The widely used green fluorescent protein (GFP) decarboxylates upon irradiation; this involves removal of the acidic function of the glutamic acid at position 222, thereby resulting in the irreversible photoconversion of GFP. To suppress this phenomenon, the photostable, non-photoconvertible histidine was introduced at position 222 in GFP. The variant E222H shows negligible photodynamic processes and high expression yield. In addition, the stable and bright fluorescence over a wide pH range makes the E222H protein an alternative for GFP in fluorescence imaging and spectroscopy. Other fluorescent proteins are predicted to benefit from replacement of the catalytic glutamic acid by histidine.
En la vida cotidiana, las experiencias se ‘enmarcan’ o se presentan desde perspectivas sociológicas, culturales, políticas, filosóficas, comunicacionales, históricas o psicológicas, y esto se ha vuelto cada vez más frecuente en nuestro mundo digital dominado por las redes sociales. Los marcos dan forma a la manera en que interactuamos y damos sentido a los eventos según cómo los presentemos o experimentemos. El cambio de marco puede resultar en una modificación del punto de vista que tenemos sobre nuestras circunstancias y ayudarnos a interpretar y dar sentido a las situaciones vividas. El término ‘marco’ se utiliza ampliamente en Drama in Education (DiE) y generalmente se remonta a la concepción sociológica de marco de Goffman (1974). En DiE, el marco es un principio importante de planificación y organización y un enfoque eficaz para estimular el pensamiento crítico, la práctica y la evaluación. En esta presentación, la Profesora Carmel O'Sullivan identifica los principios clave del marco y explora cómo el marco dramático ayuda a los/as participantes a identificarse con un rol, asumir un punto de vista asociado con ese rol en un escenario ficticio y lidiar con los obstáculos a los que se ven enfrentados/as (tensión dramática). Ella discute cómo se puede profundizar el marco para prevenir el mono-perspectivismo y cumplir varias funciones, como colocar a los/as participantes en relación con el evento dramático, aumentar la participación afectiva y al mismo tiempo proteger a quienes participan a través de la distancia del marco, crear tensión dramática y proporcionar nuevas perspectivas sobre los problemas (Davis, 2014; Eriksson, 2011; Heathcote, 1984).
