Artificial oligo(ethylene amino) acids, together with selected natural amino acids and fatty acid modifications, have been used for the solid‐phase‐assisted synthesis of polymers with precise sequence, topology, and modifications (see picture). [2,3] Hartmann, Bçrner und Kollegen verçffentlichten eine Syntheseroute zu Polyamidoaminen, die auf der abwechselnden Kupplung von Bernsteinsäureanhydrid und Diamino-Nmethyldipropylamin bzw. geschützter Spermin-Derivate beruht.[4] Um die Vorteile der Peptidsynthese mit der breiten chemischen Diversität synthetischer Polymere zu kombinieren, entwickelten wir künstliche Fmoc/Boc-geschützte Aminosäuren, die definierte Diaminoethan-Einheiten enthalten.[5] Dieses Motiv verfügt als "Protonenschwamm" über die Fähigkeit zur Endosomen-Destabilisierung, welche für die Transfektionseffizienz von Polyethylenimin (PEI) [6, 7] und weiteren kationischen Polymeren entscheidend ist.[8] Die biologische Aktivität ist dadurch viel hçher als bei Diaminopropan-Einheiten, die bereits bei neutralem pH vollständig protoniert sind.
