A rhamnolipid released by Pseudomonas aeruginosa 196 Aa into the culture medium reduced the number of local lesions induced by tobacco mosaic virus on leaves of the hypersensitive host iiicotiana glutinosa L. by up t o 90%. The content of potato virus X in the systemically infected host Nicotiana tabacum L. 'Samsun' is decreased in inoculated as well as in secondarily infected leaves by up to 50%. In a smaller degree red clover mottle virus is influenced in the systemic host Pisum sativum convar. speciosum (Dierb.) Alef 'Sadja'. I n den letzten Jahrzehnten ist eine Anzahl an Verbindungen aufgef linden iind entwickelt worden, mit denen eine Hemniung der Vermehrung oder Ausbreitung von Pflanzenviren moglich ist. Einige von ihnen werden heute bereits zur Viruseliminierung in %ell-, Meristem-und SproBspitzenkulturen eingesetzt [ 1-41. Antiphytovirale Verbindungen finden sich sowohl unter Synthetika als auch unter Extrakten oder Ausscheidungsprodukten von hoheren Organismen und Vikroorganismen. Letztere geben Ansatzpunkte zur biotechnologischen Produktion bzw. fur neue Einsatzgebiete entsprechender Priipamte, z. B. fur lipophile Fraktionen aus der Hefe Lodderomyces elongisprus IMET H 128 [5]. Da letztere ebenso wie antiphytoviral aktive Lysolecithine [6, 71 und Alkansulfonsiiuren [8] neben einer oder zwei hydrophoben, zumeist langeren Alkanketten hydrophile Bereiche unterschiedlicher Struktur besitzen, lag es nahe, auch andere Substenzen ahnlicher Struktur bezuglich antiphytoviraler Aktivitaten zu iintersuchen. Nachfolgend wird uber antiphytovirale Aktivitiiten eines Rhamnolipids berichtet, das von Psezcdomonas aervginosa in das Kulturmedium ausgeschieden wird.
Naterial und MethodenKultiviemng und Wachstumskontrolle v m P . aeruginosa P . aeruginosa 196 Aa (Inst. Franpais du Petrole, Frankreich) wurde in Schuttelkultur in einem Minimalmedium nach [9] mit Glycerol (30 g/l) als Kohlenstoff-und Ammoniumsulfat (1 g/l) als Stickstoff-Quelle bei 30°C kultiviert. Six nucleobase and nucleoside analogues (6‐aminouracil [6‐AU], 6‐aminothymin [6‐AT], 6‐fluorothymin [6‐FT], 2,4‐dioxohexahydro‐1,3,5‐triazine [DHT], 9‐(2,3‐dihydroxypropyl) adenine [DHPA] and 1‐β‐D‐ribofuranosyl‐1,2,4‐triazol‐3‐carboxamid [ribavirin]) with potential antiphytoviral activity were tested in an in vitro replication system for their effect on tobacco mosaic virus (TMV)‐and potato virus X(PVX)‐induced RNA dependent RNA polymerase. 6‐AU, 6‐AT, 6‐FT, and DHPA inhibited both polymerases which may be a target of these compounds. A Putative noncompetitive mode of inhibition is discussed. Whereas ribavirin did not affect the in vitr RNA synthesis in the established systems, the uracil‐derivative DHT inhibited only the PVX‐specific enzyme.
