This study was conducted to evaluate the influence of occupational noise (sound stress) exposure on fasting serum lipid and lipoprotein levels [ serum total cholesterol (TC) ,Triglycerides(TGs) and lipoproteins profile : Very Low Density Lipoprotein(VLDL),Low Density Lipoprotein(LDL-C) and High Density Lipoprotein (HDL-C)] in Erbil textile factory workers .These workers are exposed to high ambient noise level which is generated by big machines used in the factory for more than six hours daily at a strength of 90 deci-Bel [dB] without using ear protectors. The date obtained indicate that under these conditions of chronic and high noise exposure levels there are a significant increase (P< 0.05) in the level of fasting serum (TC) , a non significant increase in the levels of fasting serum ( TGs ,VLDL and LDL-C) and a significant decrease (p< 0.05) in the level of fasting serum (HDL-C).
Introduction: Ischemic stroke is characterized as a sudden neurological deficit attributed to an acute focal injury of the central nervous system by a vascular cause. This study was performed to determine the frequency of G20210A mutation in the prothrombin gene and its effectiveness on the incidence of ischemic stroke in the Erbil city of Kurdistan region, Iraq. Methods: A total of 50 patients with ischemic stroke was analyzed for the detection of prothrombin gene mutation (G20210A), using polymerase chain reaction (PCR), Restriction fragment length polymorphism (RFLP) with Hind III restriction enzyme. Results: We observed no evidence of an association between ischemic stroke and G20210A mutation in the prothrombin gene in this region. Conclusion: Our finding demonstrates that prothrombotic gene variant seems not to be linked to the incidence of ischemic stroke in Erbil region.
Το αιμοστατικό σύστημα είναι ένα ζωτικός μηχανισμός για τον έλεγχο υπερβολικής αιμορραγίας μετά από τραυματισμό στο αγγειακό σύστημα, καθώς και για την πρόληψη μιας κατάστασης υπερπηκτικότητας κατά τη διάρκεια της απόκρισης πήξης. Ένα από τα πιο σημαντικά συστήματα στο ανθρώπινο σώμα είναι το αγγειακό, επειδή όλα τα όργανα και οι ιστοί εξαρτώνται από αυτό για επιβίωση. Το αίμα είναι ένα ζωτικό μέρος της ανθρώπινης φυσιολογίας, ένα σύστημα μεταφοράς που φέρει θρεπτικά και οξυγόνο για να συντηρήσει τα ζωντανά κύτταρα και ταυτόχρονα να διευκολύνει την απομάκρυνση διοξειδίου του άνθρακα και μεταβολικών άχρηστων προϊόντων από τους ιστούς του σώματος. Για τον λόγο αυτό η συντήρηση του αγγειακού συστήματος, που καλείται αιμόσταση, είναι ζωτική για την επιβίωση και είναι η ελεγχόμενη διακοπή της αιμορραγίας, ενώ διατηρεί την ακεραιότητα του αγγείου με ελάχιστη απώλεια αίματος. Για να διασφαλιστεί η συνέχεια των λειτουργιών αυτών, είναι μέγιστης σημασίας η διατήρηση της ροής του αίματος μέσα στο αγγειακό σύστημα με κάθε κόστος. Η σύνθετη διεργασία της πήξης του αίματος οδηγείται μέσω ενός πολύπλοκου ενζυμικού μηχανισμού που αφορά μερικά μονοπάτια μετατροπής ζυμογόνου σε ένζυμο και επιτρέπει μεγάλη εσωτερική ενίσχυση της αντίδρασης. Το τελικό βήμα σε αυτές τις διαδοχικές αντιδράσεις είναι η μετατροπή του διαλυτού ινωδογόνου σε ένα αδιάλυτο ινικό δίκτυο. Η θρομβίνη παίζει ένα κεντρικό ρόλο και είναι η τελευταία ένωση που ενεργοποιείται. Κατά τη διάρκεια της διαδικασίας της πήξης, η θρομβίνη είναι το μόνο ένζυμο που είναι ελεύθερα διαθέσιμο στην κυκλοφορία του αίματος, όπου