Desaturation of coenzyme-A esters of saturated fatty acids is a common feature of sex pheromone biosynthetic pathways in the Lepidoptera. The enzymes that catalyze this step share several biochemical properties with the ubiquitous acyl-CoA Δ

-desaturases of animals and fungi, suggesting a common ancestral origin. Unlike metabolic acyl-CoA Δ
9
-desaturases, pheromone desaturases have evolved unusual regio- and stereoselective activities that contribute to the remarkable diversity of chemical structures used as pheromones in this large taxonomic group. In this report, we describe the isolation of a cDNA encoding a pheromone gland desaturase from the cabbage looper moth,
Trichoplusia ni
, a species in which all unsaturated pheromone products are produced via a Δ
11
Z-desaturation mechanism. The largest ORF of the ≈1,250-bp cDNA encodes a 349-aa apoprotein (PDesat-Tn Δ
11
Z) with a predicted molecular mass of 40,240 Da. Its hydrophobicity profile is similar overall to those of rat and yeast Δ
9
-desaturases, suggesting conserved transmembrane topology. A 182-aa core domain delimited by conserved histidine-rich motifs implicated in iron-binding and catalysis has 72 and 58% similarity (including conservative substitutions) to acyl-CoA Δ
9
Z-desaturases of rat and yeast, respectively. Northern blot analysis revealed an ≈1,250-nt PDesat-Tn Δ
11
Z mRNA that is consistent with the spatial and temporal distribution of Δ
11
-desaturase enzyme activity. Genetic transformation of a desaturase-deficient strain of the yeast
Saccharomyces cerevisiae
with an expression plasmid encoding PDesat-Tn Δ
11
Z resulted in complementation of the strain’s fatty acid auxotrophy and the production of Δ
11
Z-unsaturated fatty acids.
[1] High-resolution seismic profiles and sedimentological data from grab samples and long cores provide an unprecedented picture of the structure, sedimentology, and late Quaternary development of two Gulf of Papua (GoP) clinothems, one probably Stage 3 and 4 in age and one Holocene in age. The older was partially eroded during Stage 2 and partially covered by the younger clinothem during Stage 1. The younger clinothem consists of three stratigraphic units separated by two surfaces of erosion, bypass, or correlative surfaces of lap. The surfaces were formed by changes in accommodation and sediment supply. The underlying physiography of the older clinothem also appears to play an important role in governing the shape of the younger clinothem. In the northern gulf, oblique clinoforms of the younger clinothem suggest that the rate of sediment supply from the northern rivers outstripped the formation of new accommodation, whereas in the south, sigmoidal clinoforms indicate that accommodation increased faster than sediment supply. The origin of the new accommodation remains uncertain because of limited age constraints. On the basis of sediment thickness, stratal geometry, and acoustic character, off-shelf transport appears to be the dominant sediment transport direction, with preferential accumulation on the promontories and bypass in the valleys. Presently, observed and computed modern flows and complex gyres in shallow water coupled with wave-and current-supported gravity flows or river floods can explain the form, internal clinoform shapes, and mineralogy of the younger Gulf of Papua clinothem.
Establishing landscape response to uplift is critical for interpreting sediment fluxes, hazard potential, and topographic evolution. We assess how landslides shape terrain in response to a wave of uplift traversing the northern California Coast Ranges (United States) in the wake of the Mendocino Triple Junction. We extracted knickpoints, landslide erosion rates, and topographic metrics across the region modified by Mendocino Triple Junction migration. Landslide erosion rates mapped from aerial imagery are consistent with modeled uplift and exhumation, while hillslope gradient is invariant across the region, suggesting that landslides accommodate uplift, as predicted by the threshold slope model.
