Calon induk merupakan komponen penting dalam usaha budidaya ikan nila (Oreochromis niloticus). Kualitas dan kuantitas calon induk harus terjamin sehingga dapat menghasilkan benih berkualitas sebagai pangkal siklus budidaya ikan nila. Metode pengangkutan calon induk yang baik perlu dikaji lebih dalam untuk mengurangi resiko kematian dan penurunan kualitas selama periode transportasi. Anestesi adalah salah satu langkah yang dapat dilakukan untuk menanggulangi hal tersebut. Eugenol merupakan bahan yang telah banyak diteliti sebagai bahan anestetik pada bidang kedokteran gigi ataupun bidang perikanan.. Penelitian ini menggunakan eugenol 99% yang dapat menurunkan kesadaran ikan dan menurunkan laju metabolisme sehingga mampu menekan tingkat stres yang dialami calon induk pada periode transportasi.Penelitian ini telah dilaksanakan pada tanggal 28-30 April 2021 di Balai Benih Ikan (BBI) Potrobangsan, Kota Magelang, Jawa Tengah. Metode yang diterapkan adalah eksperimen dengan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan 5 kali pengulangan. Perlakuan yang diberikan adalah penambahan eugenol 99% dengan dosis A 0,00 ml/L, B 0,004 ml/L, C 0,008 ml/L, dan D 0,012 ml/L. Jumlah sampel yang diuji yaitu 100 ekor calon induk ikan nila yang dibagi pada 20 unit percobaan dengan padat tebar 1 ekor/ 1 liter air. Hasil penelitian menunjukkan bahwa penambahan eugenol 99% pada pengangkutan calon induk ikan nila berpengaruh nyata (P<0,05) pada persentase kelulushidupan dan kadar glukosa darah. Hasil terbaik diperoleh pada perlakuan B 0,004 ml/L dengan hasil persentase kelulushidupan 96% dan rata – rata glukosa darah di akhir simulasi pengangkutan 105,2±30,32 (mg/dL).