συναντά διαφορετικά υποστρώματα. Επιπλέον, ο vWF προστατεύει τον παράγοντα πήξης VIII (FVIII) από τη γρήγορη πρωτεολυτική αδρανοποίηση. Οι καρδιαγγειακές ασθένειες είναι από τις σημαντικότερες αιτίες νοσηρότητας και θνησιμότητας στα ανεπτυγμένα κράτη. Παρά τις συνεχείς προσπάθειες από πολυάριθμα εργαστήρια ανά τον κόσμο, δεν έχουν προσδιοριστεί αποτελεσματικά θεραπευτικά εργαλεία για ασθένειες όπως το έμφραγμα του μυοκαρδίου και τα εγκεφαλικά επεισόδια. Έτσι σήμερα αυτό το ερευνητικό πεδίο βρίσκεται ακόμα στα πρώτα στάδια της εξέλιξής του. Υπάρχουν ακόμα πολυάριθμα ερωτήματα σε αυτό το επιστημονικό πεδίο τα οποία παραμένουν αναπάντητα. Εφόσον η θρομβίνη και ο FXa έχουν έναν θεμελιώδη ρόλο στον μηχανισμό της πήξης, είναι λογικό ότι ο υπολογισμός των επιπέδων τους, εκτός από πλήθος άλλων σημαντικών πληροφοριών, αποτελεί τον πιο ενδεδειγμένο τρόπο για την μελέτη της αιμοποιητικής λειτουργίας των ασθενών, επομένως βοηθά στην παρακολούθηση της κατάστασης των ασθενών. Παραδείγματος χάριν , αυτή η προσέγγιση προσφάτως έχει χρησιμοποιηθεί τόσο για τον προσδιορισμό πιθανών επιπρόσθετων αιμοστατικών επιδράσεων όσο και για την ταυτοποίηση ασθενών που βρίσκονται σε κίνδυνο επαναλαμβανόμενων φλεβικών θρομβικών εμβολών. Η πληροφορία αυτή καθιστά εξαιρετικά ενδιαφέρουσα την ανάπτυξη μεθόδων για την παρακολούθηση της παραγωγής της θρομβίνης και του FXa in vitro. Σχετικά με αυτόν τον ευρύ τομέα της επιστήμης, στην παρούσα εργασία αναπτύξαμε εξαιρετικά απλουστευμένες μεθόδους για τον προσδιορισμό της παραγωγής θρομβίνης και του FXa, οι οποίες μπορούν να αυτοματοποιηθούν. Εφαρμόστηκε το πιο ευρέως χρησιμοποιούμενο χρωμογόνο υπόστρωμα, εξετάστηκαν πολλαπλές παράμετροι που σχετίζονται με την αντίδραση και βελτιστοποιήθηκαν ώστε να καθιερωθεί μια μη χρονοβόρα ,κατάλληλη και ταχεία μέθοδος ρουτίνας για την αξιολόγηση μεγάλου αριθμού κλινικών δειγμάτων. Οι ρυθμοί παραγωγής θρομβίνης και του FXa συγκρίθηκαν σε δείγματα πλάσματος πριν και μετά την αφαίρεση θρόμβου. Στη συνέχεια, υπολογίστηκε ο συσχετισμός των τιμών της παραγωγής της θρομβίνης και του FXa. Επιπροσθέτως μελετήθηκαν οι επιδράσεις της φυσικής άσκησης, της συγκέντρωσης των αιμοπεταλίων, του FVIIa και του ιστικού παράγοντα (tissue factor TF) στην παραγωγή της θρομβίνης και του FXa. Επιπλέον προσδιορισμός ρουτίνας αυτού του παράγοντα (θρομβίνη) διεξήχθη σε νοσηλευόμενους διαβητικούς ασθενείς υπό διαφορετικές αντιπηκτικές θεραπείες. Καθώς αυτός ο μηχανισμός πήξης προχωρά σαν μια σειρά συνδεόμενων πρωτεολυτικών αντιδράσεων, τα ένζυμα που δημιουργούνται από αυτές τις διεργασίες δεν είναι διαθέσιμα άμεσα για ποσοτική μέτρηση επειδή αδρανοποιούνται ταχέως από ενδογενώς παραγόμενους αναστολείς πρωτεασών. Για την αντιμετώπιση αυτού του προβλήματος εστιάσαμε στην ανάπτυξη τεχνικών ανοσοαποτύπωσης για να μελετηθεί ο παράγοντας της πήξης FX και η ενεργή του μορφή. Επιπρόσθετα στα πλαίσια αυτής της εργασίας μελετήσαμε την πιθανή λειτουργία της απομονωμένης επικράτειας Α1 του vWF στην ενεργοποίηση των αιμοπεταλίων και στη φυσιολογική διαδικασία της αιμόστασης, ώστε να κατανοηθεί πλήρως ο ιδιαίτερος ρόλος της στην διατήρηση της ακεραιότητας της διαδικασίας της πήξης του αίματος. Γι’ αυτόν τον σκοπό η συγκεκριμένη αμινοξική περιοχή (Α1 επικράτεια) του vWF, κλωνοποιήθηκε σε ευκαρυωτικά και βακτηριακά κύτταρα, απομονώθηκε και χρησιμοποιήθηκε ως βακτηριακή αλλά και ευκαρυωτική πρωτεΐνη.
