Die Kondensation von 2‐Picolyl‐magnesium‐bromid mit ω‐Halogenolefinen und mit ω‐Halogen‐acetalen wird untersucht. Die erzielten guten Ausbeuten an 2‐Pyridylolefinen bzw. 2‐Pyridylacetalen beweisen die grundsätzliche Anwendbarkeit der Magnesiumverbindung an Stelle von Lithium für derartige Synthesen. Zur Gewinnung von 3‐(2‐Pyridyl)‐propionaldehyd und 4‐(2‐Pyridyl)‐butyraldehyd werden verschiedene Wege untersucht. Als geeignet und ausbeutemäßig günstig erwies sich derjenige über die Acetale und Hydrolyse dieser. Die Struktur der pyridinsubstituierten aliphatischen Aldehyde wird durch Oxydation zu den entsprechenden Karbonsäuren gesichert. Es werden weiterhin durch Oximierung und anschließende Quaternisierung potentielle Cholinesterase‐Reaktivierungsmittel synthetisiert, sowie Semikarbazone und Thiosemikarbazone.

Mitteilung salicylsaure nicht im Verlaufe der Trennung durch das Flieljmittel verseift werden, sondern es ist eine thermische Spaltung wahrend des Trocknens zu vermuten. Um diese Annahme zu beweisen, wurde Acetylsaure in methanolischer Losung auf die Sorptionsschicht aufgetragen und einmal i. Vak. und andererseits bei 80" getrocknet. Danach wurden beide Flecke mit Methanol in der Kalte extrahiert und die UV-Spektren mit Standardspektren von Salicylsaure und Acetylsalicylsaure verglichen, Aus Abb. 3 ist zu ersehen, da13 bei 80" die Acetylsalicylsaure nahezu vollig gespalten wird. Dagegen entsteht beim Troclrnen i. Vak. und nachfolgender Extraktion nur ein geringer Anteil Salicylsaure, der im wesentlichen durch die Extraktion bedingt sein diirfte. ZusammenfassungEinige zum Bespriihen papierchromatographisch getrennter Arzneistoffe benutzte Reagenzien von allgemeiner Bedeutung sind im wesentlichen auch zum Nachweis auf Kieselgel-und Aluminiumoxyd-Sorptionsschichten geeignct. In manchen Fallen spielt jedoch die Art des Sorptionsmittels fur die Nachweisempfindlichkeit eine Rolle, so z. B. beim Chlorieren von Aminen und nachfolgendem Bespriihen rnit Kaliumjodid-Benzidin. Mit Leuchtpigmenten impragnierte, fluoreczierende Sorptionsschichten sind zum Nachweis UV-absorbierender Substanzen allgemeiner anwendbar, als das Bespriihen mit dem Bluoreszenzfarbstoff Rhodamin; aul3erdem eignen sie sich fur quantitative Trennungen.Am Beispiel der Acetylsalicylsaure konnte gezeigt werden, da13 durch thermische Zersetzung beim Trocknen der Chromatogramme Spriihreagenzien ansprechen, mit denen sich die Substanz selbst nicht nachweisen lafit. Es w i d uber die Darstellung von a-H ydroxynitrilen 'uon 0-(2-Pyricly1)-alkanalen berichtet. H ydrol yse dieser Verbindungen fuhrte zu 2-Pyridyl-2-h ydrox ykarbonsauren bzw. ihren Amiden. DieRildung sekundarer Alkohole aus 3-(2-P yridy1)-propionaldeh yd wird untersucht. Kondensationen der w-(2-Pyridy1)-alkanale mit einigen Nitroalkanen wurden durchgefihrt. Reduktion der erhaltenen Nitroalkohole fuhrtezu den Aminoalkoholen. Die Kondensation mit zwei aktisce Meth ylengruppen enthaltenden Verbindungen wird untersucht. In der ersten Mitt.') haben wir u. a. einen in guten Ausbeuten gangbaren Weg zur Synthese zweier Pyridylalkanale, des 3-(2-Pyridyl)-propionaldehyds und des l) E. Pmfft und R. Stump/. J. prakt. Chem. (im Druck). Profft und StumpfArcHiv der Pharmazie 2-(2-Pyridyl)-butyraldehyds, aufgezeigt. Diese beiden Aldehyde sind in pharmazeutischer Hinsicht recht interessante Verbindungen. Sie bieten die Moglichkeit, zu w-(2-Pyridyl)-a-hydroxynitrilen zu gelangen, iiber die bisher wenig bekannt geworden ist. Jungst erst berichteten Mathes und SauermiZch2) iiber die Darstellung der Cyanhydrine der Pyridinaldehyde selbst, gewonnen durch Umsetzung der Aldehyd-hydrochloride mit Kaliumcyanid in waBriger Losung.Bequemer erschien uns zur Darstellung der Hydroxynitrile der Weg, der von den SO,-Anlagerungsprodukten, den a-Hydroxysulfonsauren, ausgeht, da diese aus den Acetalen...
