Currently, due to abuse in the use of human antibiotics and the weak regulatory control that the authorities have over sewage discharge and manure management, antibiotic resistance genes (ARGs) have become a new type of environmental pollutant. Three different natural water bodies (Poyang Lake, Haihe River and Qingdao No.1 Bathing Beach seawater) were sampled during the same periods to conduct a longitudinal comparison of distribution. The distribution and expression of 11 ARGs in 20 species were studied, and the correlations between the expression and the distribution of time and space of the ARGs in different water bodies were also analyzed. With the exception of ermA, blaNDM-1 and vanA, which were not detected in seawater, the other ARGs could be detected in all three water bodies. Tetracycline resistance genes (tetC, tetM and tetQ) in the seawater and Haihe River had even reached 100%, and sulfa ARGs (sul1 and sul2) in the seawater and Poyang Lake, as well as sul2 and sul3 in the Haihe River, had also reached 100%. The ARG pollution in Haihe River was much more serious, since 14 and 17 of 20 ARG species were significantly higher compared with seawater and Poyang Lake, respectively. Some ARGs also had a high absolute abundance. The absolute abundance of macrolide resistance genes (ermB) in seawater was as high as 8.61 × 107 copies/L, and the anti-tuberculosis resistant genes (rpoB and katG) in the Haihe River Basin were highly abundant at 1.32 × 106 copies/L and 1.06 × 107 copies/L, respectively. This indicates that ARGs have gradually become more diverse and extensive in natural water bodies. The results of a redundancy analysis (RDA) of the three water bodies showed that although each water body is affected by different factors in space and time, overall, the presence of AGRs is closely related to the production and life of human beings and the migration of animals.

In this study, we used metagenomic sequencing to analyze the composition of ARGs in certain original habitats of northeast China, comprising three different rivers and riverbank soils of the Heilongjiang River, Tumen River, and Yalu River.Results: Twenty types of ARG were detected in every water sample. The major ARGs were multidrug resistance genes, at approximately 0.5 copies/16s rRNA, accounting for 57.5% of the total ARG abundance. The abundance of multidrug, bacitracin, beta-lactam, macrolide-lincosamide-streptogramin, sulfonamide, fosmidomycin, and polymyxin resistance genes covered 96.9% of the total ARG abundance.No signi cant ecological boundary of ARG diversity was observed. The compositions of the resistance genes in the three rivers were very similar to each other, and 92.1% of ARG subtypes were shared by all water samples. Except for vancomycin resistance genes, almost all ARGs in riverbank soils were detected in the river water. About 31.05% ARGs were carried by Pseudomonas. Opportunistic pathogenic bacteria carrying resistance genes were mainly related to diarrhea and respiratory infections. Multidrug and betalactam resistance genes correlated positively with mobile genetic elements (MGEs), indicating a potential risk of diffusion.Conclusions: The composition of ARGs in three different rivers was similar, indicating that climate played an important role in ARG occurrence. ARG subtypes in river water were almost completely the same as those in riverbank soil. ARGs had no signi cant geographical distribution characteristics. Many ARGs were carried by human pathogenic bacteria related to human diarrhea and respiratory infections, such as Pseudomonas aeruginosa and Aeromonas caviae. In general, our results provide a valuable dataset of river water ARG distribution in northeast China. The related ecological geography distribution characteristics should be further explored.

Background Under anoxic conditions, the body will undergo a series of changes to compensate for the low oxygen environment. Changes in gut microbes are an important part of this. Currently, there are relatively few studies on the changes of intestinal microorganisms and metabolites in the low-oxygen environment of plateau. This study employed a non-targeted metabolomics approach coupled with shotgun metagenomic sequencing to analysis the changes of intestinal microorganisms and metabolites in rats under hypoxic environment. Results Significant difference in biodiversity were observed. At the genus level, the relative abundance of Lactobacillus, Alistipes in the hypoxic cohort were significantly increased, while Flavonifractor, Faecalibacterium and Dorea were significantly decreased(P < 0.05). Non-targeted metabolomics analysis showed significant differences in 4 metabolic pathways (pyrimidine metabolism, aminoacyl-tRNA biosynthesis, nicotinate and nicotinamide metabolism, thyroid hormone synthesis) between T group and C group (P < 0.05). Furthermore, correlation analysis of metagenomic and metabolomic data sets showed that there was a certain relative quantitative relationship between some bacterial genera and differential metabolites. Conclusions Our results indicate that a plateau hypoxic environment can significantly alter the composition of intestinal microbes in the host.Compared with the plain control group, the relative increase of some bacteria genera was accompanied by the increase of related metabolites, suggesting that these bacteria genera and metabolites were closely related to the low-oxygen environment in the plateau.It provides a certain foundation for the next research.
