ABSTRAKRigidoporus microporus merupakan patogen penyebab penyakit jamur akar putih (JAP) pada karet yang sangat sulit untuk dikendalikan. Penggunaan jamur antagonis diharapkan mampu mengendalikan patogen tersebut karena memiliki kemampuan penghambatan melalui mekanisme kompetisi, antibiosis, atau parasitisme. Penelitian ini bertujuan mengevaluasi 10 isolat jamur antagonis dalam menghambat R. microporus penyebab penyakit JAP pada tanaman karet serta mengidentifikasi mekanisme penghambatannya secara in vitro. Penelitian dilaksanakan bulan Januari sampai April 2013, di Laboratorium Proteksi Tanaman, Balai Penelitian Tanaman Industri dan Penyegar (Balittri) Sukabumi. Penelitian terdiri dari: (1) uji kecepatan pertumbuhan koloni, (2) uji daya hambat, (3) uji metabolit sekunder, dan (4) (3) antagonistic mechanism. Each test using a completely randomized design. The antagonist fungi used are: Trichoderma virens, T. hamatum, T. amazonicum, Eupenicillium javanicum, Penicillium simplicissimum, P. citrinum, P. pinophilum, Paecilomyces lilacinus, Aspergillus fijiensis, and Hypocrea atroviridis (=T. atroviride), in which each of these fungi repeated 3 times. The results showed that the Trichoderma genus have inhibitory capability against R. microporus stronger than the others. Four genera of Trichoderma fungi have better competition mechanism against R. microporus compared to Penicillium, Eupenicillium, Paecilomyces, and Aspergillus. In addition, T. virens and H. atroviridis also have parasitism ability, while P. lilacinus and E. javanicum have antibiosis mechanism against R. microporus. The antagonistic fungi which were most potential to be developed as a biological control for white root disease, i.e.: T. virens, T. hamatum, and H. atroviridis. <p>Ceratobasidium theobromae merupakan patogen penyebab penyakit vascular streak dieback (VSD) pada tanaman kakao, dapat menurunkan produksi bahkan kematian tanaman. Penyakit ini sulit dikendalikan karena berada dalam jaringan pembuluh. Penelitian bertujuan menguji keefektifan minyak cengkeh dan serai wangi serta ekstrak bawang putih terhadap C. theobromae penyebab penyakit VSD. Penelitian telah dilakukan pada perkebunan kakao rakyat hasil sambung samping berumur 2 tahun yang terserang penyakit VSD di Desa Andomesinggo, Kecamatan Besulutu, Kabupaten Konawe, Sulawesi Tenggara dari bulan April sampai Desember 2013. Rancangan yang digunakan adalah acak kelompok (RAK) 6 perlakuan dengan 6 ulangan, masing-masing perlakuan diamati 15 tanaman. Perlakuannya adalah minyak cengkeh, serai wangi, cengkeh + serai wangi, ekstrak bawang putih, fungisida kimia ditiokarbamat (sebagai pembanding) dan kontrol (tanpa perlakuan). Ekstrak dan minyak diaplikasikan setiap bulan dengan konsentrasi 5 ml/l, dengan cara menyemprotkan suspensi ke seluruh bagian tanaman (250 ml/pohon). Pengamatan gejala serangan, perkembangan penyakit, persentase dan intensitas serangan dilakukan setiap bulan, sedangkan tingkat keefektifan fungisida nabati dihitung pada akhir pengamatan. Hasil penelitian memperlihatkan minyak cengkeh, serai wangi, dan bawang putih dapat menurunkan persentase dan intensitas serangan penyakit VSD pada tanaman kakao. Persentase penurunan intensitas serangan terbesar dan nyata diperoleh pada perlakuan minyak cengkeh dan serai wangi, masing-masing 38,6% dan 31,6% dan keduanya potensial digunakan sebagai fungisida nabati untuk mengendalikan penyakit VSD.</p><p>Kata kunci: Kakao, VSD, minyak cengkeh, minyak serai wangi, ekstrak bawang putih</p><p>Vascular streak dieback disease caused by Ceratobasidium theobromae is a major disease that causes yield loss and even kill a mature cocoa tree. This disease is difficult to control due to located inside the vascular tissue. The objective of this research was to study the antifungal activity of clove oil, citronella oil and garlic extract against C. theobromae causing VSD disease. The research was carried out at cocoa plantations which derived from side grafting and attacked by VSD disease in Andomesinggo Village, Besulutu District, Konawe Regency, Southeast Sulawesi, from April to December 2013. The design used in this study was a randomized block design with 6 treatments and 6 replications, each treatment consisted of 15 plants. The treatments used were clove oil, citronella, clove + citronella, garlic extract, chemical fungicide (as a comparison) and control (without treatment). The extracts and oil applied every month at a concentration of 5 ml/l, by spraying the suspension onto all parts of the plant (250 ml/tree). Observation of attack symptoms, progression of the disease, the percentage and intensity of the attacks were carried out every month, whereas the level of effectiveness of botanical fungicides is calculated at the end of experiment. The results showed that clove oil, citronella and garlic extract can reduce the percentage and intensity of VSD disease attacks on cocoa plant. The highest percentage of the reduction of disease intensity were obtained in the use of clove oil and citronella at about 38.6% and 31.6% respectively, and both of them are potential to be used as botanical fungicide to control VSD disease.</p>
