A series of novel substituted (E)‐N′‐benzylidene‐4‐methyl‐2‐(2‐propylpyridin‐4‐yl)thiazole‐5‐carbohydrazide derivatives (6a‐l) have been synthesized by following the multistep synthetic route starting from prothionamide. The resulting compounds were characterized via 1H, 13C NMR, and HRMS spectral data. The synthesized carbohydrazides were evaluated for their in vitro antimicrobial and antioxidant activities. Tested molecules have displayed moderate to good growth inhibition activity. Among the screened compounds, 6b, 6e, 6j, and 6k are found to be the more promising antimicrobial agents. A 2,2‐diphenyl‐1‐picrylhydrazyl assay was used to test the antioxidant activity of the carbohydrazides. The carbohydrazide derivatives 6b and 6i have shown better free‐radical scavenging ability than the other investigated compounds.

Emergence of multidrug resistant and extensively drug resistant tuberculosis has prompted to develop new molecular entities to treat the disease. A series of new 4‐thiazolidinones with pyrazinyl and thiazolyl scaffolds has been synthesized, and their antitubercular activity is reported. The title 4‐thiazolidinones, N‐(pyrazinyl substituted thiazoloylamino)‐2‐aryl‐4‐thiazolidinones (6a, 6b, 6c, 6d, 6e, 6f, 6g, 6h, 6i, 6j) have been first time prepared using pyrazinamide as a starting material via five successive steps. The purity and the structures of the intermediates (carboethoxythiazole, acid hydrazide, and azomethines) and title thiazolidinones (6a, 6b, 6c, 6d, 6e, 6f, 6g, 6h, 6i, 6j) have been confirmed by TLC and spectral analyses, respectively. An antitubercular screening of the new 4‐thiazolidinones has been performed on bacterial strains, Mycobacterium tuberculosis H37Ra and Mycobacterium BCG using the solutions of different concentrations of the compounds (6a, 6b, 6c, 6d, 6e, 6f, 6g, 6h, 6i, 6j) and the screening results are presented. Compound 6a has displayed notable antitubercular activity.

In search of new anticancer and antimicrobial agents, herein we report the synthesis of new substituted (E)-N'-benzylidene-2-(2-ethylpyridin-4-yl)-4-methyl thiazole-5-carbohydrazide derivatives (6 a-n), starting from ethionamide. The newly synthesized compounds were characterized by 1 H NMR, 13 C NMR and HRMS analyses. All the synthesized compounds have been evaluated for their anticancer activity. Among fourteen synthe-sized compounds, five carbohydrazide derivatives have displayed more promising anticancer activity against the lung carcinoma A-549 cells. Also, these compounds were screened for their in vitro antibacterial and antifungal activities. The three carbohydrazide derivatives have exhibited good antibacterial and antifungal activities.
