Cryopreservation Technique for Conservation of Plant Germplasm: Its Possible Use in Indonesia. Semuel Leunufna. Increasing rate of plant germplasm lost in Indonesia has promoted the implementation of various methods for their conservation. Cryopreservation is a technique applicable for a long-term preservation (base collection) of plants possessing non-orthodox (recalcitrant and semi-recalcitrant) seeds and those propagated vegetatively. The technique can be used as an alternative method for orthodox seed plants preservation in the ex situ conservation system. Although field and in vitro collection methods can be applied for the non-orthodox seed plants, a number of disadvantages possesed by these methods, especially in the tropics or the developing countries, deny their use for the establishment of a long-term germplasm collection. Successful implementation of the cryopreservation technique is supported by the development of protocols, which are able to provide a high recovery rate for species understudy, using vitrification based methods which are simple, economical, applicable to complex organs, and able to implement a high number of explants per experiment. The availability of infrastructures including in vitro culture laboratories, continue supply of liquid nitrogen is highly supporting the use of cryopreservation technique in Indonesia.

PENDAHULUANIndonesia merupakan wilayah yang padat keragaman hayatinya termasuk keragaman flora. Dengan luas area 1,3% luas daratan dunia, Indonesia menyimpan 11% spesies tumbuhan dunia (FWI-GFW 2001). Kekayaan ini sebagian besar tersimpan dalam hutanhutan sebagai spesies liar dan lainnya berada pada lahan-lahan pertanian serta perkebunan yang tersebar pada lebih kurang 17 ribu pulau.Sejalan dengan meningkatnya jumlah penduduk, eksploitasi hutan yang tak terkendali, serta meningkatnya jumlah varietas unggul hasil pemuliaan tanaman, kekayaan keragaman hayati yang tersedia dewasa ini mengalami penyusutan dengan sangat pesat. Berbagai pendekatan telah diupayakan guna mempertahankan keragaman yang ada agar dapat digunakan secara berHak Cipta 2007, BB-Biogen kelanjutan dari generasi ke generasi. Selain pendekatan konservasi in situ yang berupaya melestarikan organisme beserta lingkungannya dalam bentuk hutan lindung, taman nasional, dan sebagainya, pendekatan ex situ yang berupaya melestarikan organisme diluar habitat aslinya telah pula diupayakan dalam beberapa bentuk termasuk kebun raya, koleksi lapang maupun bank gen.Dari 6 juta koleksi ex situ yang dikelola di berbagai bank gen di dunia, 39% di antaranya berasal dari tanaman biji-bijian dan 15%-nya tanaman kacangkacangan. Kelompok tanaman lain seperti golongan sayuran, umbian, rerumputan, dan lain-lain, berjumlah 8% atau kurang (Scarascia-Mugnozza dan Perrino 2002).