ABSTRAKPenyakit utama busuk buah kakao disebabkan oleh Phytophthora palmivora dapat menurunkan hasil 20%-30%. Pengendalian penyakit dengan fungisida nabati saat ini banyak dikembangkan, dengan tujuan mengurangi dampak negatif dari fungisida sintetik. Fungisida nabati yang digunakan adalah minyak cengkeh dan serai wangi karena mudah didapat dan bersifat fungisidal. Penelitian bertujuan menganalisis pengaruh formula fungisida nabati minyak cengkeh dan serai wangi terhadap perkembangan penyakit busuk buah kakao (BBK) yang disebabkan oleh Phytophthora palmivora. Penelitian dilaksanakan di kebun petani Kabupaten Mamuju, Sulawesi Barat, menggunakan rancangan acak kelompok 7 perlakuan, 4 ulangan. Setiap perlakuan diamati 20 buah kakao berukuran 8-10 cm. Perlakuan yang diuji adalah 1) minyak cengkeh+serai wangi, 2) minyak cengkeh+asam salisilat, 3) minyak cengkeh+silikon, 4) serai wangi+asam salisilat, 5) serai wangi+silikon, 6) fungisida sintetik sebagai pembanding, dan 7) kontrol. Larutan formula (5ml/liter) disemprotkan pada buah setiap 2 minggu sekali sampai buah masak atau dipanen. Parameter yang diamati adalah persentase serangan, intensitas serangan, kadar senyawa fenol, dan bobot biji kakao. Hasil penelitian menunjukkan bahwa formula fungisida nabati cengkeh dan serai wangi yang diperkaya dengan asam salisilat dan silikon dapat menekan intensitas serangan penyakit busuk buah kakao sebesar 20,48%-65,62%, tidak berbeda nyata dengan fungisida sintetik (73,15%). Besarnya tingkat penekanan penyakit sejalan dengan kandungan senyawa fenol pada kuit buah kakao. Semakin tinggi kadar fenol pada kulit buah, maka semakin tinggi penekanan penyakit busuk buah kakao. Penggunaan formula fungisida nabati dapat menekan kehilangan produksi kakao 23,94%-43,02%. Formula terbaik dan dapat dianjurkan untuk pengendalian penyakit busuk buah kakao adalah minyak cengkeh+serai wangi, cengkeh+asam salisilat, dan serai wangi+silikon.Kata Kunci: Kakao, Phytophthora palmivora, busuk buah kakao, fungisida nabati ABSTRACT Black pod disease caused by Phytophthora palmivora is a major disease on cacao crops, which can cause yield losses until 20%-30%. ABSTRAKTrichoderma spp. merupakan jamur bermanfaat yang dapat menghasilkan molekul bioaktif (metabolit sekunder) dengan kandungan antibiotik, enzim, hormon, dan toksin yang berperan penting dalam biokontrol penyakit tanaman. Tujuan penelitian adalah mengetahui potensi metabolit sekunder Trichoderma spp. untuk mengendalikan penyakit vascular streak dieback (VSD) pada bibit kakao. Penelitian dilaksanakan di Laboratorium Proteksi Tanaman, Balai Penelitian Tanaman Industri dan Penyegar (Balittri), dan kebun petani di Desa Balubus, Kabupaten Lima Puluh Kota, Sumatera Barat, mulai bulan April sampai Agustus 2016. Penelitian menggunakan rancangan acak lengkap (RAL) dengan 7 perlakuan dan 5 ulangan, masing-masing perlakuan menggunakan 5 tanaman. Perlakuannya adalah metabolit sekunder yang dihasilkan oleh T. virens LP1, T. hamatum LP2, T. amazonicum LP3, T. atroviride JB2, dan T. viride PRD, kontrol (tanpa perlakuan metabolit sekunder), dan fungisida kimia sebagai pembanding. Bibit kakao berumur 3 bulan diperlakukan dengan metabolit sekunder Trichoderma spp. dengan cara menyemprotkan suspensi metabolit ke seluruh permukaan daun. Aplikasi metabolit sekunder dilakukan setiap minggu sebanyak 6 kali. Inokulasi Ceratobacidium theobromae dilakukan secara alami dengan cara meletakkan bibit di bawah pohon kakao yang terserang VSD. Pengamatan dilakukan terhadap masa inkubasi dan intensitas serangan VSD serta pertumbuhan bibit kakao. Hasil penelitian menunjukkan metabolit sekunder Trichoderma spp. berpotensi untuk mengendalikan penyakit VSD pada bibit kakao. Metabolit sekunder yang paling potensial untuk mengendalikan VSD adalah T. amazonicum LP3 dan T. virens LP1 dengan penekanan intensitas serangan masing-masing sebesar 81,8% dan 63,2% atau lebih besar dan setara dengan fungisida kimia komersial (63,6%) serta dapat meningkatkan tinggi tanaman, jumlah daun, dan diameter batang bibit kakao.
