Increased salt tolerance is needed for crops grown in areas at risk of salinisation. This requires new genetic sources of salt tolerance, and more efficient techniques for identifying salt-tolerant germplasm, so that new genes for tolerance can be introduced into crop cultivars. Screening a large number of genotypes for salt tolerance is not easy. Salt tolerance is achieved through the control of salt movement into and through the plant, and salt-specific effects on growth are seen only after long periods of time. Early effects on growth and metabolism are likely due to osmotic effects of the salt, that is to the salt in the soil solution. To avoid the necessity of growing plants for long periods of time to measure biomass or yield, practical selection techniques can be based on physiological traits. We illustrate this with current work on durum wheat, on selection for the trait of sodium exclusion. We have explored a wide range of genetic diversity, identified a new source of sodium exclusion, confirmed that the trait has a high heritability, checked for possible penalties associated with the trait, and are currently developing molecular markers. This illustrates the potential for marker-assisted selection based on sound physiological principles in producing salt-tolerant crop cultivars. The problem
The hypothesis that ABA produced by roots in drying soil is responsible for stomatal closure was tested with grafted plants constructed from the ABA-deficient tomato mutants, sitiens and flacca and their near-isogenic wild-type parent. Three types of experiments were conducted. In the first type, reciprocal grafts were made between the wild type and sitiens or flacca. Stomatal conductance accorded with the genotype of the shoot, not the root. Stomates closed in all of the grafted plants in response to soil drying, regardless of the root genotype, i.e. regardless of the ability of the roots to produce ABA. In the second type of experiment, wild-type shoots were grafted onto a split-root system consisting of one wild-type root grafted to one mutant (flacca or sitiens) root. Water was withheld from one root system, while the other was watered well so that the shoots did not experience any decline in water potential or loss of turgor. Stomates closed to a similar extent when water was withheld from the mutant roots or the wild-type roots. In the third type of experiment, grafted plants with wild-type shoots and either wild-type or sitiens roots were established in pots that could be placed inside a pressure chamber, and the pressure increased as the soil dried so that the shoots remained fully turgid throughout. Stomates closed as the soil dried, regardless of whether the roots were wild type or sitiens. These experiments demonstrate that stomatal closure in response to soil drying can occur in the absence of leaf water deficit, and does not require ABA production by roots. A chemical signal from roots leading to a change in apoplastic ABA levels in leaves may be responsible for the stomatal closure.
A significant part of the world's land area is salt-affected. Most crops are not able to handle large amounts of salt without a negative impact on their growth and metabolism, so understanding the physiological mechanisms by which plants deal with salinity is essential to minimize detrimental effects and reduce the multibillion dollar penalties imposed by salt stress on crop production. This chapter summarizes our current knowledge of physiological mechanisms conferring plant adaptive responses to salinity. The classification of saline soils is given, with causes of primary and secondary types of salinity. Major physiological constraints are then summarized, and physiological and genetic diversity of plant responses to salinity are discussed. Key physiological and anatomical mechanisms conferring salinity tolerance in plants are then analysed in detail, and practical ways of ameliorating salinity stress are considered. Altogether, this chapter shows that plants have evolved numerous mechanisms to prevent accumulation of toxic Na+ levels in cell cytosol. This ability is complemented by mechanisms enabling efficient osmotic adjustment and maintenance of the cell turgor, as well as mechanisms of coping with oxidative stress imposed by salinity. A more complete physiological and genetic understanding of these processes will enable breeders to produce new salt-tolerant plants for the future.
