Caanine monocytic ehrlichiosis caused by Ehrlichia canis has gained wider significance owing to its potential to inflict significant deleterious effect on the health of companion animals. In the present study, 46 confirmed ehrlichiosis positive dogs were evaluated for the alterations in clinical, haematological and biochemical attributes. Depression, anorexia, pyrexia, anaemia, weakness, jaundice, melana, vomition and diarrhoea were the main clinical symptoms onserved. Haematological alterations included pancytopenia especially thrombocytopenia. Significant changes were noticed in WBC, RBC, Hgb, McHc, Platelets, ALT values while rest all the studied haematological and biochemical parameters showed non-significant alterations within normal range in comparison to normal healthy controls. The findings substantiate that ehrlichiosis cause significant clinical, haematological and biochemical alterations of the varied intensity in dogs, even with lower grades of parasitaemia.
The haemato-biochemical indices and oxidative stress markers in horses naturally infected with Trypanosoma evansi were evaluated by analyzing the level of these parameters between T. evansi infected (microscopically positive patent group and PCR positive latent group) and infection free horses. To compare the hemato-biochemical indices and oxidative stress indicators, horses were divided into three categories based on diagnostic test employed and positive results obtained. These included Romanowsky stained slide positive group (Group I; n = 6), PCR positive group (group II; n = 28) and negative control group (group III, n = 30), revealing parasitologically positive patent, molecular positive latent and disease free status of horses. A significant reductions in total erythrocytes count (TEC, P = 0.01), haemoglobin (Hb, P = 0.01) and packed cell volume (PCV, P = 0.04) was noticed both in group I and group II while significant neutrophilia and lymphocytopenia was observed in group I when compared to negative control group. Substantial increase in creatinine (CRTN, P = 0.032) and gamma glutamyl transferase (GGT, P = 0.012) in group I while significant decrease in glucose (GLU, P = 0.04) and iron (Fe, P = 0.01) were noticed in both group I and group II in comparison to group III. A significant difference in lipid peroxides (LPO, P = 0.01) with highest level in patent group I (15.33 ± 0.53) followed by PCR positive latent group (14.09 ± 1.66) indicates higher lipid peroxidation in erythrocytes and oxidative stress in decreasing order when compared with infection free control horses (9.83 ± 0.97). Catalase (CAT, P = 0.01) was significantly lower in parasitological (0.82 ± 0.14) and molecular positive cases (1.27 ± 0.35) in comparison to control group (3.43 ± 0.96). The levels of superoxide dismutase (SOD, P = 0.01), reduced glutathione (GSH, P = 0.01) and ferric reducing antioxidant power (FRAP, P = 0.01) were significantly lower in parasito-molecular positive cases as compared to infection free control horses. An inverse correlation of RBC count with LPO and GSH and a direct correlation with catalase, SOD and FRAP was revealed. Overall, the observed substantial decreases in the oxidative parameters like catalase CAT, SOD, GSH and FRAP activities with remarkably elevated levels of LPO indicate high exposure of erythrocytes to oxidative damage in T.evansi infected horses.
Trypanosomosis and bovine tropical theileriosis are important vector-borne protozoan diseases imposing some of the serious constraints on the health and productivity of domestic cattle in tropical and subtropical regions of the world. Following recovery from primary infection of both these conditions, animals become persistent carriers and act as reservoirs of infection thereby playing a critical role in disease epidemiology. The present study describes development and evaluation of duplex polymerase chain reaction (PCR) assays for simultaneous detection of Trypanosoma evansi and Theileria annulata in buffaloes. Following in silico screening for candidate target genes representing each of the pathogens, an optimized duplex PCR assay was established using TBR F/R and TAMS F/R as primer sets encoding for products of 164 and 721 bp for T. evansi and T. annulata, respectively. The results were compared and correlated with conventional Giemsa-stained thin blood smear examination and the single PCR assay. The duplex PCR detected each pathogen with the same level of sensitivity, irrespective of whether its DNA was amplified in isolation or together with DNA of another pathogen. Moreover, single and duplex PCRs were able to detect each species with equal sensitivity in serially diluted DNA representing mixtures of T. evansi and T. annulata, and no evidence of nonspecific amplification from nontarget species was observed. The developed assay may be seen as a good tool for epidemiological studies aiming at assessing the burden of dual infections and improving control of the associated diseases in endemic regions.
