In Versuchen an Tieren und Menschen haben wir beobachtet, daß g r o ß e Salizylatdosen (3,4) als eine a 11 g em e in e S c h ä d j g u n g die unspezifischen Schutzvorrichtungen des Organismus mobilisieren und daß somit ein Faktor der Salizylwirkung die Mobilisierung ,,antiarthritischer" Nebennierenhormone darstellt, während kleine Salizylatdosen einen solchen alarmierenden Effekt nicht ausüben können2. Der Organismus setzt sich gegen die Schädigung zur Wehr, wobei unter seinen homöostatischen Mechanismen die Mobilisierung des Cortison-Anteils der Rindensteroide eine wichtige Rolle spielt3. Auf die Bedeutung des in den Forschungen der letzten Jahre vielleicht etwas in den Hintergrund gedrängten A d r e n a 1,i n s kommen wir später zu sprechen. Eigene UntersuchungenTierversuche: Wir haben an Ratten die Zahl der zirkulierenden Eosinophilen und Lymphozyten, die morphologischen und histoloVor kurzem hat L e c h 1 e it n e r (1) in dieser Zeitschrift mitgeteilt, daß unter den therapeutischen Salizylatwirkungen der Einfluß auf die Leber an erster Stelle steht. In Anbetracht der gegenwärtigen Renaissance der Frage nach dem Salizylatwirkungsmechanismus möchte ich zu diesem Thema mit einigen Bemerkungen und experimentellen Daten beitragen. Bereits A r c h e r (1949) schrieb auf Grund klinischer und biochemischer Beobachtung der Remissionen unspezifischer Arthritiden, daß der gemeinsame Faktor der Remissionen die Schädigung oder Dysfunktion der Leber sein dürfte (2), Zweifellos lösen große Salizylat-Dosen eine ganze Reihe von Veränderungen a u ch in d e r L e b e r aus, doch könnte ähnliches -besonders nach Verabreichung großer Dosen eines Mittels -von zahlreichen Pharmaka gesagt werden. Natürlich gilt dies auch für die im folgenden voiX uns angeführten Nebennierenwirkungen. Die Salizylatschädigung berührt den ganzen Organismus.2 H e r z und S t a mp f1 (1951) konnten bei einer täglichen Salizylsäuregabe von 0,1 mg/g und einer über zwei Monate währenden Versu.chsdauer an Mäusen und Kaninchen keine eindeutigen Veränderun-gen in den inneren Organen feststellen, während solche bei täglich 0,3 mg/g schon nach kürzerer Zeit deutlich in Erscheinung traten (5). Dasselbe war der Fall in unseren (unveröffentlichten) Experimenten, in welchen wir Ratten zwei Monate lang 0,3 mg/g Na-Salicylicum verabreichten.Neuerdings haben S a y e r s (6) und I n g 1 e (7) die Auffassung vertreten, daß die Antwort des hypophyseo-adrenalen Systems nicht der Initiator, sondern der Unterstützer der vielseitigen Stoffwechselveränderungen des ,,Stress" zu sein scheint. THERAPIE nischen Zeichen der S e 1 y e schen Alarmreaktion, die Höhe des Ascorbinsäure-und Lipoidgehaltes der Nebennierenrinde, den akuten Pormalin-Test von S e 1 y e , sowie den von uns angegebenen Hyaluronidase-Fuflödem-Test (4, 8) untersucht. Große Salizylatdosen -500 mg/kg mit einem Serumsalizyispiegel von 35 mg°/o und mehrhaben eine bedeutende Cortison-Wirkung produziert. Ihr Einfluß auf die Wundheilung -an Mäusen -ist aber sehr gering (9). Kleine Dosen -mit einem Spiegel von 20 mg°...
