This paper reports the antifouling properties of the dichloromethane crude extract (DC) and 3 pure compounds isolated from the Brazilian brown seaweed Canistrocarpus cervicornis against establishment of the mussel Perna perna. DC extract showed a strong inhibition activity against byssal threads. Two natural dolastanes and one seco-dolastane diterpene, namely (4R, 9S, 14S)-4α-acetoxy-9β,14α-dihydroxydolast-1(15),7-diene (1), (4R,7R,14S)-4α,7α-diacetoxy-14-hydroxydolast-1(15),8-diene (2) and isolinearol (3), were isolated from DC extract. Dolastane (1) inhibited 60% of byssal fixation, while compound 2 and the seco-dolastane (3) strongly inhibited (82%) the establishment of P. perna. This is the first report of this type of chemical skeleton in three powerful compounds that could be further explored for the development of antifouling technology.

This paper has identified, for the first time in a member of the Rhodophyta, a vacuolar organelle containing enzymes that are involved in the mevalonate pathway-an important step in red algal isoprenoid biosynthesis. These organelles were named mevalonosomes (Mev) and were found in the cortical cells (CC) of Plocamium brasiliense, a marine macroalgae that synthesizes several halogenated monoterpenes. P. brasiliense specimens were submitted to a cytochemical analysis of the activity of the 3-hydroxy-3-methylglutaryl-CoA synthase (HMGS). Using transmission electron microscopy (TEM), we confirmed the presence of HMGS activity within the Mev. Because HMGS is necessary for the biosynthesis of halogenated monoterpenes, we isolated a hexanic fraction (HF) rich in halogenated monoterpenes from P. brasiliense that contained a pentachlorinated monoterpene as a major metabolite. Because terpenes are often related to chemical defense, the antifouling (AF) activity of pentachlorinated monoterpene was tested. We found that the settlement of the mussel Perna perna was reduced by HF treatment (2.25 times less than control; 40% and 90% of fouled surface, respectively; P = 0.001; F9,9 = 1.13). The HF (at 10 μg · mL(-1) ) also inhibited three species of fouling microalgae (Chlorarachnion reptans, Cylindrotheca cloisterium, and Exanthemachrysis gayraliae), while at a higher concentration (50 μg · mL(-1) ), it inhibited the bacteria Halomonas marina, Polaribacter irgensii, Pseudoalteromonas elyakovii, Shewanella putrefaciens, and Vibrio aestuarianus. The AF activity of P. brasiliense halogenated monoterpenes and the localization of HMGS activity inside Mev suggest that this cellular structure found in CC may play a role in thallus protection against biofouling.

Various benthic organisms have chemical defenses which reduce their predators' consumption. Although their efficiency may be noticed in many organisms, many of their effects are not well- known yet. Multiple ecological roles of secondary metabolites are shown in some sponges, which may represent an adaptative advantage considering the high amount of energy used to produce these chemical compounds. The goal of this work was to investigate the defensive property of the extracts from the sponge Hymeniacidon heliophila against the tropical predators: hermit crabs (Calcinus tibicens), sea urchins (Lytechinus variegatus) and generalist fishes. Extracts obtained with n-hexane, ethyl acetate and acetone/methanol were used in assays and all of them were effective in reducing the consumption by C. tibicens; n-hexane extract reduced the consumption by L. variegatus; and medium polarity extracts reduced fish consumption. Either the variation in action or the multiple ecological roles of the extracts indicates that different types of compounds can be associated to the defensive system produced by H. heliophila.
Diversos organismos bênticos possuem defesas químicas que ajudam a diminuir a predação. Embora sua eficácia seja comprovada, muitos de seus efeitos são ainda desconhecidos. Múltiplas funções dos metabolitos secundários foram evidenciadas em algumas esponjas e isso pode representar uma vantagem adaptativa, considerando o alto valor energético gasto pelas espécies para produzí-los. O objetivo desse trabalho foi investigar as propriedades defensivas de extratos da esponja Hymeniacidon heliophila contra predadores tropicais: paguros (Calcinus tibicens), ouriços-do-mar (Lytechinus variegatus) e peixes generalistas. Extratos em n-hexano, acetato de etila e acetona/metanol foram usados nos ensaios e todos foram eficientes na redução do consumo por C. tibicen; extratos em n-hexano reduziram o consumo por L. variegatus; e extratos de média polaridade reduziram o consumo por peixes. A variação na ação ou as funções ecológicas múltiplas dos extratos indicam que diferentes tipos de substâncias podem ser associadas ao sistema defensivo produzido por Hymeniacidon heliophila

Benthic marine organisms are constantly exposed to fouling, which is harmful to most host species. Thus, the production of secondary metabolites containing antifouling properties is an important ecological advantage for sessile organisms and may also provide leading compounds for the development of antifouling paints. High antifouling potential of sponges has been demonstrated in the Indian and Pacific oceans and in the Caribbean and Mediterranean seas. Brazilian sponges remain understudied concerning antifouling activities. Only two scientific articles reported this activity in sponges of Brazil. The objective of this study was to test crude extracts of twelve species of sponges from Brazil against the attachment of the mussel Perna perna through laboratorial assays, and highlight promising species for future studies. The species Petromica citrina, Amphimedon viridis, Desmapsamma anchorata, Chondrosia sp., Polymastia janeirensis, Tedania ignis, Aplysina fulva, Mycale angulosa, Hymeniacidon heliophila, Dysidea etheria, Tethya rubra, and Tethya maza were frozen and freeze-dried before extraction with acetone or dichloromethane. The crude extract of four species significantly inhibited the attachment of byssus: Tethya rubra (p = 0.0009), Tethya maza (p = 0.0039), Petromica citrina (p = 0.0277), and Hymeniacidon heliophila (p = 0.00003). These species, specially, should be the target of future studies to detail the substances involved in the ability antifouling well as to define its amplitude of action.Keywords: Porifera, Demospongiae, antifouling, Perna perna, Brazil. Assim, a produção de metabólitos secundários com propriedades anti-incrustantes é uma vantagem ecológica importante para organismos sésseis e pode também orientar o estudo de substâncias para o desenvolvimento de tintas anti-incrustantes. O alto potencial anti-incrustante de esponjas tem sido demonstrado nos oceanos Índico e Pacífico, nos mares Mediterrâneo e Caribenho. Esponjas brasileiras permanecem pouco estudadas em relação à atividade anti-incrustante. Apenas dois artigos científicos registraram essa atividade em esponjas do Brasil. O objetivo desse estudo foi testar os extratos brutos de doze espécies de esponjas do Brasil contra a fixação do molusco Perna perna através de ensaios laboratoriais e também destacar espécies promissoras para estudos futuros.
