Makorin ring finger proteins (MKRNs) are part the of ubiquitin-proteasome system; a complex system important for cell functions. Ubiquitin fate through proteolytic, non-proteolytic pathways varies, depending on covalent linkage between ubiquitin and protein substrates. Makorin ring finger protein 3 is an integral part of covalent linkage of ubiquitin to protein substrates. Similar to others imprinted genes, MKRN3 also evolve under positive selection; however, which codons are specifically selected in MKRN3 during evolution are needed to be explored. Different maximum-likelihood (ML) codon-based methodologies were used to ascertain positive selection signatures in 22 mammalian sequences of MKRN3 to probe an individual codon for positive selection signatures. By applying the HyPhy software package implemented in the Data Monkey Web Server and CODEML implemented in PAML, evolutionary analysis based on two Ml frameworks were conducted. The analysis was executed by comparing M1a against M2a, M7 against M8, and PAML models and 2∆Lnl ( LRT) was resulted by likelihood logs. M1a contributed ω1 ( dN/dS) with LRT value ( ∆Lnl) 12.01, and positive selection was found in M2a with ω3 = 2.23603. To further improve selection test, M8 was compared to M7 with 2∆ Lnl ( LRT) 30.17, and M8 showed positive selection with ω = 1.55759. The data were fit to M8 than M7, which suggests that M8 was the most significant model of selection. M8 was judged encouraging for this analysis and used to establish a positive selection of MKRN3 proteins. We found Gly312 as a positively selected amino acid in a zinc finger motif/Really Interesting New Gene (RING) finger motif; the former ones’ region is involved in RNA binding and the later ones in ubiquitin ligase activity of the protein, vital for protein function. Selection analyses of MKRNs might advance the developments in unique approaches that could lead to genetic progress over the selection of superior individuals with the breeding values higher for certain traits as ancestries to get the next generation.

| Genetic correlations between any two traits indicate that some of the same genes are controlling them and in case of positive genetic correlation, selection for improvement in one trait will result in improvement in the other trait as a correlated response. In case of absence of any milk recording system under field conditions, farmers generally select animals on the basis of phenotype. Information on genetic correlation of body measurements with milk yield can be used for indirect selection for improved milk yield. Objective of this study was to record different body measurements in Nili Ravi buffaloes and estimation of phenotypic and genetic correlations with milk yield. These correlations were estimated in 437 Nili Ravi buffaloes maintained at 5 Livestock Experiment Stations in Punjab and few private breeders. Data included 1180 records of milking buffaloes. Correlations were estimated using BLUP techniques. Age of the buffalo at scoring, stage of lactation, parity, herd and season of scoring were included in the model. Data were analysed using the mixed model procedure of the SAS. Fixed effects observed to be significant in the initial analysis were included in the model for estimation of genetic and phenotypic correlation of each trait with milk yield using bivariate analysis in ASREML computer program. Buffaloes in the data were the progeny of 88 sires and 303 dams with 119 base animals. The least squares means for height at withers, diagonal body length and heart girth were found as 132.04±4.56, 154.01±7.60 and 194.46±10.31cm, respectively. Phenotypic correlations of 305 days milk yield with body measurements were low. Positive genetic correlations were observed as 0.16±0.0003 for diagonal body length, 0.08±0.00 for height at withers and 0.14±0.0005 for heart girth. Diagonal body length has shown a low but positive genetic correlation with milk yield and this trait might be considered in the selection program for Nili Ravi buffaloes. Genetic correlation of heart girth with milk yield can be considered for indirect selection for milk yield.

This trial was undertaken to study the effect of breeding season on haematological and blood biochemical parameters of Barela male camel under extensive conditions in Desert Thal Pakistan. The clinically healthy male Barela camels (n=5) from 6-10 years old having mean bodyweight of 680±50 kg was studied in breeding and Non-breeding season. The camels were fed with gram-straws, natural vegetation and twice day water. Deworming was performed before the start of trial. Haematological and blood biochemical parameters such as haemoglobin (Hb), red-blood-cells (RBC) and white-blood-cells (WBC) count, packed-cell-volume (PCV), glucose, cholesterol, triglycerides, total protein, urea and creatinine concentrations were analyzed on haematology and biochemistry analyzer. The mean Hb concentration (P<0.05) was found to be 14.77±0.76 and 14.16±0.97 g/dl in non-breeding and breeding males, respectively. The hematological values of RBC, WBC and PCV were found to be varied (P<0.05) as higher in non-breeding animals, while the biochemical indices including cholesterol, triglycerides and total protein were also significantly (P<0.05) higher in nonbreeding males except glucose concentration which was found higher in rutting males. Whereas the urea and creatinine non-significantly (P>0.05) varied between non-breeding and breeding males. These results were compared with available relevant literature and it showed that these parameters may be affected due to breeding stress in the male camels which could be the indication of high performance in the rutting season.
