RESUMOOs parasitas gastrintestinais causam enorme prejuízo econômico na bovinocultura, tanto nacional como mundial, ocasionado principalmente por Bunostumom sp., Cooperia sp. e Trichostrongylus sp. O objetivo deste trabalho foi determinar a eficácia in vitro do extrato hidroalcoólico de Artemisia annua (H.7) frente a esses endoparasitas. O H.7 foi produzido com sete dias de percolação a 4ºC e posteriormente liofilizado. Com esse fitoterápico, realizaram-se testes de eclodibilidade de ovos (TEO) e de migração larvar em ágar (TMLA), com seis repetições, com concentrações crescentes (0,78 a 50mg/mL). Para analisar a composição química do fitoterápico, procedeu-se à marcha fitoquímica completa. No TEO, a eficácia variou de 94,08±2,58% na maior concentração a 15,67±0,97% na menor concentração. Já no TMLA os valores encontrados variaram de 90,05±0,55% a 4,12±0,46%. Nas análises fitoquímicas, foram encontrados diversos compostos com propriedades de combater os nematódeos, tanto direta como indiretamente. Os resultados obtidos nos testes in vitro evidenciam que o extrato produzido possui potencial de combater nematódeos gastrintestinais de bovinos. The marine diatom Coscinodiscus wailesii has attracted ecological interest because their blooms affect fishing areas. The aim of this work was the isolation, extraction and partial chemical characterization of soluble exopolysaccharide and bound exopolysaccharide from C. wailesii. Cultures were grown in Guillards f/2 medium under controlled conditions of temperature, aeration, photoperiod and light intensity. Percentage of carbohydrate, uronic acids, sulfates groups and cellular lipids was determined. Ion exchange chromatography of exopolysaccharides produced three fractions whose partial chemical structures were disclosed using 13 C NMR and methylation techniques.Keywords: Coscinodiscus wailesii; diatom; exopolysaccharides. INTRODUÇÃOAs florações de algumas espécies de microalgas podem trazer consequências negativas ao ecossistema prejudicando áreas de pesca e cultivo comerciais, através da produção de toxinas, mucilagens e várias alterações nas cadeias tróficas aquáticas. 1 A microalga marinha Coscinodiscus wailesii Gran et Angst, uma diatomácea cêntrica, quando em floração pode diminuir o índice de nutrientes e de oxigênio da água, provocando a morte ou migração de peixes. Efeitos desta espécie no ecossistema resultaram em sé-rios danos em culturas de "Nori" (Porphyra), macroalga marinha muito utilizada comercialmente como alimento, devido à absorção acelerada de nutrientes por esta diatomácea durante sua floração na costa japonesa. 2 A distribuição da espécie C. wailesii, antes restrita ao Pacífico Tropical e ao Atlântico Ocidental, ampliou-se até a Europa, Estados Unidos, Japão e Austrália, tornando-se, em algumas áreas, importante componente da comunidade fitoplanctônica. 3 No Brasil, na Baía de Paranaguá -PR, a espécie C. wailesii tem sido encontrada desde 1991 em baixas concentrações em todos os meses do ano, produzindo florações esporadicamente, durante as quais se observou uma marcada diminuição da população do fitoplâncton e zooplâncton. Embora a relação entre esses eventos ainda não tenha sido explicada, a possibilidade dessa microalga causar efeitos nocivos motiva a realização de estudos sobre essa espécie ecologicamente importante. 4 Vários autores divergem quanto à composição do polissacarídeo da cápsula mucilaginosa de microalgas ser diferente ou não da composição do exopolissacarídeo solúvel no meio de cultura. Experimentos distintos com diatomáceas do gênero Chaetoceros, com as espécies Cylindrotheca closterium, Nitzschia sp. e Navicula salinarum, mostraram que os exopolissacarídeos produzidos são diferentes daqueles constituintes da cápsula mucilaginosa, descartando a possibilidade dos polissacarídeos encontrados no meio serem provenientes da morte ou degradação celular durante a fase estacionária do cultivo. 5,6 Os autores afirmam que essas diferenças na composição, localização ...
Resumo O objetivo deste estudo foi avaliar a eficácia do extrato hidroalcoólico de Artemisia annua frente a oocistos de Eimeria sp. em camas contaminadas. O extrato foi produzido com 7 dias de percolação a 4°C, sendo posteriormente realizada a marcha fitoquímica; dosagem de fenóis totais, quantificação de artemisinina, ensaio antioxidante e teste de toxicidade. Para testar a atividade anticoccidiana, camas de aves compostas de cepilho de árvores foram contaminadas com 5000 oocistos. Foram formados quatro tratamentos, em triplicata, nos quais foram usadas diferentes concentrações, sendo G1: 12mg/mL, G2: 8mg/mL, G3: 4mg/mL e C-: água. Após a contaminação, foram aspergidos, 800 mL dos extratos nas diferentes concentrações sobre as camas e coletadas, em triplicatas, 10 cm2 de cada local, aleatoriamente, nos tempos: 0, 3, 6, 24, 48, e 72 horas após a aplicação. Nas análises fitoquímicas, foram evidenciados diversos compostos com propriedades antiparasitárias, como flavonoides e taninos. O fitoterápico continha 59,409±1,47μg/dL de artemisinina. O produto na concentração de 12mg.mL-1 apresentou eficácia entre 45,5 e 42,1%. Os resultados dos testes bioquímicos, juntamente com os encontrados no teste anticoccidiano, evidenciaram que o extrato produzido possui alto potencial para combater Eimeria sp.
Haemonchus contortus é responsável por causar enorme prejuízo econômico na produção de pequenos ruminantes, retardando o crescimento e causando anemia em animais infectados. O objetivo deste trabalho foi determinar a eficácia in vitro do extrato hidroalcoólico de Artemisia annua frente a estágios de vida livre deste parasito. O fitoterápico foi produzido com 7 dias de percolação a 4ºC, sendo posteriormente liofilizado. Foram realizados o teste de eclodibilidade de ovos (TEO) e o teste de desenvolvimento larval (TDL), com concentrações crescentes (0,78 a 50 mg/mL) em seis repetições. Para analisar a composição química do fitoterápico, procedeu-se a marcha fitoquímica completa. O produto obtido a partir da A. annua apresentou eficácia de 93,22% (±1,87) no TEO, já no TDL a eficácia foi de 90,33% (±0,27), ambos na concentração de 50 mg/ml. Nas análises fitoquímicas, foram encontrados diversos compostos que podem ter contribuído com o efeito anti-helmíntico, tanto direta como indiretamente. Os dados da marcha fitoquímica, aliados aos resultados dose-dependentes obtidos nos testes in vitro evidenciam que o extrato produzido possui potencial para combater H. contortus. Novos estudos devem ser realizados buscando maximizar a eficácia deste extrato.
