Microphthalmia-associated transcription factor (MITF) is involved in white spotting and deafness associated with lack of pigmentation in human and mice. In the present study, we employed MITF-associated markers to evaluate MITF as a candidate for canine congenital sensorineural deafness (CCSD) in Dalmatian dogs. We performed an association study using MITF flanking and intragenic markers for 88 Dalmatian dogs of different hearing and eye pigmentation status. A significant association was identified for MITF-related markers with CCSD and blue iris colour. We conclude that MITF might play a role in CCSD and blue eye colour in Dalmatian dogs.
Zusammenfassung
Gegenstand und Ziel: Für den Merle-Faktor homozygote Hunde weisen oft multiple Augenmissbildungen auf. Zwar ist die für den Merle-Phänotyp kausale SINE-Insertionsmutation im SILV-Gen auf dem Hundechromosom 10 inzwischen bekannt, doch konnte der Zusammenhang zwischen dieser Mutation und den bei homozygoten Merle-Hunden auftretenden okulären Defekten bisher nicht geklärt werden. Material und Methoden: Für vier Altdeutsche Hütehunde und vier Aus tralian Shepherds wurden die aus der Fellfarbe abgeleiteten Merle-Genotypen MM, Mm und mm über Gen-tests bestätigt. Es erfolgten Sequenz- und Haplotypanalysen für das SILV-Gen. Mittels Markern wurde eine mögliche Assoziation der Merle-Genotypen zu dem MITF-Gen, das die SILV-Expression reguliert, untersucht. Ergebnisse: Alle MM-Hunde zeigten multiple Augenanomalien wie Kolobome, Mikrophthalmie, Dyskorie und persistierende Pupillarmem-bran. In der Länge der SINE-Insertion sowie in den übrigen kodierenden Sequenzen des SILV-Gens ließen sich keine Unterschiede zwischen Hunden mit und ohne Augendefekte nachweisen. Ein mit den Merle-Genotypen vollständig segregierender Haplotyp war für das SILV-Gen feststellbar. Für drei MITF-Marker fand sich eine signifikante Assoziation zu den Augenmissbildungen der MM-Hunde. Schlussfolgerungen: Es ergaben sich keine Anhaltspunkte für Mutationen im SILV-Gen, die auf einen Zusammenhang mit den Augendefekten hindeuten. Die signifikante Assoziation eines Haplotyps zwischen den das MITF-Gen flankierenden Markern und den Augenanomalien könnte für eine Beteiligung des MITF-Gens an der Entwicklung dieser Missbildungen sprechen. Da dieser Untersuchung nur drei MM-Hunde zugrunde lagen, sind diese Resultate mit größter Vorsicht zu interpretieren und erst an umfangreichen Daten zu bestätigen. Klinische Relevanz: Unsere Untersuchungen geben erste Hinweise darauf, dass die häufig bei MM-Hunden vorkommenden Augenmissbildungen nicht durch Mutationen im SILV-Gen, sondern eventuell durch Mutationen im MITF-Gen bedingt sind. Nach Bestätigung dieser Befunde an einem größeren Material könnte die Aufklärung dieser Mutationen im MITF-Gen ermöglichen, die mit der Merle-Färbung verbundenen Augendefekte durch Selektionsmaßnahmen zu verhindern.
