Fermentation microorganisms, lactic acid bacteria (LAB) and yeast from 12 samples of tunta production chain were quantified, from the native potatoes used by the process fermentation of potatoes in the river up to the final product. During fermentation, the LAB population steadily increased from 3 to 4 to 8 log CFU/g during the first 8 days in the river and the yeast population increased from 2 to 3 to 3-4 log CFU/g. Overall, 115 LAB strains were isolated using a culture-dependent method. Molecular techniques and 16S rRNA gene sequencing enabled the identification of native species. In LAB isolates, members of the Lactobacillaceae (64%), Leuconostocaceae (9%) and Enterococcaceae (2%) families were identified. The most prevalent LAB species in the tunta production chain was Lactobacillus curvatus, followed by Leuconostoc mesenteroides and Lactobacillus sakei, Lactobacillus brevis and Enterococcus mundtii were also present. Only 13 LAB strains showed anti-listerial activity, and one of them, identified as En. mundtii DSM 4838 [MG031213], produced antimicrobial compounds that were determined to be proteins after treatment with proteolytic enzymes. Based on these results, we suggest that traditional fermented product-derived LAB strains from specific environments could be selected and used for technological application to control pathogenic bacteria and naturally protect food from post-harvest deleterious microbiota.
El objetivo de este estudio es caracterizar molecularmente bacterias con potencial probiótico aisladas de heces de neonatos humanos. Se evaluó 60 muestras de heces de neonatos (0-3 días) se enriquecieron en caldo Man Rogosa y Sharp (MRS) a 37°C/24h. Se seleccionó y se sometió a pruebas in vitro con sales biliares, resistencia a pH bajo y actividad antimicrobiana frente a Escherichia coli ATCC25922, E. coli ATCC35218, Salmonella enterica y Listeria inocua mediante el ensayo difusión en agar. La identificación molecular se realizó con amplificaciones PCR-BOX y el secuenciamiento del gen 16S rRNA. Se aislaron un total de 48 cepas y todas presentaron resistencia a pH 3 y 0.3% sales biliares; 3 cepas mostraron actividad antimicrobiana frente a E. coli ATCC25922, 1 cepa frente a E. coli ATCC35218, 5 cepas frente a L. inocua y todas frente a S. entérica. De las 48 cepas se obtuvieron dos perfiles BOX-PCR pertenecientes a los géneros de Lactobacillus y Enterococcus. Nueve cepas (C52, C61, C71, C112, C16 2, C192, C20, C35, y C42) presentaron un 100% de similaridad a L. plantarum ATCC 14917T [ACGZ01000098] y dos cepas (C15 y C40) un 99.93% y 99.80% de similaridad, respectivamente a Enterococcus faecium CGMCC 1.2136T [AJKH01000109]; estas cepas mostraron actividad en leche con diferencias significativas (p valor < 0.05) en la cinética de pH 3. En conclusión se encontró bacterias con potencial probiótico.
ResumoO carcinoma de células escamosas de laringe (CCEL) é um tumor maligno de mal prognóstico. O polimorfismo MCC c.*5077A>G (rs7033) está envolvido com o risco para outros carcinomas de cabeça e pescoço. Já é conhecido que o genótipo MCC AA deste polimorfismo está associado a risco aumentado de CCEL, especialmente se analisado junto à variável cor de pele. O objetivo deste estudo foi avaliar se o polimorfismo citado altera também o prognóstico de pacientes com CCEL. A sobrevida global dos pacientes foi considerada como indicador de prognóstico. Não há resultados preliminares, no entanto, esperamos que um resultado positivo reforce a necessidade de tratamento diferenciado para genótipos mais agressivos.
