Bei vielen Insekten wird das Zusammenfinden der Geschlechtspartner durch artspezifische Lockdüfte ermöglicht 1 " 5 . Biologische Untersuchungen über Sexuallockstoffe sind vor allem an Nachtschmetterlingen unternommen worden, und Versuche mit markierten Tieren haben gezeigt, daß die Männchen unbefruchtete Weibchen bei manchen Species über große Entfernungen hin aufzufinden vermögen. Dieses Phänomen wird im Schrifttum bis in die neuere Zeit hinein 6 mit physikalischen Fernwirkungen wie X-Strahlen, Infrarot-Strahlen oder anderen elektromagnetischen Schwingungen erklärt. Durch Extraktions-und Kondensationsversuche ist jedoch überzeugend gezeigt worden, daß ein stoffliches Prinzip vorliegen muß, das von den weiblichen Schmetterlingen aus Duftorganen der letzten Hinterleibsegmente sezerniert und von den Männchen mit ihren Antennen wahrgenommen wird 1 " 5 . Da die chemische Natur der Sexuallockstoffe unbekannt war, schien es reizvoll, die Isolierung und chemische Charakterisierung dieser hochaktiven Wirkstoffe an einem geeigneten Objekt zu versuchen. Wir wähl-ten auf Anregung von Walter Seh o eller und seinen Mitarbeitern 3 ' 4 für unsere Untersuchungen den leicht zu haltenden und in größeren Mengen zugänglichen Seidenspinner Bombyx mori L.Der Verhaltenstest am Seidenspinner-Männchen Als Test auf die Anwesenheit des Sexuallockstoffes dient seine erregende Wirkung auf Seidenspinner-Männchen 1 ' 3 ' 4 . Einzeln gehaltene Männchen bleiben oft stundenlang unbeweglich sitzen. Sie werden aber durch ein in ihre Nähe gebrachtes Weibchen, durch deren Duftdrüsen (Sacculi laterales) oder Extrakte daraus fast augenblicklich in einen charakteristischen Erregungszustand versetzt, der sich in heftigem 1
Twenty-four hours after oral administration of metformin-14C to mice (100 mg/kg), an average of 77.2% of the administered activity was recovered in the urine and 8.4% in the faeces. Under the same test conditions the rat eliminated 66.8% of the administered dose in the urine and 18.4% with the faeces. Less than 1% of the radioactivity was found in the bile. Unchanged metformin was the only radioactive substance which appeared in the urine of rats and mice. The investigation comparing the distributions of buformin-14C and of metformin-14C in the body organs gave a similar picture for both biguanides. The highest biguanide concentrations were found in the kidneys, adrenals, pancreas and liver. Serum concentrations amounted to an average of 1.2 ~g/ml after oral administration of 50 mg buformin-14C/kg and 6.2 ~g/ml after 150 mg metformin-~aC/kg. Healthy human volunteers eliminated an average of 63% of the administered dose in the urine within 36 h. Metformin was not metabolized by the human being. From the rate of elimination in the urine, the biological half-life of the biguanide was found to be 2.8 h. Compared with other biguanides the weak hypoglycaemie activity of metformin in both animals and diabetics cannot be explained by low absorption. It is due to the weak intrinsic activity of this biguanide as such.
Aus 500000 paarigen Hinterleibsdrüsen (Sacculi laterales) weiblicher Seidenspinner konnten 12mg des 4'-Nitro-azobenzol-carbonsäure-(4)-esters eines Alkohols isoliert und nach Spaltung des Esters der hochempfindliche Alkohol selbst durch seine extrem hohe und spezifische Wirksamkeit auf männliche Seidenspinner als reiner Sexuallockstoff des Spinners Bombyx mori L. charakterisiert werden. Analyse und spektrophotometrische Äquivalentgewichtsbestimmung des Azoesters stimmen am besten auf einen doppelt ungesättigten Alkohol der Bruttoformel C 16 H 30 0. Das UV-Spektrum des Derivats zeigt, daß beide Doppelbindungen konjugiert sein müssen 1 ' 2 .Weitere Einblicke in die chemische Konstitution des Bombykol genannten Lockstoffs ergab die katalytische Hydrierung des aus 4 mg NABS-Ester* durch Verseifung mit Glykolmonomethyläther/Alkali in Benzol gewonnenen Lockstoff-Alkohols. Sie führt den bei Raumtemperatur flüssigen Wirkstoff in einen gesättigten Alkohol über, der zur Reinigung sublimiert wurde und bei 47-48° schmilzt. Sein Mischschmelzpunkt mit Cetylalkohol ist nicht erniedrigt. Die Schmelzpunkte des eutektischen Gemisches 3 von authentischem Cetylalkohol bzw. des aus Bombykol erhaltenen Hydrierungsproduktes mit Azobenzol sind identisch. Zur weiteren Sicherung der Grundstruktur des Sexuallockstoffes wurde sein Hydrierungsprodukt mit öc-Naphthyl-isocyanat umgesetzt: Das oc-Naphthylurethan erwies sich nach Schmp. und Mischschmp. als mit dem <%-Naphthylurethan des Cetylalkohols 4 identisch. Der Lockstoffalkohol ist daher ein unverzweigter primärer Alkohol mit 16 C-Atomen.
Much research has been done t o elucidate the mechanism of action of the hypoglycemic biguanides. Their metabolism in man and animal has also been studied to someextent; '-lo the former, however, requires further investigation.This paper constitutes a summary of the present knowledge regarding the elimination and fate of buformin and phenformin in man, taking into account previous reports by Beckmann' and by Beckmann and Hubner. Emphasis is laid on new aspects; data corroborating earlier findings are referred to briefly. The formulae of the two biguanides are: 8 CH~-CH~-CH~-CHZ-NH-~-NH-C-NHZ * HCI II NH NH N'-n-butylbiguanide hydrochloride (buformin hydrochloride) oCHz-CH2-NH-C-NH-C-NHzs I Y HCI NH NH N'-P-phenethylbiguanide hydrochloride (phenformin hydrochloride) METHODSThe concentration of unlabeled buformin* in urine and the levels of unlabeled phenformin* in urine and serum were determined as described elsewhere. The radioactive buformin* used in these experiments was synthesized by the method of Herbert and Pichat" and had an initial specific activity of 7.3 gc/mg. The serum levels of buformin-14C were directly determined in 1-ml aliquots, using 15 ml of the scintillation medium described by Bloom and Nelp. Phenformin-'H* was synthetized by Wilzbach tritiation" with additional irradiation with micr0waves.i The specific activity was 2.7 gc/mg. The radioactivity in the serum of the subject who was given phenformin-H was measured using cellosolve-scintillator after protein precipitation.For the isolation of the phenformin metabolite, the urine of one volunteer who received 50 mg (140 gc) phenformin-JH was collected over 24 hr and mixed with urine from subjects who were given inactive phenformin. The mixed urine was chromatographed on Dowex 50-X8 (200-400 mesh, H tform). The metabolite was eluted with 4 N hydrochloric acid just before the unchanged biguanide left the column.