The rapid development of anthelminthic resistance has limited the success of traditional control programmes in several countries, thereby forcing the researchers to search for alternatives. In vitro anthelmintic activities of crude aqueous and hydro-alcoholic extracts of the leaves of Eucalyptus globulus were investigated against the egg and larvae of naturally infected sheep. In the phytochemical analyses, tannins and flavonoids were the main metabolites identified in the extract. The aqueous extract of E. globulus was also investigated for in vivo anthelmintic activity in naturally infected sheep. The various blood parameters, coupled with effects on marker enzymes and antioxidant status, were evaluated during the trial period. Methanolic extract showed better ED50 (3.756 mg/ml) and ED99 (33.809 mg/ml) values in comparison with aqueous extract (ED50 = 1.502 and ED99 = 7.10 mg/ml) in the egg hatch assay. Inverse was true in larval development and larval paralysis tests. The aqueous extract's ED50 = 19.994 and ED99 = 108.931 mg/ml values in the larval development test and ED50 = 19.994 and ED99 = 108.931 mg/ml in the larval paralysis test were more potent than those of its methanolic counterpart with ED50 = 15.595 and ED99 = 94.493 mg/ml and ED50 = 15.595 and ED99 = 94.493 mg/ml, respectively. A significant amount of 66% faecal egg count reduction was observed in in vivo trail using the aqueous extract on day 21 post treatment, although in initial stages it showed 58.0 and 80% effectiveness on days 7 and 14 post treatment. Though the FCER reduction was somewhat lower in terms of comparison with albendazole, nevertheless, significant and prolong reduction was noticed. No deleterious ill effect was found in any of the haematological and biochemical parameters suggesting that the plant could be safer for use in sheep. Though significant changes were observed in SGPT, RBCs, Hb and RDWc levels, other parameters showed nonsignificant variations within the normal range in the stipulated time of of herbal trial period. Based on the results of the present study, it could be very well concluded that leaves of E. globulus possess good level of anthelminthic efficacy; further research is thereby warranted before recommending it for use in nematode control programme in ovines.
Trypanosomosis is caused by different species of unicellular eukaryotic haemofl agellate Trypanosoma. Though human infection by animal species of trypanosomes is "not possible" as these species fails to infect humans due to innate immunity of the host due to presence of trypanolytic factor in human serum, however, across the world 20 patients with atypical human trypanosomosis are documented, eight of which are confi rmed between 1974 and 2014 due to improved molecular diagnostic assays. However, the numbers of cases are atypical human trypanosomosis caused by animal trypanosomes might be underestimated. Ten atypical cases of human trypanosomosis have so far been reported form Indian subcontinent. Out of these, nine cases were from India and one from Sri Lanka. Three cases of human T. evansi have been reported from the Indian subcontinent (one from Sri Lanka and two from India) during the last decade. Apart from these three cases, at least six more atypical human cases of trypanosomosis caused by rat trypanosome, T. lewisi have been reported. Two casualties due to non-tsetse transmitted trypanosomosis (NTTT) were also reported from India. High prevalence of these two animal trypanosomes in India is now a matter of concern for public health specialists. These raise it as an alarming situation of emerging new zoonotic disease and debatable one health issue to public health experts and parasitologist. There is a need to investigate disease with modern diagnostics by epidemiological based surveys in the fi eld to know the exact situation of the disease. Veterinarian can play very crucial role in diagnosis and control of disease, also by educate and re-educate people about the transmission, prevention and control of disease.
