To investigate the global expression profile of miRNAs in primary breast cancer (BC) and normal adjacent tumor tissues (NATs) and its potential relevance to clinicopathological characteristics and patient survival, the genome-wide expression profiling of miRNAs in BC was investigated using a microarray containing 435 mature human miRNA oligonucleotide probes. Nine miRNAs of hsa-miR-21, hsa-miR-365, hsa-miR-181b, hsa-let-7f, hsa-miR-155, hsa-miR-29b, hsa-miR-181d, hsa-miR-98, and hsa-miR-29c were observed to be up-regulated greater than twofold in BC compared with NAT, whereas seven miRNAs of hsa-miR-497, hsamiR-31, hsa-miR-355, hsa-miR-320, rno-mir-140, hsa-miR-127 and hsa-miR-30a-3p were observed to be down-regulated greater than twofold. The most significantly up-regulated miRNAs, hsa-mir-21 (miR-21), was quantitatively analyzed by TaqMan real-time PCR in 113 BC tumors. Interestingly, among the 113 BC cases, high level expression of miR-21 was significantly correlated with advanced clinical stage (P = 0.006, Fisher's exact text), lymph node metastasis (P = 0.007, Fisher's exact text), and shortened survival of the patients (hazard ratio [HR]=5.476, P < 0.001). Multivariate Cox regression analysis revealed this prognostic impact (HR=4.133, P = 0.001) to be independent of disease stage (HR=2.226, P = 0.013) and histological grade (HR=3.681, P = 0.033). This study could identify the differentiated miRNAs expression profile in BC and reveal that miR-21 overexpression was correlated with specific breast cancer biopathologic features, such as advanced tumor stage, lymph node metastasis, and poor survival of the patients, indicating that miR-21 may serve as a molecular prognostic marker for BC and disease progression.
Supported by MSS images in the mid and late 1970s, TM images in the early 1990s and TM/ETM images in 2004, grassland degradation in the "Three-River Headwaters" region (TRH region) was interpreted through analysis on RS images in two time series, then the spatial and temporal characteristics of grassland degradation in the TRH region were analyzed since the 1970s. The results showed that grassland degradation in the TRH region was a continuous change process which had large affected area and long time scale, and rapidly strengthen phenomenon did not exist in the 1990s as a whole. Grassland degradation pattern in the TRH region took shape initially in the mid and late 1970s. Since the 1970s, this degradation process has taken place continuously, obviously characterizing different rules in different regions. In humid and semi-humid meadow region, grassland firstly fragmentized, then vegetation coverage decreased continuously, and finally "black-soil-patch" degraded grassland was formed. But in semi-arid and arid steppe region, the vegetation coverage decreased continuously, and finally desertification was formed. Because grassland degradation had obviously regional differences in the TRH region, it could be regionalized into 7 zones, and each zone had different characteristics in type, grade, scale and time process of grassland degradation.
Inter-annual dynamics of grassland yield of the Three Rivers Headwaters Region of Qinghai-Tibet Plateau of China in 1988-2005 was analyzed using the GLO-PEM model, and the herbage supply function was evaluated. The results indicate that while grassland yield in the region showed marked inter-annual fluctuation there was a trend of increased yield over the 18 years of the study. This increase was especially marked for Alpine Desert and Alpine Steppe and in the west of the region. The inter-annual coefficient of variation of productivity increased from the east to the west of the region and from Marsh, Alpine Meadow, Alpine Steppe, Temperate Steppe to Alpine Desert grasslands. Climate change, particularly increased temperatures in the region during the study period, is suggested to be the main cause of increased grassland yield. However, reduced grazing pressure and changes to the seasonal pattern of grazing could also have influenced the grassland yield trend. These findings indicate the importance of understanding the function of the grassland ecosystems in the region and the effect of climate change on them especially in regard to their use to supply forage for animal production. Reduction of grazing pressure, especially during winter, is indicated to be critical for the restoration and sustainable use of grassland ecosystems in the region.
