Mutations have been identified in the beta2-microglobulin gene of tumor cells of two metastatic melanoma patients who received immunizations with MAGE peptides. One mutation abolishes the start codon whereas the other introduces a premature stop codon. The second beta2-microglobulin allele of both tumors appears to be lost on the basis of sequence data and loss of microsatellite heterozygosity. The lack of beta2-microglobulin gene product results in the absence of HLA class I antigens on the surface of the tumor cells. This may explain why the tumors of both patients progressed despite the immunization treatment and shows the necessity of analyzing in depth the antigen presentation capability of the tumor cells for the interpretation of clinical trials involving anti-tumor vaccination.
Beta2 microglobulin mutations are an important mechanism for HLA class I total loss, (phenotype No. I) and have been described in colon carcinomas, melanomas and lymphomas. We describe a new beta2 microglobulin mutation detected in the melanoma cell line GR-34. The new mutation reported here was identified as a deletion of 4 bases (TTCT) in the highly repetitive sequence CTCTCTCTTTCT located in the leader sequence of the beta2 microglobulin gene at codon 15-16 of exon 1. The mutation produces a frameshift in the open reading frame sequence with the appearance of a stop codon at position 42. We also demonstrate that the second beta2 microglobulin gene is deleted. Comparisons with beta2 microglobulin mutations in other tumor cell lines suggest a mutation hot spot in exon 1.
Las saponinas son un tipo de metabolito secundario ampliamente estudiado por susreconocidas propiedades biológicas. Gran parte de las investigaciones en fitoquímicaestán dirigidas a encontrar nuevas fuentes naturales de saponinas con aplicaciónmedicinal. La quinua (Chenopodium quinoa Willd.) es una planta que ha alcanzadoun valioso reconocimiento por ser una fuente de alimentos altamente nutritivos, asícomo una especie rica en saponinas triterpénicas contenidas, principalmente, en lacáscara de las semillas. A la fecha, se han identificado alrededor de 30 saponinas derivadasde la hederagenina y de los ácidos oleanólico, fitolacagénico y serjanico en laplanta. El consumo del grano de quinua implica la remoción de la cáscara a fin dereducir su sabor amargo, la ingesta de niveles residuales de saponinas y la obtenciónde un subproducto rico en las mismas. Esta revisión, inicialmente, ofrece una contextualizacióngeneral de las saponinas; posteriormente, recopila las característicasestructurales de las saponinas identificadas en la quinua, describe el efecto del procesamientodel grano en su contenido de saponinas y, finalmente, expone los efectosbiológicos explorados con extractos de saponinas de quinua, los cuales pueden serconsiderados como punto de partida en investigaciones futuras dirigidas al fortalecimientode su uso en el campo farmacéutico y/o nutracéutico.
Los sistemas de producción industrial actuales usan metales pesados para la extracción de un material o como elemento en la refinación de un producto en particular lo que en principio resulta en la obtención del producto deseado con relativos bajos costos de producción. Sin embargo, usar estos metales presenta un grave problema a nivel ambiental debido a sus altos niveles de toxicidad para los organismos con los que interactúen una vez son descargados. Por tanto, se han desarrollado diversos métodos para tratar con estos metales una vez están en el agua y los métodos para analizarlos. Se realizó una búsqueda amplia y actualizada de las fuentes, efectos y toxicidad en los seres vivos, métodos de análisis y las diversas técnicas de tratamiento existentes para metales pesados. Como resultado el investigador estará más preparado al tener información actual sobre los tratamientos a tener en cuenta en la manipulación de metales pesados.
