Livestock are the largest source of anthropogenic methane (CH 4 ) emissions, and in intensive dairy systems, manure management can contribute half of livestock CH 4 . Recent policies such as California's short-lived climate pollutant reduction law (SB 1383) and the Global Methane Pledge call for cuts to livestock CH 4 by 2030. However, investments in CH 4 reduction strategies are primarily aimed at liquid dairy manure, whereas stockpiled solids remain a large source of CH 4 . Here, we measure the CH 4 and net greenhouse gas reduction potential of dairy manure biochar-composting, a novel manure management strategy, through a composting experiment and life-cycle analysis. We found that biochar-composting reduces CH 4 by 79%, compared to composting without biochar. In addition to reducing CH 4 during composting, we show that the added climate benefit from biochar production and application contributes to a substantially reduced life-cycle global warming potential for biochar-composting: −535 kg CO 2 e Mg −1 manure compared to −194 kg CO 2 e Mg −1 for composting and 102 kg CO 2 e Mg −1 for stockpiling. If biochar-composting replaces manure stockpiling and complements anaerobic digestion, California could meet SB 1383 with 132 less digesters. When scaled up globally, biochar-composting could mitigate 1.59 Tg CH 4 yr −1 while doubling the climate change mitigation potential from dairy manure management.
MicroRNAs (miRNAs) play important roles in the fibrosis of systemic sclerosis (SSc). However, the underlying miRNA-mRNA regulatory network is not fully understood. A systemic investigation of the role of miRNAs would be very valuable for increasing our knowledge of the pathogenesis of SSc. Here, we combined miRNA and mRNA expression profiles and bioinformatics analyses and then performed validation experiments. we identified 21 miRNAs and 2698 mRNAs that were differentially expressed in SSc. Among these, 17 miRNAs and their 33 target mRNAs (55 miRNA-mRNA pairs) were involved in Toll-like receptor, transforming growth factor β and Wnt signalling pathways. Validation experiments revealed that miR-146b, miR-130b, miR-21, miR-31 and miR-34a levels were higher whereas miR-145 levels were lower in SSc skin tissues and fibroblasts, normal fibroblasts and endothelial cells that were stimulated with SSc serum. ACVR2B, FZD2, FZD5 and SOX2 levels were increased in SSc skin fibroblasts, normal fibroblasts and endothelial cells that were stimulated with SSc serum. We did not identify any negative correlations among these miRNA-mRNA pairs. miR-21 was specifically expressed at higher levels in SSc serum. Six miRNAs and 4 mRNAs appear to play important roles in the pathogenesis of SSc are worth investigating in future functional studies.
In this study, fourteen wheat cultivars with contrasting yield and N use efficiency (NUE) were used to investigate the agronomic and NUE-related traits, and the N assimilation-associated enzyme activities under low and high N conditions. Under deficient-N, the cultivars with high N uptake efficiency (UpE) and high N utilization efficiency (UtE) exhibited higher plant biomass, yields, and N contents than those with medium and low NUEs. The high UpE cultivars accumulated more N than other NUE type cultivars. Under sufficient-N, the tested cultivars showed similar patterns in biomass, yield, and N content to those under deficient-N, but the varietal variations in above traits were smaller. In addition, the high UpE cultivars displayed much more of root biomass and larger of root length, surface area, and volume than other NUE type cultivars, indicating that the root morphological traits under N deprivation are closely associated with the plant biomass through its improvement of the N acquisition. The high UtE cultivars showed higher activities of nitrate reductase (NR), nitrite reductase (NIR), and glutamine synthetase (GS) at stages of seediling, heading and filling than other NUE type cultivars under both low and high N conditions. Moreover, the high UpE and UtE cultivars also displayed higher photosynthetic rate under deficient-N than the medium and low NUE cultivars. Together, our results indicated that the tested wheat cultivars possess dramatically genetic variations in biomass, yield, and NUE. The root morphological traits and the N assimilation enzymatic acitivities play critical roles in regulating N accumulation and internal N translocation under the N-starvation stress, respectively. They can be used as morphological and biochemical references for evaluation of UpE and UtE in wheat.
Cytokine-induced apoptosis inhibitor 1 (CIAPIN1), initially named anamorsin, a newly indentified antiapoptotic molecule is a downstream effector of the receptor tyrosine kinase-Ras signaling pathway. Current study has revealed that CIAPIN1 may have wide and important functions, especially due to its close correlations with malignant tumors. However whether or not it is involved in the multi-drug resistance (MDR) process of breast cancer has not been elucidated. To explore the effect of CIAPIN1 on MDR, we examined the expression of P-gp and CIAPIN1 by immunohistochemistry and found there was positive correlation between them. Then we successfully interfered with RNA translation by the infection of siRNA of CIAPIN1 into MCF7/ADM breast cancer cell lines through a lentivirus, and the expression of the target gene was significantly inhibited. After RNAi the drug resistance was reduced significantly and the expression of MDR1mRNA and P-gp in MCF7/ADM cell lines showed a significant decrease. Also the expression of P53 protein increased in a statistically significant way (p ≤ 0.01) after RNAi exposure. In addition, flow cytometry analysis reveals that cell cycle and anti-apoptotic enhancing capability of cells changed after RNAi treatment. These results suggested CIAPIN1 may participate in breast cancer MDR by regulating MDR1 and P53 expression, changing cell cycle and enhancing the anti-apoptotic capability of cells.
