The separation of DNA by capillary electrophoresis using poly(ethylene oxide) (PEO) containing gold nanoparticles (GNPs) is presented. The impacts of PEO, GNPs, ethidium bromide (EtBr), and pH on the separation of double-stranded DNA have been carefully explored. Using a capillary dynamically coated with 5.0% poly(vinylpyrrolidone) and filled with 0.2% PEO containing 0.3 x GNPs (the viscosity less than 15 cP), we have demonstrated the separation of DNA markers V and VI within 5 min at pH 8.0 and 9.0. In terms of resolution and reproducibility, GNPs have a greater impact on the separation of DNA at pH 9.0. Resolution improvements for large DNA fragments (> 300 base pairs, bp) are greater than those for small ones in the presence of GNPs. It is important to point out that reproducibility is excellent (relative standard deviations for the migration times less than 0.5%) and thus no further dynamic coating is required in at least 20 consecutive runs in the presence of GNPs. Using 0.2% PEO (pH 9.0) containing 0.3 x GNPs, the separation of DNA fragments ranging in size from 21 to 23,130 bp was accomplished in 7 min. The results presented in this study show the advantage of PEO containing GNPs for DNA separation, including rapidity, high resolving power, excellent reproducibility, and ease of filling capillaries.

This paper describes the photosynthesis of gold nanoparticles (GNPs) in the presence of bovine serum albumin (BSA). The concentration of NaAuCl4 and the relative ratio of NaAuCl4 to BSA are important parameters for controlling the size of the GNPs. We prepared GNPs having average diameters ranging from 7 to 50 nm by illumination (Hg-Xe lamp) of phosphate-buffered solutions (pH 3.0-11.0) containing 1 mM NaAuCl4 and 10 microM BSA for 9 h. The size distribution of the GNPs synthesized at pH 7.0 is narrower relative to that of those prepared at other values of pH. Based on the observation that there are no GNPs formed at 25 degrees C in the absence of either BSA or illumination, we conclude that photolytic reduction is the main mechanism for the formation of the GNPs and that BSA acts as a capping agent to stabilize the as-synthesized GNPs. In addition to BSA, several other proteins, such as beta-casein, conalbumin, hemoglobin, beta-lactoglobulin, lysozyme, myoglobin, ovalbumin, pepsin, and trypsinogen play the same role.

This review focuses on rapid, efficient, and sensitive characterization of the proteome by capillary electrophoresis (CE) and microchip capillary electrophoresis (MCE) in conjunction with laser-induced fluorescence (LIF). A number of advanced CE-LIF and MCE-LIF techniques, including on-column concentration techniques, on-line chemical reaction and enzyme-assay techniques, integrated microdevices, and high-efficiency multidimensional separation techniques that have been developed within the past few years, are capable for the analysis of trace proteins in biological samples and/or in single cells. These powerful techniques also show great potential for drug screening and diagnosis. The basic principle of each technique and its advantages and shortcomings for proteomics studies are discussed in detail. We also highlight the importance of techniques that possess improved analytical sensitivity, sample throughput, and quantitation capabilities for identifying the complexities and dynamics of the proteome expressed by cells, tissues, or an organism.
