Corynespora cassiicola is an Ascomycetes fungus with a broad host range and diverse life styles. Mostly known as a necrotrophic plant pathogen, it has also been associated with rare cases of human infection. In the rubber tree, this fungus causes the Corynespora leaf fall (CLF) disease, which increasingly affects natural rubber production in Asia and Africa. It has also been found as an endophyte in South American rubber plantations where no CLF outbreak has yet occurred. The C. cassiicola species is genetically highly diverse, but no clear relationship has been evidenced between phylogenetic lineage and pathogenicity. Cassiicolin, a small glycosylated secreted protein effector, is thought to be involved in the necrotrophic interaction with the rubber tree but some virulent C. cassiicola isolates do not have a cassiicolin gene. This study set out to identify other putative effectors involved in CLF. The genome of a highly virulent C. cassiicola isolate from the rubber tree (CCP) was sequenced and assembled. In silico prediction revealed 2870 putative effectors, comprising CAZymes, lipases, peptidases, secreted proteins and enzymes associated with secondary metabolism. Comparison with the genomes of 44 other fungal species, focusing on effector content, revealed a striking proximity with phylogenetically unrelated species (Colletotrichum acutatum, Colletotrichum gloesporioides, Fusarium oxysporum, nectria hematococca, and Botrosphaeria dothidea) sharing life style plasticity and broad host range. Candidate effectors involved in the compatible interaction with the rubber tree were identified by transcriptomic analysis. Differentially expressed genes included 92 putative effectors, among which cassiicolin and two other secreted singleton proteins. Finally, the genomes of 35 C. cassiicola isolates representing the genetic diversity of the species were sequenced and assembled, and putative effectors identified. At the intraspecific level, effector-based classification was found to be highly consistent with the phylogenomic trees. Identification of lineage-specific effectors is a key step toward understanding C. cassiicola virulence and host specialization mechanisms.
Corynespora cassiicola causa mancha alvo em folhas de diversas espécies de plantas, entre elas as cucurbitáceas. Este trabalho objetivou analisar a patogenicidade de 15 isolados de C. cassiicola originados de diferentes espécies hospedeiras a híbridos de pepino (Cucumis sativus) tipo 'japonês', assim como avaliar a severidade da doença em condições de casa-de-vegetação. Verificou-se que somente três isolados originados de pepino e um isolado de abóbora foram capazes de infectar os quatro híbridos de pepino. Os híbridos Natsubayashi e Tsuyataro também foram infectados pelos dois isolados originados de soja e um originado de tomateiro. Quanto à severidade, os isolados de C. cassiicola originados de pepino mostraram-se significativamente mais agressivos. Além disso, Tsuyataro mostrou-se mais suscetível dentre os híbridos testados. As diferenças na agressividade dos isolados de C. cassiicola sugerem maior adaptação dos isolados originados de pepino para os quatro híbridos, uma vez que estes foram significativamente mais agressivos do que os demais isolados.

Este trabalho teve como objetivos estudar o progresso da ferrugem-asiática da soja e quantificar os urediniósporos no ar na região de Campo Mourão no Estado do Paraná, região Sul do Brasil. Três experimentos foram instalados em condições de campo, nas safras 2006/07 e 2007/08. Utilizando-se o método do gradiente de doenças, proporcionado pela aplicação de doses crescentes do fungicida tebuconazole, foram obtidas quatro epidemias em cada experimento com diferentes intensidades de ferrugem asiática. Para quantificar os uredinióporos, foram instalados coletores de esporos tipo catavento durante as duas safras e entressafras. As curvas foram ajustadas ao modelo logístico por apresentarem maiores coeficientes de determinação e melhor aleatoriedade dos resíduos plotados no tempo. As análises da área abaixo da curva de progresso da doença mostraram que as maiores epidemias ocorreram na safra 2006/07 e as taxas de progresso foram maiores para a severidade, mesmo em plantas protegidas com o fungicida. A quantidade de urediniósporos coletados no ar foi relacionada com a presença de plantas de soja em lavouras cultivadas. A quantidade de urediniósporos também apresentou correlação positiva com a severidade, com a incidência da doença, com a precipitação acumulada e com os dias favoráveis a infecção por P. pachyrhizi. Climate is quite important among the factors that favor the epidemiology of ASR. Climate variations are influenced by macro-, meso-and microclimate. They can affect the different processes of the disease cycle and influence the rate of progress and severity of the epidemics. In soybean fields, rainfall seems to be the most important factor that influences the severity of the disease on a regional scale. In v.38, n.4, p.280-287, 2012. curves were plotted for each epidemic, and maximum severity was estimated. The curves were fit to the logistic model, which provided higher coefficients of determination and more randomly distributed residuals plotted over time. Analyses of the area under the disease progress curve showed that the largest epidemics occurred in the 2007/2008 crop season and that the progress rates were higher for severity, even among plants protected with the fungicide. The number of urediniospores collected in the air was related to the presence of soybean plants in the cultivated crops. ABSTRACT. The powdery mildew caused by Oidium spp. is an important disease for several crops of the Cucurbitaceae family. Although the teleomorphs, Podosphaera xanthii and Golovinomyces cichoracearum, currently have already been described as the causal agents of powdery mildew in Brazil, only P. xanthii is considered the main causal agent of powdery mildew field epidemics. The objective of this work was to identify and determine the prevalence of the species causing powdery mildew in cucumber (Cucumis sativus) and melon (Cucumis melo var. reticulatus) grown in greenhouses in the State of Paraná in Brazil. The morphological traits of the conidial stages, such as the presence of fibrosin bodies and a germinative tube, were used to identify the species. Leaves exhibiting high severity of powdery mildew were collected from plants of 13 plastic greenhouses during different seasons in 2003/2004 and in different regions of Paraná State. In all environments, a significant prevalence of P. xanthii (80-100%) was observed affecting parthenocarpic or ordinary cucumber and melon. Golovinomyces cichoracearum was observed in six greenhouses, with up to 20% of conidia of this species on the samples.Keywords: morphological characterization, grafting, Podosphaera xanthii, Golovinomyces cichoracearum.Espécies de fungos que causam oídio em casas de vegetação cultivadas com pepino e melão no Estado do Paraná, Brasil RESUMO. O oidio, causado por Oidium sp. é uma importante doença para espécies de plantas cultivadas da família das cucurbitáceas. Apesar das espécies teleomórficas Podosphaera xanthii e Golovinomyces cichoracearum já terem sido citadas como causadoras de oídio no Brasil, geralmente em trabalhos publicados atualmente tem-se referenciado somente a P. xanthii como agente causal dessa doença em cucurbitáceas em cultivo convencional. Por isso, este trabalho teve como objetivo identificar e quantificar a freqüência de ocorrência dessas duas espécies causadoras de oídio nas culturas de pepino (Cucumis sativus) e melão nobre (Cucumis melo var. reticulatus) conduzidas em estufas plásticas no Estado do Paraná. Para a identificação de P. xanthii e G. cichoracearum utilizaram-se as características morfológicas dos conídios (fase anamórfica), presença de corpos de fibrosina no interior dos conídios e características do tubo germinativo dos conídios. A amostragem foi realizada coletando-se folhas das plantas apresentando alta severidade de oidio em 13 estufas plásticas em 2003/2004 em diferentes regiões do estado do Paraná. Em todas as 13 estufas plásticas ocorreu à presença de P. xanthii em alta prevalência (80-100%). Entretanto, a presença de G. cichoracearum foi observada em seis estufas plásticas, apresentando freqüência de conídios característicos da espécie de até 20% nas amostras avaliadas.
