In the three-dimensional extracellular matrix of the insect cuticle, horizontally aligned microfibrils composed of the polysaccharide chitin and associated proteins are stacked either parallel to each other or helicoidally. The underlying molecular mechanisms that implement differential chitin organization are largely unknown. To learn more about cuticle organization, we sought to study the role of chitin deacetylases (CDA) in this process. In the body cuticle of nymphs of the migratory locust Locusta migratoria, helicoidal chitin organization is changed to an organization with unidirectional microfibril orientation when LmCDA2 expression is knocked down by RNA interference. In addition, the LmCDA2-deficient cuticle is less compact suggesting that LmCDA2 is needed for chitin packaging. Animals with reduced LmCDA2 activity die at molting, underlining that correct chitin organization is essential for survival. Interestingly, we find that LmCDA2 localizes only to the initially produced chitin microfibrils that constitute the apical site of the chitin stack. Based on our data, we hypothesize that LmCDA2-mediated chitin deacetylation at the beginning of chitin production is a decisive reaction that triggers helicoidal arrangement of subsequently assembled chitin-protein microfibrils.
Attention-deficit hyperactivity disorder (ADHD) seriously affects children’s health, and the gut microbiome has been widely hypothesized to play a role in the regulation of ADHD behavior. The present study aims to the biosynthesize of zinc oxide nanoparticles (ZnONPs) by using Acinetobacter johnsonii strain RTN1, followed by their characterization through state-of-the-art material characterization techniques, viz., UV–vis spectroscopy, Fourier transform infrared spectroscopy, and transmission and scanning electron microscopic analyses with energy dispersive spectrometry. Moreover, we investigated and compared the population composition of gut microbiota and their susceptibility to biogenic ZnONPs between healthy and ADHD children based on the traditional plate method and 16S rRNA amplicon sequence analysis. The antibacterial effect of ZnONPs against gut bacteria was also determined by measurement of live cell number, living/dead bacterial staining test, and flow cytometry observation. The present study revealed that the number of live gut bacteria in healthy children was more than 10-fold higher than that in ADHD children; however, the community structure of gut bacteria has changed, while greater diversity was found in gut bacteria from ADHD children. In addition, we found that the number of live gut bacteria in healthy and ADHD children was reduced by ZnONPs, which shows an increased and reduced effect in composition of gut bacteria from healthy and ADHD children, respectively. It was also noted that the main mechanism of ZnONPs may be to inhibit the growth of gut bacteria rather than to kill them, while the nanoparticle-resistant strains in healthy children is also different from that in ADHD children. Some representative bacteria, in particular nanoparticle-resistant bacteria, were successfully isolated and identified. Overall, this study revealed the potential correlation of ADHD with gut bacteria and provided a new possibility to prevent ADHD by the combination of nanoparticle and its resistant bacteria.
With the outset of Industrial Revolution 4.0 (IR 4.0), every sector is escalating to get enrichment out of it, whether they are research- or industry-oriented. The Architecture Engineering and Construction (AEC) industry lags a bit in adopting it because of its multi-faceted dependencies and unique nature of work. Despite this, a trend has been seen recently to hone the IR 4.0 multitudes in the AEC industry. The upsurge has been seen in the usage of Immersive Technologies (ImTs) as one of the disruptive techniques. This paper studies the literature based on ImTs, which are Virtual Reality (VR), Augmented Reality (AR), and Mixed Reality (MR) integrating with Building Information Modelling (BIM) in the AEC sector. A total number of 444 articles were selected from Scopus following the preferred reporting items for systematic reviews and meta-analysis (PRISMA) protocol of reviewing the literature. Among the selected database, 64 papers are identified as the result of following the protocol, and the articles are divided into eight domains relevant to the AEC industry, namely client/stakeholder, design exploration, design analysis, construction planning, construction monitoring, construction health/safety, facility/management, and education/training. This study adopts both a scientometric analysis for bibliometrics visualization and a critical review using Strength Weakness Opportunity Threat (SWOT) analysis for finding gaps and state of play. The novelty of this paper lies in the analysis techniques used in the literature to provide an insight into the literature, and it provides directions for the future with an emphasis on developing sustainable development goals (SDGs). In addition, research directions for the future growth on the adoption of ImTs are identified and presented based on categorization in immersive devices, graphical/non-graphical data and, responsive/integrative processes. In addition, five subcategories for each direction are listed, citing the limitations and future/needs. This study presents the roadmap for the successful adoption of ImTs for industry practitioners and stakeholders in the AEC industry for various domains. The paper shows that there are studies on ImTs with or without BIM; however, future studies should focus on the usage of ImTs in various sectors such as modular integrated construction (MiC) or emerging needs such as SDGs.
This article presents the results from exploring the impact of using a parametric design tool on designers’ behavior in terms of using design patterns in the early conceptual development stage of designing. It is based on an empirical cognitive study in which eight architectural designers were asked to complete two architectural design tasks with similar complexity, respectively, in a parametric design environment and a geometric modeling environment. The protocol analysis method was employed to study the designers’ behavior. In order to explore the development of design patterns in the empirical data, Markov model analysis is utilized. Through Markov models analysis of the parametric design environment and geometric modeling environment results, it was found that there are some significantly different design patterns being used when designing in a parametric design environment compared to designing in a geometric modeling environment. The article articulates these differences and draws conclusions from these results.
In 2010, the outbreak of a disease with symptoms similar to bacterial brown stripe was observed in rice seedlings planted in a perlite culture system in China. The causal bacterium was identified as Acidovorax avenae subsp. avenae based on its biochemical and physiological characteristics, cellular fatty acid composition, Biolog data, specific PCR detection and 16S rRNA gene sequence analysis. The bacterial isolates caused similar symptoms after inoculation of rice seedlings. This report is the first of bacterial brown stripe of rice in a soil-less culture system caused by A. avenae subsp. avenae in China.
