Vitrification is becoming a preferred method for pre-implantation embryo cryopreservation. The objective of this study was to determine the differentially expressed genes of in vivo- and in vitro-produced bovine embryos after vitrification. In vitro- (IVF) and in vivo-derived (IVV) bovine blastocysts were identified as follows: in vitro-produced fresh (IVF-F), in vitro-produced vitrified (IVF-V), in vivo-derived fresh (IVV-F), in vivo-derived vitrified (IVV-V). The microarray results showed that 53 genes were differentially regulated between IVF and IVV, and 121 genes were differentially regulated between fresh and vitrified blastocysts (P < 0.05). There were 6, 268, 962, and 17 differentially regulated genes between IVF-F × IVV-F, IVF-V × IVV-V, IVF-F × IVF-V, and IVV-F × IVV-V, respectively (P < 0.05). While gene expression was significantly different between fresh and vitrified IVF blastocysts (P < 0.05), it was similar between fresh and vitrified IVV blastocysts. Significantly up-regulated KEGG pathways included ribosome, oxidative phosphorylation, spliceosome, and oocyte meiosis in the fresh IVF blastocyst samples, while sphingolipid and purine metabolisms were up-regulated in the vitrified IVF blastocyst. The results showed that in vitro bovine blastocyst production protocols used in this study caused no major gene expression differences compared to those of in vivo-produced blastocysts. After vitrification, however, in vitro-produced blastocysts showed major gene expression differences compared to in vivo blastocysts. This study suggests that in vitro-produced embryos are of comparable quality to their in vivo counterparts. Vitrification of in vitro blastocysts, on the other hand, causes significant up-regulation of genes that are involved in stress responses.

The aim of the study was to show whether there were some differences among 9 Holstein bulls and within their replications on their ability of in vitro fertilization for in vitro embryo production as cleavage and coming into the blastocyst stage. Semen collected and frozen from nine Holstein bulls with satisfactory in vivo fertilization capabilities for artificial insemination was used for in vitro fertilization. Direct washing method by Brackett and Oliphant medium and 5 or 6 h incubation period were used for in vitro fertilization. Charles Rosenkrans medium was used for in vitro embryo culture. An atmosphere with a higher than 95% relative humidity, 39°C, 5% CO 2 and was used for all in vitro embryo production processes. Totally 2519 A and B quality oocytes were treated for in vitro embryo production. As a result statistically significant (P<0.05) variation was found for cleavage and blastocyst development among bulls. However there was no significant difference (P>0.05) between the replications for each bull as cleavage and coming into blastocyst stage. The results showed varied capabilities of bulls for in vitro fertilization and embryo production and male factor can affect success of in vitro embryo production. Suni tohumlama boğası olarak kullanılan ve fertilite sorunu olmayan ve tatminkar düzeyde fertilite oranlarına sahip dokuz Holştayn boğadan alınan ve dondurulan spermalar in vitro fertilizasyon amacıyla kullanılmıştır. İn vitro fertilizasyon için Brackett ve Oliphant mediumu ile Direkt yıkama metodu ve 5-6 saat inkubasyon periyodu; in vitro embriyo kültürü için de Charles Rosenkrans mediumu kullanılmıştır. Kültür periyotlarında %95'in üzerinde bağıl nem, 39°C, %5 CO 2 içeren bir kültür ortamı sağlanmıştır. Toplam 2519 A ve B kalite oosit in vitro embriyo elde etme sürecine alınmıştır. Sonuç olarak yarıklanma ve blastosiste ulaşma oranları bakımından boğalar arasında önemli düzeyde istatistiki farklılık bulunmuştur (P<0.05). Fakat her bir boğanın kendi içindeki tekrarları arasında yarıklanma ve blastosiste ulaşma oranları bakımından önemli düzeyde fark görülmemiştir (P>0.05). Bu sonuçlar boğaların değişik düzeyde in vitro fertilizasyon ve embriyo üretim yeteneklerine sahip olduklarını ve in vitro embriyo üretim başarısı için erkek faktörünün belirleyici olduğunu göstermektedir.

