Zusammenfassung: Es wird eine Mikromethode zur Bestimmung freier bzw. gebundener N-Acetyl-Neuraminsäure (NANA) in Glykoproteinen auf der Grundlage der Periodatoxidation und Thiobarbituratreaktion beschrieben. Die Methode zeichnet sich durch einfache Durchführbarkeit bei Verwendung des Mikrolitersystems Eppendorf aus. Die Nachweisempfindlichkeit liegt bei 0,2 nmol NANA im Testansatz. Die Präzision in der Serie beträgt 3,0%, die Langzeitpräzision 4,3% (VK). Die Richtigkeit der erhaltenen Ergebnisse wurde durch Vergleichsuntersuchungen mit der Makromethode nach Warren ((1959), J. Biol. Chem. 234, 1971Chem. 234, -1979 an Referenzmaterial (Sialyllactose, Transferrin, Fibrinogen) unter Beweis gestellt. The intraassay precision comprises 3.0% CV, the interassay precision 4.3% CV. A specific, sensitive, and reliable radioimmunoassay for serum-conjugated deoxycholic acid has been developed with antiserum obtained after immunization of rabbits with deoxycholic acid--bovine serum albumin conjugate. The displacement curve of glyco [3H] deoxycholic acid was linear on a logit-log plot from 7.5 to 320 pmol of unlabeled glycodeoxycholic acid. At 50% displacement of bound label, the cross-reactivity of taurodeoxycholic acid was 100%, deoxycholic acid 30%, taurocholic acid 3%, and glycocholic acid 2%. No cross-reactivity was observed with free cholic acid, conjugated or free chenodeoxycholic acids and conjugated or free lithocholic acids. Fasting serum-conjugated deoxycholic acid concentrations in 10 healthy volunteers ranged from 0.18 to 0.92 mumol/1. Over a period of 5 hours following 0.5 g oral cholate administration the serum-conjugated deoxycholic acid concentration did not change in 5 fasting healthy persons, whereas an initial increase of serum cholic acid was observed.
Zusammenfassung: Es wird eine rasch durchfuhrbare Methode zur Bestimmung von Lipoprotein-gebundenen Kohlenhydraten in Humanseren beschrieben. Die Methode beruht auf der Abtrennung der Lipoproteine aus Vollserum durch sequentielle Präzipitation mit Natriumphosphorwolframat und Magnesiumchlorid. Nach Reinigung der abgetrennten Lipoproteine durch zweimalige Repräzipitation werden Neutralzucker, Aminozucker und N-Acetylneuraminsäure (Sialinsäure) kolorimetrisch bestimmt. Die Variationskoeffizienten für die Präzision in der Serie liegen zwischen 3,2% und 7,3%. Die Bestimmung der Lipoprotein-gebundenen Kohlenhydrate nach Präzipitation wird verglichen mit den Ergebnissen nach Ultrazentrifugation. Die Gehalte an Neutralzucker, Aminozucker und N-Acetylneuraminsäure in den 3 Lipoproteinklassen VLDL, LDL und HDL stoffwechselgesunder Kontrollpersonen werden mitgeteilt. The method is based on a successive precipitation of lipoproteins with sodium phosphotungstate and magnesium chloride. After purification of the separated lipoproteins by a twofold reprecipitation, a colorimetric determination of hexoses, hexosamines and N-acetylneuraminic acid (sialic acid) is performed. The coefficients of Variation in the series are between 3.2% and 7.3%. The results for lipoprptein-bound carbohydrates after precipitation are compared with the corresponding values after Separation by ultracentrifügätion. The concentrations of hexoses, hexosamines and N-acetylneuraminic acid in the 3 jipopfotein classes VLDL, LDL and HDL of control persons are reported. Einführung
RESUMO: o objetivo deste trabalho é verificar o impacto do comportamento das variáveis macroeconômicas sobre o desempenho do setor têxtil e do vestuário de Santa Catarina no período de 1990 a 2003. O Valor da Produção Industrial, calculado pelo IBGE, foi considerado como representativo do desempenho do setor, e seu comportamento comparado com exportações, importações, PIB brasileiro e catarinense, taxa de câmbio e taxa real de juros, através de análise de gráficos e de correlação. O Valor da Produção Industrial tem uma relação inversa estatisticamente significante com a taxa real de juros e muito próxima de significante em relação às exportações. Conclui-se que a queda das exportações, influenciada pela taxa de câmbio e taxa de juros teve papel decisivo na crise do setor, mais que a elevação das importações.
