Aims: It was our aim to investigate whether AMP-activated protein kinase (AMPK) mediates the considerable increase in reactive oxygen species (ROS) and cell apoptosis induced by bupivacaine in the human neuroblastoma cell line SH-SY5Y. Methods: The recombinant plasmids pGPU6/GFP/Neo-shRNA AMPKα2 and pEGFP-N1-AMPKα2 were constructed and transfected into the SH-SY5Y cell line. The expression of AMPKα2 was determined by RT-PCR and Western blot after transfection. The SH-SY5Y cells transfected with recombinant plasmid were exposed to 1 mmol/l bupivacaine. Cell viability, intracellular ROS and apoptosis were determined. Results: The plasmid pEGFP-N1-AMPKα2 can upregulate the expression of AMPKα2, and the pGPU6/GFP/Neo-shRNA AMPKα2 can downregulate the expression of AMPKα2 in cells. Inhibition of AMPKα2 expression attenuated ROS production and cell apoptosis, and overexpression of AMPKα2 promoted ROS production and cell apoptosis after bupivacaine treatment. Conclusion: AMPK probably mediated ROS production and cell apoptosis induced by bupivacaine.
According to the characteristic of synchronizer signal, multi-channel synchronizer signal generator is designed based on USB and C8051F340. Software implementation is given using Aitken interpolation algorithm. It greatly reduces the cost of development and effectively improves the precision. Meanwhile, using the USBXPress development kit for PC and firmware development shortens the development cycle. Compared with the special numeric synchronizer converter, the price of designed Synchronizer signal generator cheap 100 times, the full range error less than 0.04º and the accuracy improved 25 times. At the same time, it has work stability, strong anti-interference and high practical value.
Masonry infills are commonly used in reinforced concrete (RC) frame structures. In this paper a separate finite element modelling method is proposed and used to study the lateral behaviour of single-storey one-bay masonry infilled frames with openings under monotonic horizontal loading. The interface friction model and the surface-based cohesive model are combined to simulate masonry interface and the interface between masonry wall and RC frame. Damage plastic model of material property is adapted to capture the brittle behaviour of concrete and bricks. Appropriate experimental data available from the literature are utilized to verify the model. The comparison between the numerical analysis and test results indicates that the proposed FE modelling method can predict the masonry infill wall’s crack patterns and the lateral strength and stiffness of the structure with window or door opening successfully.
The determination of the physical property cutoffs of the effective reservoirs is a major factor that affects the result of the reserve calculation and is a difficult problem in the research of the reservoir evaluation. It is related to the exploration, development and decision-making. This paper used the empirical statistical method, the minimum pore throat radius method, the oil attitude method and the porosity permeability cross plot method to exploratorily strike the physical property cutoffs of the effective reservoirs of Yaoyi Formation in Cha 48 zone. It was based on the actual situation of the data of cores, the well testing and the well logging and combined with the applicability of various methods. The results achieve the attest of the data from the well testing and the producing test and provide the basis for the reservoir calculation.
