Don't take this antivitamin! 2-Phenylethynylcobalamin was prepared in a newly developed radical reaction using cob(II)alamin and 1-iodo-2-phenylethyne. It has an exceptionally short organometallic bond and is a remarkably light-stable and heat-resistant organometallic cobalamin. It is bound well by two important proteins of the human B12 transport system and has properties that are as expected for a new type of an "antivitamin B12 ".

Antivitamins B12 have a growing biomedical impact as robust B12-dummies. Here, the potential antivitamin B12 2,4-difluorophenyl-ethynyl-cobalamin (F2PhEtyCbl) was prepared and its 3D-structure was studied in solution and in the crystal. Chemically inert F2PhEtyCbl resisted thermolysis of its (Co-C)-bond at 100°C, was stable to bright day light and also remained intact during prolonged storage in aqueous solution at room temperature. It bound to the human B12-processing enzyme CblC with high affinity (KD = 130 nM) in the presence of the co-substrate glutathione (GSH). F2PhEtyCbl withstood tailoring by CblC, and it also stabilized the ternary complex with GSH. The crystal structure of this abortive assembly afforded first insights into the binding interactions of an antivitamin B12 with CblC, as well as into the organization of GSH and a base-off cobalamin in the active site of this enzyme.

Alkynylcorrinoids are a class of organometallic B12 derivatives, recently rediscovered for use as antivitamins B12 and as core components of B12-based biological vectors. They feature exceptional photochemical and thermal stability of their characteristic extra-short Co–C bond. We describe here the synthesis and structure of 3-hydroxypropynylcobalamin (HOPryCbl) and photochemical experiments with HOPryCbl, as well as of the related alkynylcobalamins: phenylethynylcobalamin and difluoro-phenylethynylcobalamin. Ultrafast spectroscopic studies of the excited state dynamics and mechanism for ground state recovery demonstrate that the Co–C bond of alkynylcobalamins is stable, with the Co–N bond and ring deformations mediating internal conversion and ground state recovery within 100 ps. These studies provide insights required for the rational design of photostable or photolabile B12-based cellular vectors.

Geburtstag gewidmet Die biologisch wichtigen Rollen der B 12 -Cofaktoren Coenzym B 12 (1, 5'-Desoxyadenosylcobalamin) und Methylcobalamin (2) sind mit ihrer metallorganischen Chemie und Redoxchemie verbunden. [1,2] Enzym-gesteuerte metallorganische Reaktionen, die Homolyse der schwachen Co-C-Bindung von Coenzym B 12 (1) und eine Nucleophil-induzierte heterolytische Abstraktion der Cobaltgebundenen Methylgruppe von Methylcobalamin (2), sowie Prozesse im (formal) umgekehrten Sinne, stellen die bekannten wichtigsten Schritte in B 12 -abhängigen Enzymen dar. [3][4][5][6] Um mçgliche biologische Funktionen der B 12 -Cofaktoren, wie 1 und 2, zu erforschen, wurde erheblicher Aufwand in Untersuchungen der chemischen Eigenschaften von metallorganischen B 12 -Derivaten gesteckt. [1,7] Ein Merkmal von metallorganischen B 12 -Derivaten ist ihre effiziente und selektive homolytische Spaltung der Co-C-Bindung durch Thermolyse oder Bestrahlung mit sichtbarem Licht. [8,9] Metallorganische Arylcobalamine weichen von dieser Regel zum Teil ab und weisen Thermolysebeständige Co-C sp 2-Bindungen auf. Sie repräsentieren eine neue Linie konzipierter B 12 -Antimetaboliten und sind potenzielle Antivitamine B 12 .[10] In diesem Zusammenhang kçnnen auch metallorganische Alkinyl-B 12 -Derivate mit einer Co-C sp -Bindung von besonderem Interesse sein. Es gibt einen überraschenden Mangel an relevanten Informationen über Alkinylcorrine, [1,7] und aus den älteren Arbeiten über Alkinylcobalamine [11][12][13] sind nur weitgehend vorläufige analytische und strukturelle Informationen verfügbar. Metallorganische B 12 -Derivate mit einem direkt Cobalt-gebundenen Alkinyl-Liganden haben außergewçhn-liche Struktureigenschaften und chemische Reaktivitäten, wie wir hier für Co b -2-Phenylethinylcobalamin (3; Schema 1) berichten. Co b -2-Phenylethinylcobalamin (3) wurde in einer Eintopfreaktion in Sauerstoff-freier wässriger Lçsung hergestellt, beginnend mit Aquocobalaminchlorid (6), 2-Phenylethinyliodid und Triethylammoniumformiat (Schema 2). Die Formiatreduktion von 6 lieferte Cob(II)alamin (4; siehe Lit.[10]), das mit 2-Phenylethinyliodid über eine mutmaßliche radikalische Reaktion reagierte. Wir schlagen vor, dass die Bildung von 3 durch Rekombination eines 2-Phenylethinylradikals mit in situ generiertem 4 erfolgt (Schema 3). Das Organocobalamin 3 wurde in 68 % Ausbeute nach Kristallisation des rohen roten Isolats aus wässrigem Aceton erhalten.Das UV/Vis-Spektrum von 3 (siehe Hintergrundinformationen, Abbildung S1) zeigte ein Base-on-Cobalamin und war ähnlich dem von Vitamin B 12 (5).
