Volatile compounds of cajá and taperebá fruits, both classified as Spondias mombin, but from different geographic origins, were extracted (and analyzed) using solid phase microextraction (SPME) and simultaneous distillation and extraction (SDE). Forty-eight compounds were identified in taperebá and 47 in cajá by SPME using a DVB/CAR/PMDS fiber. (E)-Caryophyllene (18.7%), ethyl butyrate (10.0%), and ethyl hexanoate (7.0%) were the most abundant components in taperebá volatiles extracted by SPME, whereas myrcene (41.1%) and beta-phellandrene (8.5%) were the major compounds in cajá. In the taperebá SDE extract, 46 substances were identified, and (Z)-caryophyllene (13.2%) and limonene (9.5%) were predominant. From the 42 substances found in the SDE extract of cajá, the major components were myrcene (38.0%) and p-cymene (6.2%). The two fruits showed similar chromatograms upon the use of SDE and SPME. These methods made it possible to determine 30 identical components in both fruits by using SDE and 32 by using SPME.
Ceiba pentandra (L.) GAERTNER (Bombacaceae) is a tropical tree and the capsules are known as kapok. It is widely used in traditional medicines as a diuretic, an antidiarrheic and an emollient.1) From the root bark of Ceiba pentandra, naphtoquinones have been reported with antimicrobial activities.2) From the bark, vavain (1) and its glucoside (2) have been isolated with their inhibitory effects on cyclooxygenase-catalyzed prostaglandin biosynthesis.3) On our studies of finding inhibitory active compounds against a-glucosidase from medicinal plants, the bark of Ceiba pentandra which was called Samauma-verdadeira in Bélem, Brazil and used for the treatment of diabetes, exhibited some inhibitory activities (data not shown). A new vavain glycoside (3) was isolated in addition to 1 and 2 in the EtOAc-soluble fraction and n-BuOH-soluble fraction although these compounds showed no activities against this enzyme.Compound 3 showed a blue color under UV light (2537 Å), and a brown color when heated with 10% H 2 SO 4 on TLC plate. The presence of phenolic hydroxyl group and carbonyl group was apparent from the absorption bands at 3566 (OH), 1655 (CϭO), 1508 and 1474 (aromatic) cm Ϫ1 in the IR spectrum. These observations on TLC and IR were quite similar to those of 1 and 2. The negative secondary ion (SI)-MS of 3 revealed [MϪH] Ϫ at m/z 637.1782, which corresponded to the molecular formula C 29 H 34 O 16 . This suggested that compound 3 had an additional pentose sugar to vavain glucoside (2: C 24 H 26 O 12 ). Since 1 H-and 13 C-NMR data of 3 were similar to those of 2, it was speculated that a pentose sugar was attached to the glucose moiety of 2. This was proved by a correlation between the anomeric proton (d 4.94, Jϭ7.5 Hz) and C-3Ј carbon (d 150.7), as well as a correlation between the anomeric proton (d 4.70, Jϭ2.0 Hz) and a primary alcohol carbon (d 66.9), in the heteronuclear multiple bond connectivity (HMBC) spectrum. Since the anomeric carbons attached to the anomeric protons at d 4.94 and d 4.70 were assigned at d 101.3 and d 108.3 by the heteronuclear multiple quantum coherence (HMQC), respectively, the terminal pentose sugar was connected to the 6-hydroxyl group of the glucosyl moiety. This was also supported by HMBC connectivities between a hydroxyl proton ( , the terminal pentose sugar was determined as a-arabinofuranoside, whose 13 C-NMR chemical shifts demonstrated the characteristic pattern.5) To confirm the configuration of sugars, 3 was hydrolyzed to afford 1 and D-glucose and L-arabinose detected by the chromatography. Thus, the structure of 3 was elucidated as 5-hydroxy- ExperimentalGeneral Procedures Melting points were determined on a Yanaco micro melting point apparatus and uncorrected. UV and IR spectra were recorded using a Hitachi U-3010 spectrometer in MeOH and on a Shimadzu FTIR-8200 spectrometer in KBr, respectively. Optical rotation were measured on JASCO DIP-370 digital polarimeter in EtOH or pyridine.1 H-and 13 C-NMR spectra were taken on a Varian VXR-500 instrument at 500 MHz, and 125 MHz...