IntroductionIschemic stroke is sudden neurologic deficits that result from focal cerebral ischemia associated with permanent brain infarction. It is a major cause of disability and death worldwide which occurs as a result of atherothrombotic occlusion of large arteries; cerebral embolism; nonthrombotic occlusion of small, deep cerebral arteries; and proximal arterial stenosis with hypotension that decreases cerebral blood flow in arterial watershed zones.1 A multifunctional multimeric plasma glycoprotein von Willebrand factor (vWF), has long been known to be a key player in thrombus formation at sites of vascular damage, vWF act as a carrier protein for blood clotting FVIII and promotes platelet adhesion and aggregation at sites of vascular injury. The release of vWF is increased when endothelial cells are activated or damaged. Therefore, plasma vWF level is considered a marker of endothelial dysfunction, a condition that predisposes to atherosclerosis and thrombosis. Because of its direct role in hemostasis, and its indirect role as a marker of endothelial dysfunction, vWF is a potential risk indicator for cerebrovascular disease.2 Recent studies revealed that abnormal elevation of any or all lipids and/or lipoproteins which known as hyperlipidemia is considered a modifiable risk factors that play a crucial role in the pathogenesis of atherosclerosis and biological etiology of the ischemic stroke. Hyperlipidemia usually classified as either familial caused by specific genetic abnormalities, or acquired when resulting from endocrine, renal or hepatic diseases. 4 Deposition of cholesterol and cholesteryl ester from the plasma lipoproteins into Background and objective: Ischemic stroke is classically characterized as a neurological deficit attributed to an acute focal injury of the central nervous system by a vascular cause, it occurs as a result of obstruction by a blood clot (thrombus) or plugs within a blood vessel supplying blood to the brain. In this study, we investigated the association of von Willebrand factor-antigen and serum lipids with ischemic stroke. Methods: The following retrospective study was conducted on 138 participants; 88 patients with ischemic stroke and 50 healthy controls. Results: In crude analyses, Plasma von Willebrand factor antigen, fasting serum Total cholesterol, Triglycerides and LDL-C were significantly higher in patients with ischemic stroke than in controls (P <0.001, P <0.001, P = 0.003, P <0.001, respectively), while the difference in the level of serum HDL-C between patients with ischemic stroke and control was significantly lower (P = 0.023). Conclusion: These data suggest that von Willebrand factor, total cholesterol, triglycerides, LDL-C and HDL-C gives some contribution to stroke risk even in the elderly and that von Willebrand factor antigen and lipid profile assessment must be taken into account in estimating the individual risks of stroke.
Introduction Giardiasis is an infectious disease, usually starts with ingestion of the cysts through contaminated food or water, followed by excystation and release of the trophozoite in the small intestine. 1-3 It is a dangerous disease that affect children and adults and lead to malabsorbtion syndrome and weight loss in the infected persons. 4,5 Giardia lamblia has a unique metabolic pathway that allows it to survive and multiply by scavenging nutrients from the host; 6 which commonly causes acute or chronic diarrhea, abdominal pain, nausea, anorexia, vomiting, flatulence, weight loss and fatigue. However, a significant proportion of infected individuals are completely asymptomatic. 7 Trophozoites, the active forms of the parasite, colonize the upper small intestine by adhering to the apical surface of the epithelium which leads to malnutrition as well as malabsorption in the infected persons. 6,8 Once a person becomes infected with Giardia lamblia many biochemical changes are often found during infectious period like alteration in lipid profiles, alteration of some micronutrients, certain enzymes and vitamins. 9-12 Micronutrients such as iron, chromium, copper, manganese, selenium and zinc are essential cofactors of enzymes and as organizers of the molecular structures of the cell. 13,14 The main impact of human intestinal parasitic infections is its relation to trace elements and vitamin deficiency, which may have Background and objective: The micro aerophilic flagellated protozoan parasite, Giardia lamblia, is the most common intestinal pathogen infecting human and a major diarrheal disease found throughout the world. It can cause acute or chronic diarrhea contributing to nutritional status, growth, and intellectual function. This prospective study evaluated the association between giardiasis and blood minerals disorders in Hawler city. Methods: The present study was carried out at the College of Medicine, Hawler Medical University, via collaborating study between the Department of Biochemistry and the Department of Microbiology. The study was carried out on 50 patients with giardiasis and 65 age and sex matched healthy controls. Examination of fecal samples for detection of Giardia cyst and/or trophozoite was carried out using the direct wet smear, while serum zinc, iron and copper levels were measured for both groups spectrophotometrically using commercial diagnostics kits. Results: The levels of serum zinc and iron in patients infected with Giardia lamblia were significantly lower (P <0.01) compared to normal healthy control, while the differences in the level of serum copper was significantly higher (P <0.01). Conclusion: : In the frame of this work we delineate that Giardia lamblia infection had a significant effect on serum trace elements reflected by, significant decrease in serum zinc and iron levels, whereas the level of copper increased too in a statistically significant manner.
