RATIONALE: Nei-like 2 (NEIL2) is a DNA glycosylase enzyme that can excise oxidized damaged DNA bases like 5-hydroxyuracil. Neil2-null mice show more innate inflammatory response in the lungs stimulated by oxidative stress-inducing challenges. Here we hypothesized that NEIL2 regulates cat dander extract (CDE)-induced allergic airway inflammation and sensitization. METHODS: Wild-type (WT) and Neil2 KO mice were sensitized with 5 intranasal doses of CDE and challenged with CDE to elucidate allergic airway inflammation. Allergic airway inflammation and the lung mRNA expression of 84 genes known to be associated with allergic-inflammation were quantified. RESULTS: Multiple challenge with CDE in WT mice induced allergic inflammation characterized by an increase in eosinophilic airway inflammation, stimulation of mucin in airway epithelial cells, and levels of cat dander specific IgE in serum at 72-hours post challenge. Multiple CDE challenges in Neil2 KO mice enhanced these increases observed in WT mice. PCR array and qPCR identified Ccl11 and Il5ra as two eosinophilia-associated genes that were higher 4 h-post challenge in Neil2 KO mice. CONCLUSIONS: These observations indicates that NEIL2 suppress CDE-induced eosinophilic airway inflammation. NEIL2 may be a novel target for attenuating eosinophilic airway inflammation and antigenspecific IgE.

Introduction:The Brachial artery (BA) is used for various clinical procedures, particularly with respect to blood pressure monitoring and percutaneous arterial catheterization. A sound and accurate knowledge of anatomical variations regarding the BA especially higher division of BA are important for clinical diagnosis and surgical management of diseases of superior extremities. Because of such importance, we undertook the study to investigate the course and branching pattern of BA with special reference to high up division and to discuss its clinical significance. Subjects and Methods: A total of 50 formalin fixed upper limbs were utilized in the present study. The branching pattern of BA variability was studied. Results: Variation in the branching pattern of BA was observed in 8 specimens (16%). The variations encountered were superior brachial artery (2%), deep brachial artery (2%), superficial ulnar artery (2%), and superficial radial artery (10%). Conclusion: The present study highlights the necessity for a thorough knowledge of brachial artery with respect to the high up division, to enhance the efficacy of diagnosis and for better surgical management of vascular diseases and injuries.
RATIONALE: Leukocytoclastic vasculitis (LCV) can be due to drugs, infections, connective tissue diseases, allergens, or be idiopathic. METHODS: A medical report of LCV induced by unknown cause was reviewed for possible causative factors. RESULTS: A 26-year female had conjunctivitis, pruritic dermatitis for 8 months with use of Diclofenac and prednisolone due to joints symptoms. Two years before she had birch pollen allergy without skin symptoms. The new rash did not respond to antihistamines. NSAIDS were used for the joint symptoms but worsened the rash. The patient had pruritic erythematous rashes on the face and extremities and conjunctival erythema. Leukopenia, moderate eosinophilia (1.9 G/L), increased erythrocyte sedimentation rate and fibrinogen, increased total IgE (1198 IU/ml), and increased levels of specific IgE (>0.35 kU/l) to D. pteronyssinus., D. farinae, rBetv1 and rBetv2, positive antinuclear antibody and cANCA (anti-proteinase-3) were noted. EBV was found in the pharynx, blood and saliva. Skin biopsy showed leukocytoclastic vasculitis with eosinophils and lymphocytes within the vessel walls. According to the definitions of the Chapel Hill Consensus Conference and ACR Criteria LCV was diagnosed associated with EBV, IgE and NSAIDS. Antiviral drugs, prednisolone, and antihistamines provided decreased skin and eyes findings. CONCLUSIONS: EBV in association with allergens may be associated with development of LCV. Detection of triggers of vasculitis allows etiological treatment and prevention of complications.
