Latar belakang: mahasiswa profesi kedokteran gigi merupakan salah satu petugas kesehatan yang harus memenuhi kriteria standar dalam memberikan pelayanan kesehatan gigi dan mulut, antara lain melakukan pencegahan dan pengendalian infeksi (PPI). Penerapan PPI seperti cuci tangan menurut WHO dan penggunaan APD merupakan hal yang penting untuk mencegah penyebaran infeksi silang dan Health-care Associated Infection (HAIs). Tujuan: untuk mengetahui gambaran pengetahuan penerapan cuci tangan dan alat pelindung diri pada mahasiswa Fakultas Kedokteran Gigi DKI Jakarta. Metode: penelitian ini menggunakan metode observasional deskriptif dengan metode cross sectional. Pengumpulan data menggunakan kuesioner melalui google form yang berisi 14 pertanyaan mengenai pengetahuan penerapan cuci tangan menurut WHO dan 15 pertanyaan mengenai penggunaan APD dengan 197 responden dari mahasiswa program profesi angkatan 2016 dan 2017 FKG DKI Jakarta. Hasil: tingkat pengetahuan penerapan cuci tangan menurut WHO dengan kategori baik sebanyak 44,7%, kategori cukup 53,8%, dan kategori kurang 1,5%. Tingkat pengetahuan penggunaan APD dengan kategori baik sebanyak 62,4%, kategori cukup 36,5% dan kategori kurang 1%. Kesimpulan: tingkat pengetahuan responden terbanyak untuk penerapan cuci tangan menurut WHO adalah kategori cukup, yaitu sebesar 53,8%. Tingkat pengetahuan responden terbanyak untuk penerapan alat pelindung diri adalah kategori baik, yaitu sebesar 62,4%.
