Fluorogens with aggregation-induced emission (AIEgens) have emerged as a powerful and versatile platform for the development of novel biosensors. In this study, a series of water-soluble fluorescent probes based on tetraphenylethylene (TPE) were designed and synthesized for the detection of monoamine oxidases (MAOs) based on specific interactions between the probes and the proteins. Among the six probes developed, t-TPEM displays a significant fluorescence increase upon introduction of MAOs. Of particular significance is that the fluorescence of t-TPEM in the presence of MAO-A is 21-fold higher than other proteins including MAO-B. Lineweaver-Burk plots reveal that t-TPEM acts as an uncompetitive inhibitor of MAO-A with Ki = 17.1 μM, which confirms its good binding affinity toward MAO-A. Furthermore, a cell imaging experiment reveals that t-TPEM is able to selectively monitor the activity of MAO-A which is localized in mitochondria of MCF-7 cells.

Bile acids (BAs) were originally known as detergents to facilitate the digestion and absorption of lipids. And our current knowledge of BAs has been extended to potential carcinogenic or cancer suppressor factors due to constant research. In fact, BAs were regarded as a tumor promoters as early as the 1940s. Differential bile acid signals emitted by various bile acid profiles can produce distinct pathophysiological traits, thereby participating in the occurrence and development of tumors. Nevertheless, in recent years, more and more studies have noticed the value of BAs as therapeutic targets. And several studies have applied BAs as a therapeutic agent for various diseases including cancer. Based on the above evidence, we acknowledge that the role of BAs in cancer has yet to be exploited, although considerable efforts have been made to probe the functions of BAs. In this review, we describe the characteristics of BAs as a double-edged sword in cancer, hoping to provide references for future cancer treatments.

Pancreatic ductal adenocarcinoma (PDA) is a worldwide health problem. Early diagnosis and assessment may enhance the quality of life and survival of patients. The present study investigated the potential correlations between the gene and protein expression of laminin-332 (LM-332 or laminin-5) and clinicopathological factors as well as evaluating its influence on the survival of patients with PDA. The expression of LM-332 subunit mRNAs in pancreatic carcinoma specimens from 37 patients was investigated by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) analysis. Using immunohistochemical methods, the protein expressions of the three chains of LM-322 (LNα3, LNβ3 and LNγ2) were determined in 96 pancreatic carcinoma specimens, for association analysis with clinicopathological characteristics from patient data. The results of the prognosis analysis of three mRNAs expression datasets were validated in The Cancer Genome Atlas datasets. RT-qPCR results indicated that the overall relative values of LNα3 and LNγ2 mRNAs were increased in pancreatic carcinoma compared with the control. In immunostaining analyses LNα3 and LNγ2 expression was observed in all tumor tissues from the 96 patient samples. The expression levels of LNα3, LNβ3 and LNγ2 were associated with each other. LNα3 and LNγ2 positivity was significantly associated with differentiation, depth of invasion and advanced stage (P<0.05). The samples were classified into three groups: Basement membrane (B) type, cytoplasmic (C) type and mixed (M) type, according to their LNγ2 immunohistochemical expression patterns. The B type correlated significantly with differentiation (P=0.010) and the M type was significantly associated with hepatic metastasis (P=0.031). Patients with B-type LNγ2 demonstrated significantly better outcomes than patients with the C or M type (P=0.012 and P=0.003, respectively). Overexpression of the α3, β3 and γ2 chains of LM-332 may serve an important role in the progression and prognosis of PDA.

A fluorogenic probe based on the novel fluorophore 5-triazole-quinoline was developed for the detection of hydrogen sulfide, an endogenous signaling molecule associated with the development of various diseases. The lysosome-targeted probe Lyso-HS was synthesized via C-H direct azidation from 8-aminoquinoline; it was able to detect HS in 1 min and exhibited excellent turn-on ability with 95-fold fluorescence enhancement based on a new fluorochrome. The high quenching efficiency was further verified using time-dependent density functional theory (TDDFT). The probe also exhibited high selectivity and a low detection limit (as low as 214.5 nM), which has practical applications for disease detection and monitoring.

Herein, we report as elf-quenching nanoparticle composed of dualt wo-photon dyes of naphthalimides, displaying almost no fluorescencea tb oth channels, owing to aggregation-caused quenching (ACQ). A dual two-photon-channel fluorescent probe for GSH was further developed by inserting ad isulfide bond between two naphthalimides. Upon treatment with GSH, the disulfide bond was cleaved, and two napthalimide fluorophores were separated, leadingt ot urn-on fluorescencea tb othc hannels. This dual-two-photon-channeld esign strategy can be easily extended to other analytes, simply by changing the linker molecules, which createsn ew opportunities to accurately monitorvariousbiological processes in living cells.