As a powerful predictor of 5-year DSS among patients with NPC, the newly developed NPC-SVM classifier based on tumor-associated biomarkers will facilitate patient counseling and individualize management of patients with NPC.
Background: Nasopharyngeal carcinoma (NPC) is one of the most common malignancies in southern China. The China 1992 TNM staging system has been widely used for prognosis prediction of NPC patients in China. Although NPC patients can be classified according to their clinical stage in this system, their prognosis may vary significantly.
BackgroundThe current World Health Organization (WHO) classification of nasopharyngeal carcinoma (NPC) conveys little prognostic information. This study aimed to propose an NPC histopathologic classification that can potentially be used to predict prognosis and treatment response.MethodsWe initially developed a histopathologic classification based on the morphologic traits and cell differentiation of tumors of 2716 NPC patients who were identified at Sun Yat-sen University Cancer Center (SYSUCC) (training cohort). Then, the proposed classification was applied to 1702 patients (retrospective validation cohort) from hospitals outside SYSUCC and 1613 patients (prospective validation cohort) from SYSUCC. The efficacy of radiochemotherapy and radiotherapy modalities was compared between the proposed subtypes. We used Cox proportional hazards models to estimate hazard ratios (HRs) with 95% confidence intervals (CI) for overall survival (OS).ResultsThe 5-year OS rates for all NPC patients who were diagnosed with epithelial carcinoma (EC; 3708 patients), mixed sarcomatoid-epithelial carcinoma (MSEC; 1247 patients), sarcomatoid carcinoma (SC; 823 patients), and squamous cell carcinoma (SCC; 253 patients) were 79.4%, 70.5%, 59.6%, and 42.6%, respectively (P < 0.001). In multivariate models, patients with MSEC had a shorter OS than patients with EC (HR = 1.44, 95% CI = 1.27–1.62), SC (HR = 2.00, 95% CI = 1.76–2.28), or SCC (HR = 4.23, 95% CI = 3.34–5.38). Radiochemotherapy significantly improved survival compared with radiotherapy alone for patients with EC (HR = 0.67, 95% CI = 0.56–0.80), MSEC (HR = 0.58, 95% CI = 0.49–0.75), and possibly for those with SCC (HR = 0.63; 95% CI = 0.40–0.98), but not for patients with SC (HR = 0.97, 95% CI = 0.74–1.28).ConclusionsThe proposed classification offers more information for the prediction of NPC prognosis compared with the WHO classification and might be a valuable tool to guide treatment decisions for subtypes that are associated with a poor prognosis.
The first-stage of an ecological conservation and restoration project in the Three-River Source Region (TRSR), China, has been in progress for eight years. However, because the ecological effects of this project remain unknown, decision making for future project implementation is hindered. Thus, in this study, we developed an index system to evaluate the effects of the ecological restoration project, by integrating field observations, remote sensing, and process-based models. Effects were assessed using trend analyses of ecosystem structures and services. Results showed positive trends in the TRSR since the beginning of the project, but not yet a return to the optima of the 1970s. Specifically, while continued degradation in grassland has been initially contained, results are still far from the desired objective, 'grassland coverage increasing by an average of 20%-40%'. In contrast, wetlands and water bodies have generally been restored, while the water conservation and water supply capacity of watersheds have increased. Indeed, the volume of water conservation achieved in the project meets the objective of a 1.32 billion m 3 increase. The effects of ecological restoration inside project regions was more significant than outside, and, in addition to climate change projects, we concluded that the implementation of ecological conservation and restoration projects has substantially contributed to vegetation restoration. Nevertheless, the degradation of grasslands has not been fundamentally reversed, and to date the project has not prevented increasing soil erosion. In sum, the effects and challenges of this first-stage project highlight the necessity of continuous and long-term ecosystem conservation efforts in this region.