The issue of pregnancy planning in these patients makes treatment tricky.Glatiramer acetate (GA), one of the first-line therapies currently approved for relapsing-remitting multiple sclerosis (RRMS) (34), was originally designed as a synthetic analogue of myelin basic protein (1). Data about exposure to GA during early pregnancy period in humans is limited but animal studies have revealed no fetal risk to date so GA is classified by █ INTRODUCTIONNeural tube (NT) defects are uncommon malformations occurring 6 in 10000 pregnancies (60). Congenital malformation incidence is 3-5% in newborns and NT defects cause 7% of newborn deaths related to congenital malformations (13).Multiple Sclerosis (MS), a chronic demyelinating and degenerative disease of the central nervous system (CNS), is the most common chronic neurologic disability in young adults in AIM: To demonstrate the effect of glatiramer acetate (GA) in chick embryos on neural tube (NT) development, and to explore its effects of FOXP1, apoptosis, and N-cadherin. MATERIAL and METHODS:One hundred fertile, specific pathogen free eggs were divided into 5 groups for this study. The eggshell was windowed specifically at 24 hours of incubation. The embryos in Group 1 (n=20) were treated with 10 μl physiological saline; in Group 2 the embryos (n=20) were given 10 μl GA (equal to daily human therapeutic dose); 20 μl GA (equal to twice daily human therapeutic dose) was injected to embryos in Group 3 (n=20); in Group 4 and 5, 30 μl and 40 μl GA were administered to the embryos (n=20) (equal to x3 and x4 daily human therapeutic dose, respectively). Each egg was re-incubated for 24 hours more. Then, histological and immunohistochemical evaluation of the subjects were done. RESULTS:The embryos with NT defect showed FOXP1 expression without N-cadherin or staining with N-cadherin in another location in our study. We interpreted this result as GA leading to an NT closure defect by increasing FOXP expression. Moreover, we also showed the reverse relation between FOXP1 and N-cadherin at the immunohistochemical level for the first time. CONCLUSION:GA affects the spinal cord development through FOXP in the chick embryo model at high doses.

Bu çalışmanın amacı, etilen glikol direkt transfer metoduna göre dondurulan in vivo sığır embriyolarının taşıyıcı hayvanlara nakledilerek gebelik elde edilmesidir. Projenin ilk aşamasında; 56 adet iyi kalite in vitro üretim sığır embriyosu etilen glikol direkt transfer metoduna göre dondurulmuştur. Çözündürme sonrası TCM-199 + 0.1mM β-merkaptoetanol + %10 fötal buzağı serumu (FCS) içeren solüsyon içerisinde, 24-48 saat inkubasyonda, 36 adet embriyonun re-ekspansiyonu ve zonadan çıktığı görülmüş, canlılık oranı %64.28 olarak tespit edilmiştir. İkinci aşamada; süperovulasyon protokolü uygulanan kültür ırkı ineklerden 8 adet iyi kalite embriyo toplanmıştır. Embriyolar 1.8M etilen glikol + 0.1M sakkaroz içeren %20 yeni doğmuş buzağı serumlu (CS) Dulbecco'nun fosfat tampon solüsyonu (D-PBS) içerisine alınarak 0.25 ml'lik steril payetlere çekilmiştir. Yerleştirme her bir payete bir embriyo olacak şekilde yapılmış ve payetler oda sıcaklığında 15 dk. ekilibrasyona bırakılmıştır. Programlanabilir dondurma cihazının, metil alkol bulunan haznesi içerisine payetler yerleştirilerek,-7 o C'den itibaren soğutma işlemine başlanmıştır.-7 o C'de 2 dk. sonra payetlere seeding yapılmış ve 8 dk aynı ısıda bekletilmiştir. Soğutma işlemine-30 o C'ye kadar 0.3 o C/dak. düşürülecek şekilde devam edilmiş ve direkt-196 o C'deki sıvı azot içerisine alınarak embriyolar muhafaza altına alınmıştır. Çözündürmede, sıvı azot içerisinden alınan payet önce 5-6 sn. havada tutulmuş, 30 o C'deki su banyosunda 10 sn. tutularak çözdürülmüştür. Önceden prostaglandin F2α ile senkronize edilen 8 baş taşıyıcı inekte kızgınlığının yedinci gününde rektal palpasyon ile korpus luteum (CL) tespiti yapılmıştır. CL tespit edilen taşıyıcılara, kriyoprotektan uzaklaştırılmadan embriyolar, cerrahi olmayan yöntemle, ipsilateral olarak nakledilmiştir. Nakil sonrası 60. günde taşıyıcı hayvanların rektal palpasyon ile yapılan gebelik kontrollerinde, 5 taşıyıcı hayvanın gebe olduğu tespit edilmiş ve gebelik oranı %62.5 olarak bulunmuştur. Gebeliklerin hepsi doğumla sonuçlanmıştır. Bu çalışmada, kriyoprotektan olarak etilen glikol'ün sığır embriyolarının dondurulması ve direkt transferinde kullanılabileceği sonucuna varılmıştır.

Nafion, a sulfonated tetrafluoroethylene copolymer, consists of a hydrophobic fluorocarbon backbone (-CF2-CF2-) to which hydrophilic sulfonate groups ( SO −3 ) are attached. The presence of negatively charged sulfonate groups in a nanomembrane brings about a high level of proton conductivity. In this study, Saccharomyces cerevisiae was used for production of bioelectricity in a 2-chambered microbial fuel cell (MFC). We selected 9-cm 2 Nafion 117 and Nafion 112 as nanomembranes to transport the produced proton from the anode chamber to the cathode compartment at ambient temperature and pressure. Initial glucose concentration was 30 g/L. The maximum obtained voltage, current, and power density for Nafion 117 were 668 mV, 60.28 mA/m 2 , and 9.95 mW/m 2 , respectively. For Nafion 112, those results were 670 mV, 150.6 mA/m 2 , and 31.32 mW/m 2 , respectively.
