The patterns of exochorion ornaments on eggs of seven South American Lutzomyia sand fly species were analyzed by scanning electron microscopy (SEM): Lutzomyia (Lutzomyia) cruzi (Mangabeira 1938), Lutzomyia (Micropygomyia) evandroi (Costa Lima and Antunes 1936), L. (Nyssomyia) intermedia (Lutz and Neiva 1912), L. longipalpis (Lutz and Neiva 1912), L. migonei (Franca 1920), L. (Nyssomyia) neivai (Pinto 1926), and L. renei (Martins, Falcao, and Silva 1957). Different patterns were observed, which showed the distinction between some species. Egg ornaments in L. cruzi and L. longipalpis appear as single, parallel, unconnected ridges, whereas eggs of L. migonei appear as single, parallel, connected ridges. Eggs of L. (Nyssomyia) intermedia and L. (N.) neivai present a new variation of the single, unconnected, parallel ridges pattern: small tubercles are present, distributed between the ridges. Eggs of L. renei present an elliptical pattern, with most structures connected by straight ridges. Eggs of L. (M.) evandroi present a polygonal pattern, with alternate rows of small and large hexagons. Our data emphasize the advantages of the SEM approach in the study of the exochorion patterns of Lutzomyia eggs and in the distinction of the sand fly species.
RESUMO-Este trabalho teve como objetivo avaliar a substituição total ou parcial do ágar por amidos de milho e mandioca e da esterilização física em autoclave pela esterilização química com hipoclorito de sódio (NaClO) no desenvolvimento de mudas de abacaxizeiro 'Gold' (Ananas comosus). Como agentes geleificantes, foram utilizados: M1: ágar (6,0 g L -1 ); M2: amido de milho (60,0 g L -1 ); M3: ágar (3,0 g L -1 ) + amido de milho (30,0 g L -1 ), e M4: ágar (3,0 g L -1 ) + amido de mandioca (30,0 g L -1 ). As esterilizações foram feitas por meio da autoclavagem a 121°C e da utilização de soluções de NaClO nas concentrações de 0,003% para o enxágue das vidrarias e 0,0003% para adição ao meio de cultivo. Após um mês de cultivo in vitro, não houve influência negativa do NaClO e dos agentes geleificantes no enraizamento das brotações para a maioria dos parâmetros avaliados. As mudas produzidas in vitro foram levadas para aclimatização em casa de vegetação, por 90 dias. Na aclimatização, as mudas oriundas dos tratamentos com esterilização química não diferiram das oriundas dos meios autoclavados em nenhum parâmetro avaliado, enquanto a combinação ágar + amido de milho proporcionou resultados inferiores entre os agentes geleificantes. STARCH AND SODIUM HIPOCHLORITE ON IN VITRO ROOTING OF PINEAPPLE 'GOLD' AND ITS EFFECTS ON ACCLIMATIZATIONABSTRACT -This study aimed to evaluate the total or partial replacement of agar by corn starch and cassava starch and physical sterilization in autoclave for chemical sterilization with sodium hypochlorite (NaClO) in development of pineapple 'Gold' plantlets. The gelling agents used were: M1: agar (6.0 g L -1 ); M2: corn starch (60.0 g L -1 ); M3: agar (3.0 g L -1 ) + corn starch (30.0 g L -1 ) and M4: agar (3.0 g L -1 ) + cassava starch (30.0 g L -1 ). The sterilizations were made by autoclaving at 121°C or by using the solutions of NaClO at the concentrations of 0,003% to rinse the glassware and 0,0003% for addition to the culture medium. After one month of in vitro cultivation, it was observed no negative influence of NaClO and gelling agents in the rooting in vitro of shoots for most parameters evaluated. The plantlets derived from in vitro phase were taken to acclimatization in greenhouse for 90 days. During the acclimatization, the plantlets derived from treatments with chemical sterilization did not differ from the ones derived of autoclaved media to any parameter evaluated, whereas the combination agar + corn starch provided inferior results among gelling agents.
RESUMO-O objetivo deste trabalho foi avaliar a eficiência da esterilização química do meio de cultura e de agentes geleificantes alternativos e sua influência no desenvolvimento in vitro e ex vitro de plantas de abacaxizeiro 'Vitória'. O meio de cultivo utilizado foi composto pelos sais do MS e vitaminas de White, com 30 g L -1 de sacarose e 100 mg L -1 de mioinositol. Foram realizadas duas formas de esterilização dos meios de cultivo e das vidrarias: em autoclave a 121°C e com solução de hipoclorito de sódio (NaClO) a 0,003% para enxágue das vidrarias e 0,0003% no meio de cultivo. Os agentes geleificantes testados foram:). Após 30 dias de cultivo in vitro, constatou-se que o NaClO e os agentes geleificantes alternativos não interferiram no crescimento e no desenvolvimento das mudas. Parte das mudas de cada tratamento foi aclimatizada durante 90 dias, após os quais se verificou que não houve influência dos agentes geleificantes no crescimento destas, e, para a maioria dos parâmetros avaliados, as mudas oriundas dos tratamentos envolvendo a esterilização química apresentaram resultados semelhantes ou superiores às dos meios autoclavados. CHEMICAL STERILIZATION AND ALTERNATIVE GELLING AGENTS IN THE IN VITRO VEGETATIVE PROPAGATION OF PINEAPPLE 'VITÓRIA'ABSTRACT -The objective of this study was to evaluate the efficiency of chemical sterilization of the culture medium and alternative gelling agents and their influence on development in vitro and ex vitro plants of pineapple 'Vitória'. The culture medium used was composed of MS salts and White vitamins, with 30 g L -1 of sucrose and 100 mg L -1 of myo-inositol. Two forms of sterilization of the media culture and the glassware were performed: in autoclave at 121°C and with solution of sodium hypochlorite (NaClO) at 0.003% to rinse the glassware and 0.0003% in the culture medium. The gelling agents tested were: M1: agar (6.0 g L ). After 30 days of in vitro cultivation was found that the NaClO and alternative gelling agents did not affect the growth and development of plantlets. Part of the plantlets of each treatment was acclimatized during 90 days, after which it was found that there was no influence of gelling agents on plantlets growth. For most of the parameters evaluated, the plantlets derived from treatments involving the chemical sterilization showed results similar or superior to the autoclaved media.
