The purpose of the present study was to evaluate the efficacy of propolis extract in maintaining the viability of human periodontal ligament (PDL) cells, and to radiographically analyze tooth replantation and the adjacent periodontium in dogs after storage in this extract. Human PDL cells were incubated with the experimental media propolis, milk, saliva, Hank's balanced salt solution (HBSS), and Dulbecco's modified Eagles medium (DMEM, positive controls), and distilled water (negative control). Cell viability was determined 0, 1, 3, 6, 12, and 24 h later by colorimetric MTT assay. Thirty incisors from dogs were divided into two storage time blocks (1 and 3 h) and were maintained in the experimental media. HBSS served as a positive control, and dry teeth (on gauze) as a negative control. The replanted teeth were radiographed once per month for 6 months. The radiographic images were standardized by the shortening/lengthening factor, and were both qualitatively and quantitatively analyzed. The in vitro results showed that the efficacy of propolis in maintaining functional viability of PDL cells was similar to that of milk. Propolis and milk were significantly better than controls from the 6-h time period. The in vivo results showed that teeth maintained in propolis medium exhibited replacement resorption with significant reduction in tooth length, similar to teeth maintained in saliva and dried teeth. This resorption was less intense with the 3-h storage time than the 1-h storage time. Conditions close to normal were found in teeth maintained in milk, similar to the HBSS control. Therefore, although propolis was effective in maintaining the viability of human PDL cells, resorption of the tooth replantation in dogs occurred under these experimental conditions.

Extratos etanólicos de própolis obtidos a partir de diferentes concentrações de própolis e com álcool hidratado com diferentes teores de álcool/água foram analisados. A ação antioxidante destes extratos foi determinada através da captura do radical livre 1,1-difenil-2-picril-hidrazila (DPPH•) e do poder de redução férrico (FRAP) utilizando o complexo férrico-tripiridiltriazina (Fe 3+ -TPTZ). O teor de compostos fenólicos totais e a quantificação de flavonóides também foram verificados e obtidos por espectrometria de massas com ionização por electrospray (ESI(-)-MS) dos extratos. Observou-se que a composição química dos extratos de própolis e sua atividade antioxidante variam com a concentração de própolis e, principalmente, com o teor de álcool/água do etanol hidratado utilizado na extração. Foi possível perceber também que a atividade antioxidante dos extratos é significativamente dependente destes parâmetros de extração e que as composições de compostos fenólicos e flavonóides também variam muito. Os valores de FRAP não apresentaram, porém, boa correlação com o teor de flavonóides.Ethanolic extracts of propolis obtained with different concentrations of propolis and made using hydrous ethanol with various water/ethanol contents were analyzed. The antioxidant action of the extracts was determined by free radical 1,1-diphenyl-2-picryl-hydrazyl (DPPH•) scavenging and by ferric reducing ability power (FRAP), using the ferric-tripyridyl triazine (Fe 3+ -TPTZ) complex. The total phenolic and flavonoid contents were also determined, and the electrospray ionization mass spectrometry spectra (ESI(-)-MS) of the extracts were acquired. The chemical composition of propolis extracts and their antioxidant activity were observed to vary with the concentration of propolis, and especially with the water/ethanol contents of the hydrous ethanol used for extraction. The antioxidant activity of the extracts was also found to be significantly dependent on these extraction parameters, as well as on the composition of the phenolic compounds and flavonoids. FRAP values showed, however, poor correlation with the flavonoid content. No presente trabalho relata-se caso de paciente, funcionário de hospital veterinário, infectado através de arranhadura de gato doméstico portador de esporotricose. Inquérito domiciliar junto aos proprietários do animal fonte de infecção, revelou dois outros casos presuntivos de esporotricose humana transmitida por gatos, e confirmou o diagnóstico, por cultivo do Sporotrix schenckii, em 3 gatos domésticos adicionais. A esporotricose felina caracteriza-se por lesões cutâneas ulceradas e tendência à disseminação sistêmica e evolução fatal. A transmissão intra e inter-espécie é facilitada pela exuberância de fungos nas lesões cutâneas de felinos infectados.

Carcinogenesis is frequently linked to genetic background, however, exposure to environmental risk factors has gained attention as the etiologic agent for several types of cancer, including prostate. The intrauterine microenvironment has been described as a preponderant factor for offspring health; and maternal exposure to insult has been linked to chronic disease in older offspring. Using a model of maternal exposure to low protein diet (LPD; 6% protein), we demonstrated that impairment of offspring rat prostatic growth on postnatal day (PND) 21 was associated with prostate carcinogenesis in older offspring (PND 540). One explanation is that maternal LPD consumption exposed offspring to an estrogenic intrauterine microenvironment, which potentially sensitized prostate cells early during glandular morphogenesis, increasing cellular response to estrogen in older rats. The onset of accelerated prostatic growth, observed on PND 21, associated with an unbalanced estrogen/testosterone ratio and increased circulating IGF-1 in older offspring appears to contribute to the development of prostate carcinoma in gestational groups on low protein (GLP) and gestational and lactational low protein (GLLP) diets (33% and 50%, respectively). Our study strongly indicated maternal exposure to LPD as a potential risk factor for induction of slow-growing prostate carcinogenesis in rat offspring later in life.
