O objetivo deste estudo foi descrever a diversidade genética de S. adstringens em três populações localizadas em Unidades de Conservação do Estado de Minas Gerais, utilizando-se marcadores isoenzimáticos. Foram amostradas as populações dos Parques Estaduais (PE) do Biribiri e Serra Nova e Parque Nacional (PN) das Sempre Vivas. Foram empregados 14 marcadores isoenzimáticos, dos quais nove locos foram polimórficos (est-2, est-3, est-4, got-1, pgi-1, mdh-4, idh-1, adh-1 e skdh-1). Os testes de ajustamento às proporções genotípicas de EHW, teste exato de Fisher e χ² não foram significativos nos locos isoenzimáticos das três populações, exceto est-4 no PE Serra Nova, mdh-4 e idh-1 no PE do Biribiri. Os valores de heterozigosidade média esperada (He) variaram entre 0,217 e 0,255, estando próximos dos encontrados em espécies arbóreas tropicais (0,204 e 0,211) com as mesmas características de distribuição geográfica de S. adstringes. O índice de fixação (F) foi significativamente menor do que zero na população do PE do Biribiri (-0,149) e não significativo no PN das Sempre Vivas (-0,031) e no PE Serra Nova (-0,138). O excesso significativo de heterozigotos estimado na população do PE do Biribiri pode significar seleção a favor desses genótipos, porém, para verificar tal possibilidade, são requeridos estudos que consideram variáveis ambientais.
Recebido para publicação em 21/08/2012 Aceito para publicação em 08/10/2013 RESUMO: O S. adstringens, árvore típica do Cerrado, tem sido explorada visando suas propriedades medicinais e tanantes. Em razão do ainda incipiente conhecimento genético da espécie, este trabalho teve como objetivo caracterizar a diversidade e a estrutura genética de S. adstringens por meio de marcadores aloenzimáticos. Foram coletadas sementes em cinco mesorregiões brasileiras, sendo amostrados 627 indivíduos divididos em 16 populações localizadas nos Estados de Minas Gerais e Goiás. Foram testados 14 sistemas isoenzimáticos; destes, sete foram polimórficos com o total de 10 locos e 28 alelos. O valor de diversidade genética média (H) foi 0,226, a proporção média de locos polimórficos (P) foi 68,75, o número médio de alelos por loco polimórfico (AP) foi 2,65 e o número efetivo de alelos (A e ) foi igual a 1,29. Resultados do índice de fixação total (F= 0,003), do índice de fixação dentro de populações (f = -0,114) e, da medida de diferenciação genética (θ =0,105) foram não significativos, indicando a inexistência de estruturação genética. Na análise de agrupamento (UPGMA) foram observados dois grupos principais, o primeiro formado pela população do Parque Estadual (PE) do Rio Preto (MG), e outro, formado pelas demais populações. Se excluída a população do PE do Rio Preto das análises, G ST é drasticamente reduzido de 0,077 para 0,026. Assim, aproximadamente 2/3 do valor total de G ST verificado em S. adstringens foi devido à variação entre a população do PE do Rio Preto e as demais populações. De modo geral, os valores H e P observados em S. adstringens são compatíveis aos constatados em árvores tropicais comumente distribuídas. Por outro lado, excluindo a população do PE do Rio Preto, o valor da medida de diferenciação genética G ST foi menor que o verificado em árvores tropicais nativas e pinheiros de zonas temperadas. A semelhança entre populações avaliadas indica que o fluxo gênico ainda é alto o suficiente para prevenir a diferenciação genética, pelo menos em nível local.Palavras-chave: locos aloenzimáticos, diversidade genética, barbatimão, plantas medicinais.ABSTRACT: Characterization of the diversity and genetic structure in natural populations of Stryphnodendron adstringens (Mart.) This study aimed to evaluate the effects of substituting wheat bran (WB) by corn gluten feed without steep water (CGF) in cats feeds regarding the nutritional value of the diet, stool characteristics, time of food passage through the gastrointestinal tract (GIT), intestinal gas production and concentrations of short chain fatty acids (SCFA) in blood and faeces. Twenty eight cats were distributed in a randomized block design with four treatments (0, 2.6, 5.3 or 8.0% CGF, based on dry matter, substituting for WB) and seven replicates. Substitution of WB by CGF decreased (P<0.01) the digestibility of acid detergent fibre without influencing (P>0.05) the digestibility of other nutrients and apparent metabolizable energy. There was also no effect (P>0.05) on faeces moisture, faecal pH and score, food transit time in the GIT, intestinal gas area and SCFA concentrations in faeces, while blood concentrations of propionate were influenced differently by the different feeds and sampling times (P<0.05). Thus replacement of up to 8% of WB by CGF may be done safely in cats feed.
-(Allozyme variation in a natural population of Stryphnodendron adstringens in the Rio Preto State Park, southeastern Brazil). Leaves and fruits from 63 Stryphnodendron adstringens trees were sampled in the Rio Preto State Park to analyze allozyme segregation, tissue specific expression of allozyme loci, and their genetic parameters. The enzyme systems ADH, EST, ACP, PGM, PGI, GDH, G6PDH, GOT, IDH, LAP, MDH, PER and SKDH were assessed by means of starch-gel electrophoresis. The polymorphic systems PGI, IDH, MDH and GOT demonstrated a dimeric quaternary structure, while EST and PER were monomeric. The total expected genetic diversity (H E ) for leaves and seeds were 0.325 and 0.244 respectively. The effective number of alleles per locus (A E ) was 1.58 in leaves and 1.42 in seeds. The values of H E and A E observed in S. adstringens were comparatively higher than the average values seen in allozyme studies of other woody plants. The values of the fixation indices for the population, considering leaves (f = 0.070) and seeds (f = 0.107), were not significant. The high values of genetic diversity and of effective number of alleles per locus, as well as the non-significant fixation index and the adjustments of the Hardy-Weinberg proportions between generations for the pgi-1, mdh-2 and idh-1 loci, indicated random mating in this population. The enzyme systems EST and PER demonstrated their best resolution in leaf tissues, while the MDH, IDH, PGI and GOT systems demonstrated their best resolution in seed tissues.
