραση καίριων γονιδίων που εμπλέκονται σε αυτή τη διαδικασία και βρήκαμε ότι η έκφραση των γονιδίων Gata3, ThPOK και Socs1, γονιδίων δηλαδή που επάγουν τη CD4 κυτταρική δέσμευση, μειώνεται στατιστικώς σημαντικά στα ζώα με αποσιώπηση του Erf, γεγονός που συνηγορεί υπέρ της θετικής ρύθμισης του Erf στη CD4 δέσμευση των θυμοκυττάρων. Δεδομένης της διαδοχικής και επαγόμενης έκφρασης αυτών των γονιδίων (το Gata3 επάγει την έκφραση του ThPOK ενώ το δεύτερο μπορεί να επάγει την έκφραση του Socs1) υποθέτουμε ότι ο Erf δρα θετικά, άμεσα ή έμμεσα, και ανοδικά επί του Gata3. Για να ελέγξουμε με ποιο τρόπο ο ERF συμμετέχει στη βιολογία των περιφερικών Τ κυττάρων, μελετήσαμε την έκφρασή του σε HuT78 Τ κύτταρα και βρήκαμε ότι η φωσφορυλίωσή του και ο υποκυτταρικός εντοπισμός του είναι γεγονότα εξαρτώμενα της Erk1/2 ενώ έχουμε ενδείξεις ότι δρα ανασταλτικά στον κυτταρικό κύκλο. Τέλος, βρήκαμε ότι η υπερέκφραση του ERF μειώνει στατιστικώς σημαντικά την παραγωγή της IL-2, γεγονός ιδιαίτερα ενδιαφέρον καθώς η IL-2 είναι κυτταροκίνη με ευρύ φάσμα δράσεων στο ανοσοποιητικό σύστημα. Συνολικά, η έρευνά μας αποτελεί την πρώτη ένδειξη ότι ο ERF εμπλέκεται στη βιολογία των Τ λεμφοκυττάρων αποτελώντας το έναυσμα για τη συνέχιση της μελέτης του ρόλου του ERF στη θυμοποίηση και σε άλλες διαδικασίες όπως η διαφοροποίηση των Τ βοηθητικών κυττάρων (ΤΗ).
